      Charged multivesicular body protein 1b (CHMP1B) is a member of the chromatin-modifying protein/charged multivesicular body protein (CHMP) family. CHMP1B is a possible peripherally associated component of the ESCRT-III (endosomal sorting required for transport complex III) which is involved in MVBs (multivesicular bodies) formation and sorting of endosomal cargo proteins into MVBs. The MVBs contain ILVs (intraluminal vesicles) which are created by invagination and scission from the limiting membrane of the endosome and generally are transported to lysosomes enabling degradation of membrane proteins, such as stimulated growth factor receptors, lysosomal enzymes and lipids.

      Retinitis pigmentosa 9 (RP9) is assumed to be a target protein for the PIM1 kinase. The RP9 protein may have some roles in B-cell proliferation in association with PIM1. RP9 can be bound and phosphorylated by the protooncogene PIM1 product, a serine/threonine protein kinase. RP9 localizes in nuclear speckles containing the splicing factors, and has a part in pre-mRNA splicing. RP9 gene mutations result in autosomal dominant retinitis pigmentosa-9.

    • Background

      I-TAC is a small cytokine belongs to the CXC chemokinen family which also called inducible T-cell alpha chemoattractant and IP-9. I-TAC is expressed mainly in peripheral blood leukocytes, liver and pancreas with moderate levels in spleen, thymus and lung and low levels in small intestine, placenta and prostate. IFN-g and IFN-b induces strongly gene expression of I-TAC. The I-TAC chemokine elicits its effects on its target cells by interacting with the cell surface chemokine receptor CXCR3, with a higher affinity than do the other ligands for this receptor, CXCL9 and CXCL10.

    Activation Protocol:

    2. Activate MMP3 by adding Chymotrypsin(Sigma, Catalog#C­3142,1mg/ml stock in 1mM HCl) to a final concentration of 5ug/ml.
    3. Incubate at 37°C for 30 minutes.
    4. Stop activation with 2mM PMSF. Pre-warm the PMSF to 37°C prior to adding to sample.

      Alpha 1-microglobulin (A1M) is a lipocalin superfamily member (kernal lipocalins). A1M is a low molecular weight protein component of plasma. A1M is distributed in plasma and extravascular compartments of all organs. Alpha-1 Microglobulin is found in mammals, birds, amphibians and fish. The primary sites of A1M synthesis are the liver and the kidney. Around the opening of the lipocalin pocket three lysyl residues are situated; those residues carry yellow-brown modification derived from the binding and degradation of heme and kynurenin (a tryptophan metabolite). A1-Microglobulin’s reductase and dehydrogenase have broad biological substrate specificity properties due to its’ free cysteine side-chain which is located in a flexible loop. RPL35A is a component of the 60S subunit of the ribosomal protein and a member of the L35AE family of ribosomal proteins. RPL35A is situated in the cytoplasm. The rat protein is known to bind to both initiator and elongator tRNAs, therefore, it is located at the P site, or P and A sites, of the ribosome. Though RPL35A was initially mapped to chromosome 18, it has been proven that it is located at 3q29-qter. Transcript variants utilizing alternative transcription initiation sites and alternative polyA signals exist.       Chemokine (C-C motif) ligand 8 (CCL8) is a small cytokine belonging to the CC chemokine family that was once called monocyte chemotactic protein-2 (MCP-2). The CCL8 protein is produced as a precursor containing 109 amino acids, which is cleaved to produce mature CCL8 containing 75 amino acids. The gene for CCL8 is encoded by 3 exons and is located within a large cluster of CC chemokines on chromosome 17q11.2 in humans. MCP-2 is chemotactic for and activates a many different immune cells, including mast cells, eosinophils and basophils, (that are implicated in allergic responses), and monocytes, T cells, and NK cells that are involved in the inflammatory response. CCL8 elicits its effects by binding to several different cell surface receptors called chemokine receptors. These receptors include CCR1, CCR2B and CCR5.
