The HIV1 life cycle is a complex process involving several key viral enzymes, one of which is the integrase (IN). HIV1 integrase plays a crucial role in viral replication by catalyzing the integration of viral DNA into the host cell genome, an essential step for the establishment of a persistent infection.
The study of HIV1 integrase has been of great interest to researchers due to its significance as a therapeutic target. In recent years, advances in recombinant DNA technology have allowed for the production and purification of HIV1 integrase in recombinant form, enabling detailed structural and functional studies. This research aims to characterize the HIV1 integrase recombinant and gain insights into its mechanisms of action during viral integration.
The first objective of this study is to express and purify recombinant HIV1 integrase using various expression systems such as bacterial, yeast, or mammalian cell-based systems. Recombinant DNA techniques, including cloning and expression vector design, will be employed to generate the desired constructs for protein production. The recombinant integrase will be purified using affinity chromatography, followed by characterization using biochemical and biophysical techniques.
The second objective is to investigate the enzymatic activity of the purified HIV1 integrase recombinant. In vitro assays will be performed to determine its ability to catalyze the integration of viral DNA into target DNA sequences. Various substrates, including oligonucleotides and plasmids, will be used to assess the substrate specificity and kinetics of the integrase enzyme. Furthermore, the effects of potential inhibitors or small molecules on the enzymatic activity will be evaluated.
The third objective is to elucidate the three-dimensional structure of the HIV1 integrase recombinant using techniques such as X-ray crystallography or cryo-electron microscopy. This structural information will provide valuable insights into the mechanism of integrase function and aid in the rational design of novel inhibitors targeting integrase.
By characterizing the HIV1 integrase recombinant, this research aims to contribute to our understanding of the molecular mechanisms underlying viral integration. The findings from this study may provide crucial information for the development of novel therapeutic strategies targeting HIV1 integrase, potentially leading to the discovery of more effective antiretroviral drugs.

Ebolavirus (EVD) belongs to the Filoviridae family of proteins which is comprised of a single-strand, non-infectious RNA genome. EVD genome is about 19,000 base pairs long and covers 7 genes in the order 3'-UTR-NP-VP35-VP40-GP-VP30-VP24-L-5'-UTR. There are 4 different ebolaviruses such as: Zaire (EBO-Z), Sudan (EBO-S), Cote d’Ivoire (EBO-CI) and Reston (EBO-R) that differ in amino acid sequence and location of where the gene overlaps. Zika virus (ZIKV) belongs to the family Flaviviridae and the genus Flavivirus, it is transmitted by daytime-active Aedes mosquitoes, such as A. aegypti and A. albopictus. The Zika virus is related to the dengue, yellow fever, Japanese encephalitis, and West Nile viruses. Much like the other flaviviruses, Zika virus is enveloped and icosahedral and has a nonsegmented, single-stranded, positive-sense RNA genome. Zika fever is an infection, which often causes no symptoms or only mild ones, like a mild form of dengue fever, and it is treated by rest. IL11 is a member of the gp130 family of cytokines. These cytokines drive the assembly of multisubunit receptor complexes, all of which contain at least one molecule of the transmembrane signaling receptor IL6ST (gp130). Synovial Sarcoma, X Breakpoint 1 (SSX1) is a member of a family of highly homologous synovial sarcoma X (SSX) breakpoint proteins. SSX proteins are localized to the nucleus and expressed in testis and some types of cancers and, hence, they are classified as C/T (cancer/testis) antigens. These proteins may serve as transcriptional repressors. SSX1 genes are involved in the t (X;18) translocation typically found in all synovial sarcomas. Interleukin 33 (IL-33) is a 32kDa proinflammatory cytokine that may also regulate gene transcription in producer cells. IL-33 is structurally related to IL-1, which induces helper T cells to produce type 2 cytokines and acts through the receptor IL1RL-1 (IL1 receptor-like-1), which is known also as ST2. Chikungunya is an infection caused by the chikungunya virus which is passed to humans by two species of mosquito of the genus Aedes: A. albopictus and A. aegypti. Animal reservoirs of the virus include monkeys, birds, cattle, and rodents. The features of the disease are a sudden onset of fever 2-4 days after exposure. SAA1 protein is an acute phase apolipoprotein reactant which is produced mostly by hepatocytes and under regulation of inflammatory cytokines. SAA1 (Serum amyloid A1) protein is produced mainly in the liver and circulates in low levels in the blood. The SAA1 seems to have a role in the immune system. DiGeorge syndrome critical region 6-like protein (DGCR6L) shares homology with the Drosophila gonadal protein, it is expressed in gonadal tissues and germ cells, and with the human laminin gamma-1 chain which functions in cell attachment and migration. Acidic fibroblast growth factor is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes, including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth and invasion. This protein functions as a modifier of endothelial cell migration and proliferation, as well as an angiogenic factor. It acts as a mitogen for a variety of mesoderm- and neuroectoderm-derived cells in vitro, thus is thought to be involved in organogenesis. Three alternatively spliced variants encoding different isoforms have been described. The binding growth factors are angiogenic agents in vivo and are potent mitogens for a variety of cell types in vitro. There are differences in the tissue distribution and concentration of these 2 growth factors.
