These enzymes cleave a peptide bond in C4 releasing the anaphylatoxin C4a and activating C4b. C4A and C4B are 2 variants of C4 that are common in man whereas, in animals who have only 1 type usually have C4B.

C4BP has a flower shaped structure with each alpha chain radiating out from a central core. The arms are linked by disulfide bonds in the centre and the ends of each arm bind C4b. C4BP acts as a cofactor allowing factor I to split and permanently inactivate C4b. C4b binding protein modulates complement activation of the classical and lectin pathways. C4BP binds to C4b and accelerates the dissociation of C2a from the C3/C5 convertase C4b,C2a. This inactivates the central enzyme of both pathways. Studies have shown that on surfaces densely coated with C4b, C4BP can bind up to four C4b molecules simultaneously.

Complement Component 4 Binding Protein, Beta (C4BPB) belongs to a superfamily of proteins composed primarily of tandemly arrayed short consensus repeats of approximately 60 amino acids. C4BPB has a regulatory role in the coagulation system also, mediated via the beta-chain binding of protein S, a vitamin K-dependent protein which functions as a cofactor of activated protein C. A single, unique beta-chain of the C4BPB assembles with 7 identical alpha-chains into the principal isoform of C4BPB, which is a multimeric protein that controls activation of the complement cascade via the classical pathway. C4BPB binds as a cofactor to C3b/C4b inactivator (C3bINA), which subsequently hydrolyzes the complement fragment C4b. In addition, C4BPB accelerates the degradation of the C4bC2a complex (C3 convertase) by detaching the complement fragment C2a. Furthermore, C4BPB interacts with anticoagulant protein S and with serum amyloid P component.

Complement C4c is a degradation product of C4b which is cleaved by C4b/C3b inactivator to yield C4d and C4c.
Complement C4c interacts with C1 and C2 to form C3 convertase of the classic activation pathway. Systemic lupus erythematosus (SLE) is often associated with congenital C4 deficiency. Reduced levels of components of the classic pathway (C1, C2, C4, C3) are common after such activation, e.g. in SLE, acute serum sickness and conditions associated with circulating immune complexes.

Complement C5 is cleaved into C5a and C5b and is activated by all 3 pathways of complement activation. Each pathway of complement activation generates proteolytic enzyme complexes which binds to the target surface. An activation peptide, C5a, which is an anaphylatoxin, which has potent spasmogenic and chemotactic activity, is derivative from the alpha polypeptide via cleavage with a convertase. The C5b macromolecular cleavage product forms a complex with the C6 complement component, and this complex is the basis for creation of the membrane attack complex, which includes supplementary complement components.
