      Members of the IFN regulatory factor (IRF) family regulate gene expression critical to immune response, hemopoiesis, and proliferation. IRF-3 is a member of the IRF family, and is distinct from other family members. Its transcriptional activity is regulated solely by posttranslational modifications. It plays a crucial role in activation of innate immunity and inflammation in response to viral infection. IRF-3 mediates IFN-stimulated response element (isre) promoter activation. Functions as a molecular switch for antiviral activity. Dsrna generated during the course of an viral infection leads to IRF3 phosphorylation on the c-terminal serine/threonine cluster. This induces a conformational change, leading to its dimerization, nuclear localization and association with creb binding protein (crebbp) to form dsrna-activated factor 1 (draf1), a complex which activates the transcription of genes under the control of isre. The complex binds to the ie and prdiii regions on the ifn-alpha and ifn-beta promoters respectively. IRF-3 does not have any transcription activation domains.

      IFN-tau is also known as TP-1 (trophoblast protein-1) is a new class of type I IFN that is secreted by the trophoblast and is the signal for maternal recognition of pregnancy in sheep. IFN- tau has potent immunosuppressive and antiviral activities similar to other type I IFN but is less cytotoxic than IFN-alpha and IFN-beta. The current investigation concerns the effect of recombinant ovine IFN- tau (rOIFN- tau) on the modulation of MHC class I and II expression on cloned mouse cerebrovascular endothelial (CVE) cells.
      IFN-tau induced tyrosine phosphorylation of Stat1 and upregulated the expression of MHC class I on CVE. One proposed action by which type I IFN reduces the relapse rate in MS is via interference with IFN-?-induced MHC class II expression. IFN- tau was shown to downregulate IFN-?-induced MHC class II expression on CVE and, hence, may be of potential therapeutic value in downregulating inflammation in the central nervous system (CNS). IFN- tau did not upregulate the expression of MHC class II on CVE. IFN- tau also inhibited the replication of Theiler's virus in CVE.

      IFN-alpha forms are produced by monocytes/macrophages, lymphoblastoid cells, fibroblasts, and a number of different cell types following induction by viruses, nucleic acids, glucocorticoid hormones, and low-molecular weight substances (n-butyrate, 5-bromodeoxy uridine). At least 23 different variants of IFN-alpha are known. The individual proteins have molecular masses between 19-26 kDa and consist of proteins with lengths of 156-166 and 172 amino acids. All IFN-alpha subtypes possess a common conserved sequence region between amino acid positions 115-151 while the amino-terminal ends are variable. Many IFN-alpha subtypes differ in their sequences at only one or two positions. Naturally occurring variants also include proteins truncated by 10 amino acids at the carboxy-terminal end. Disulfide bonds are formed between cysteines at positions 1/98 and 29/138. The disulfide bond 29/138 is essential for biological activity while the 1/98 bond can be reduces without affecting biological activity. All IFN-alpha forms contain a potential glycosylation site but most subtypes are not glycosylated. In contrast to IFN-gamma IFN-alpha proteins are stable at pH2.

      There are at least 23 different IFN-alpha genes. They have a length of 1-2 kb and are clustered on human chromosome 9p22. It is not known whether all these genes are expressed following stimulation of the cells. Understanding the potential uses and benefits of the protein Interferon may be particularly important due to the relationship they may play in developments towards the future of human health. Below, we examine what Interferon is, and why it matters.

Mechanism
Interferon Types I and II are members of the cytokines family, and have a crucial role to play in the way that information is conveyed between cells. Essentially, Interferon provide instructions between cells in response to pathogens, such as viruses, bacteria, and other foreign agents that the body considers to be unwanted intruders.
There are a number of different types of Interferons, but it is Type I Interferon that is best known and most researched. Type I is known to induce the onset of anti-proliferative, immunomodulatory, and anti-viral response. Type II has a similar function; it is secreted by ‘natural killer’ cells and T lymphocytes and can inhibit the growth, and sometimes destroy, viruses and cancerous cells.
Interferon Type III has been identified, but as yet its application is not particularly well understood.

Function
Interferon is a protein that acts as a battling sergeant, marshaling an army of eager soldiers, and issuing commands for an oncoming war. It is essentially the “wake up, trouble is coming” protein, helping to launch immune defenses against pathogens.
Interferon has become particularly of interest in the field of medical research due to the ability to provide immune responses to threats. Synthetic Interferon is now being manufactured and administered to patients, following successful research and trials into the potential benefits of additional Interferon proteins being used to fight a known threat. This threat may be a foreign virus, or a cancer.
At present, Interferon is prescribed for the following illnesses:
● Various forms of cancer, including melanoma, leukemia, and the AIDS-related cancer Kaposi’s Sarcoma.
● Viral infections, including the chronic strains of Hepatitis B and C.
● Type I Interferon is commonly used in the treatment of multiple sclerosis.

Interactions
Type I Interferon proteins are the defences of the body; the soldiers that help to ward off illness. In many cases, this is effective, and Type I Interferon proteins activate molecules that prevent a virus from producing and replicating.
However, certain pathogens have been found to be resistant to Interferon:
● H5NI; better known as “Bird Flu”
● Japanese Encephalitis
● Strains of the herpes virus have been shown to be resistant to Interferon
● Certain strains of the Dengue virus are also able to overcome the resistance mounted by Interferon.
