APOA1 (Apolipoprotein A-1) is a human protein with a specific role in lipid metabolism being the main protein component of HDL in the plasma. APOA1 promotes cholesterol efflux from tissues to the liver for excretion. Furthermore, APOA1 is a cofactor for LCAT, which is responsible for the formation of most plasma cholesteryl esters. In addition, APOA1 activates spermatozoa motility as part of the SPAP complex. The APOA1 gene is strongly linked with two other apolipoprotein genes on chromosome 11. Uromodulin is the most abundant protein in normal urine. Its secretion in urine follows proteolytic cleavage of the ectodomain of its glycosyl phosphatidylinosital-anchored counterpart that is situated on the luminal cell surface of the loop of Henle. The HIV-2 Gag protein plays a central role in the assembly and maturation of viral particles. Gag is a polyprotein precursor that undergoes a series of proteolytic cleavages to yield the mature structural proteins essential for viral assembly and infectivity. Research on HIV-2 Gag has been limited compared to HIV-1, and a comprehensive characterization of HIV-2 Gag recombinant is necessary to shed light on its role in viral replication.
The primary objective of this study is to express and purify recombinant HIV-2 Gag protein using various expression systems. Recombinant DNA techniques will be employed to construct expression vectors harboring the HIV-2 gag gene, followed by expression in bacterial, yeast, or mammalian cell-based systems. The recombinant Gag protein will be purified using affinity chromatography or other suitable methods, allowing for subsequent biochemical and biophysical characterization.
The second objective is to investigate the assembly and maturation processes of HIV-2 Gag protein. In vitro assays will be conducted to analyze the ability of the purified Gag protein to self-assemble into virus-like particles (VLPs). The formation and morphology of VLPs will be characterized using techniques such as electron microscopy and dynamic light scattering. Additionally, the effect of potential inhibitors or small molecules on Gag assembly and maturation will be evaluated.
The third objective is to determine the three-dimensional structure of HIV-2 Gag recombinant through X-ray crystallography or cryo-electron microscopy. Structural information will provide crucial insights into the molecular interactions involved in Gag assembly and maturation, offering a basis for rational drug design targeting HIV-2 Gag.

Introduction
EPCAM is a carcinoma-associated antigen and belongs to a family which includes at least 2 type I membrane proteins. The EPCAM protein has a role in embryonic stem cells proliferation and differentiation. EPCAM is used as a target for immunotherapy treatment of human carcinomas. EPCAM is expressed on most normal epithelial cells and gastrointestinal carcinomas and acts as a homotypic calcium-independent cell adhesion molecule. Epithelial cell adhesion molecules (EPCAM) can act as a physical homophilic interaction molecule between intestinal epithelial cells (IECs) and intraepithelial lymphocytes (IELs) at the mucosal epithelium for supplying immunological barrier as a first line of defense against mucosal infection. EPCAM gene mutations result in congenital tufting enteropathy.

Introduction
DHH is part of the Hedgehog family which encodes signaling molecules that are involved in regulating morphogenesis. DHH protein is a precursor that is autocatalytically cleaved, the N-terminal portion is soluble and contains the signalling activity while the C-terminal portion is involved in precursor processing. Additionally, the C-terminal product covalently attaches a cholesterol moiety to the N-terminal product, restricting the N-terminal product to the cell surface and preventing it from freely diffusing throughout the organism. Defects in DHH protein have been associated with partial gonadal dysgenesis (PGD) accompanied by minifascicular polyneuropathy. DHH plays a role both male gonadal differentiation and perineurial development.
DHH plays a role in intercellular signaling which is essential for a variety of patterning events during development. DHH functions as a spermatocyte survival factor in the testes & is essential for testes development. -
