Crk-like protein (CRKL) is a protein kinase containing SH2 and SH3 (src homology) domains which activates the RAS and JUN kinase signaling pathways and transforms fibroblasts in a RAS-dependent manner. CRKL is a substitute of the BCR-ABL tyrosine kinase, it also has a role in fibroblast transformation by BCR-ABL, and has oncogenic potential.

Simian immunodeficiency virus, SIV, is a retrovirusthat is found in strains, having the HIV-1and HIV-2specific strains that infect humans. CRK belongs to the signaling adapter protein family which binds to several tyrosine-phosphorylated proteins. CRK is involved in many cellular processes such as apoptosis, proliferation, and differentiation. CRK has a modular domain architecture consisting of an SH2 followed by two SH3 domains (src-homology domains). The N-terminal SH2 domain of the CRK protein functions as a positive regulator of transformation whereas the C-terminal SH3 domain functions as a negative regulator of transformation.

HRAS is part of the Ras oncogene family, whose members are related to the transforming genes of mammalian sarcoma retroviruses. The products encoded by these genes participate in signal transduction pathways. These proteins can bind GTP and GDP, and they have intrinsic GTPase activity. HRAS go trough a continuous cycle of de- and re-palmitoylation, which mediates its rapid exchange between the plasma membrane and the Golgi apparatus. Mutations in HRAS result in Costello syndrome, a disease characterized by increased growth at the prenatal stage, growth deficiency at the postnatal stage, predisposition to tumor formation, mental retardation, skin and musculoskeletal abnormalities, distinctive facial appearance and cardiovascular abnormalities. Defects in this HRAS gene are implicated in a range of cancers, including bladder cancer, follicular thyroid cancer, and oral squamous cell carcinoma.

RRAS2 (RAS viral (r-ras) oncogene homolog 2) is a member of the Ras like GTPase family that cycle between GTP-bound active and GDP-bound inactive states at the cytoplasmic face of the plasma membrane. RRAS2 is a plasma membrane-associated GTP-binding protein with GTPase activity. RRAS2 is implicated in the pathogenesis of human cancers. Hence, overexpression of active RRAS2 in EpH4 cells induces tumorigenicity through the phosphoinositide 3-kinase, p38 MAPK, and mTOR pathways, as these cells lose their sensitivity to the normal growth inhibitory role of TGF-beta. RRAS2 might transduce growth inhibitory signals across the cell membrane, exerting its effect via an effector shared with the Ras proteins but in an antagonistic manner.
RRAS2 is ubiquitously present in all tissues examined, with the highest levels in the heart, placenta, and skeletal muscle. Moderate levels of RRAS2 are found in the lung and liver and low levels in the brain, kidney, and pancreas.
RRAS2 deficiencies are linked to ovarian cancer.

Rab3D mRNA is primarily expressed in white adipose tissue and, coincident with GLUT4, increases upon differentiation of 3T3-L1 fibroblasts. Rab3D exhibits close structural similarities with Rab 3A, an isoform which is functionally associated with regulated exocytosis in various cell types.

RAB3A Interacting Protein Like 1 (RAB3IL1) is a 382 amino acid protein which is a member of the SEC2 family. RAB3IL1 is a guanine nucleotide exchange factor (GEF) for Rab3A. RAB3IL1 competes for interaction with Rab3A and IHPK1 via its coiled coil domain, which doesn’t involve IHPK1 kinase activity.

The RAN complex with BIRC5 participates in mitotic spindle formation by being as a physical scaffold to assist the delivery of the RAN effector molecule TPX2 to microtubules. RUN increases AR-mediated transactivation.

Defects in this NRAS gene result in juvenile myelomonocytic leukemia.

RAB, Member RAS Oncogene Family-Like 5 (RABL5) belongs to the Ras superfamily of small GTP-binding proteins. The Ras-related superfamily of guanine nucleotide binding proteins includes the R-Ras, Rap, Ral/Rec and Rho/Rab subfamilies. Rab proteins have an essential role in endocytosis or in biosynthetic protein transport. The transporting process of newly synthesized proteins from the endoplasmic reticulum to various stacks of the Golgi complex and to secretory vesicles includes the movement of carrier vesicles and requires Rab protein function.

RAP2A belongs to the RAS oncogene family and a Ras-like small G protein. The RAP2A protein shares 60% identity with Rap1A and exhibits a carboxy-terminal CAAX motif and 2 upstream cysteines similar to those of the H-Ras, K-Ras and N-Ras proteins. In contrast with Rap1A and Rap1B, overexpression of RAP2A doesn’t obstruct the Ras signaling pathway.

RAB31 Member RAS Oncogene Family (RAB31) is a 194 amino lipid-anchored protein which localizes to the cytoplasmic side of the cell membrane and is a member of the Ras related GTPase superfamily. RAB31 is expressed at high levels in the lung, brain and heart. RAB31 functions in a similar mode to other Rab proteins, namely playing a part in protein transport. Small GTP-binding proteins of the RAB family, for example RAB31, play critical roles in vesicle and granule targeting. RAB31 gas the highest expression in the placenta and brain with lower levels in the heart and lung. However, RAB31 is not detected in liver, skeletal muscle, kidney or pancreas.

Ras-related protein Rap-2b (RAP2B) is a part of RAS-related genes family. RAP2B has a threonine residue at aa 61Unlike normal ras proteins which have a nontransforming glutamine residue at that position. The threonine residue makes the intrinsic GTPase activity for Rap2B lower, by this means allows RAP2B to exist in the activated state for a longer period of time than normal ras proteins. RAP2B is a platelet protein which is activated by thrombin and involved in platelet activation. RAP2B may be a candidate oncogene which takes part in carcinogenesis through activation of NF-kappaB pathway.

RAB14 belongs to the RAB protein family, low molecular mass monomeric GTPases which is found on the cytoplasmic surfaces of distinct membrane bound organelles. RAB14 has a role in vesicular trafficking and neurotransmitter release throughout the body and is expressed at high levels in brain, lung, kidney, spleen and thymus. In addition, RAB14 takes part in the biosynthetic/recycling pathway between the Golgi and endosomal compartments.

RAB13 is a member of the small GTPase superfamily which are nontransforming monomeric GTP-binding proteins that hold 4 extremely conserved regions that take part in GTP binding and hydrolysis. The GTPase Rab13 regulates the assembly of functional epithelial tight junctions (TJs) and is expressed in every phase of preimplantation development.

RAB18 belongs to a family of Ras-related small GTPase which is lipid-anchored to the cytoplasmic side of the cell membrane. RAB18 takes part in apical recycling, endocytosis and protein transport between early endosomes and the plasma membrane.
