    • Introduction

The proteasome is a multicatalytic proteinase complex with a highly assembled ring-shaped 20S core structure which is composed of 4 rings of 28 non-identical subunits, 2 rings are composed of 7 alpha subunits and 2 rings are composed of 7 beta subunits. Proteasomes are found throughout eukaryotic cells at a high concentration and cleave peptides in an ATP/ubiquitin-dependent process in a non-lysosomal pathway. PSMA7 is part of the peptidase T1A family, that is a 20S core alpha subunit. PSMA7 interacts particularly with the hepatitis B virus X protein, a protein critical to viral replication. PSMA7 is involved in regulating hepatitis virus C internal ribosome entry site activity that is crucial for viral replication. PSMA7 is in charge for regulating the hypoxia-inducible factor-1alpha, a transcription factor important for cellular responses to oxygen tension. PSMA7 is characterized by its ability to cleave peptides with Arg, Phe, Tyr, Leu, and Glu adjacent to the leaving group at neutral or slightly basic pH. PSMA7 interacts specifically with two subdomains of HIF-1alpha and inhibited the transactivation function of HIF-1alpha under both normoxic and hypoxia-mimicking conditions.

CTK-19 is a member of the keratin family. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. The type I cytokeratins consist of acidic proteins which are arranged in pairs of heterotypic keratin chains. Unlike its related family members, this smallest known acidic cytokeratin is not paired with a basic cytokeratin in epithelial cells. It is specifically expressed in the periderm, the transiently superficial layer that envelopes the developing epidermis. The type I cytokeratins are clustered in a region of chromosome 17q12-q21.

    • Introduction

ICAM-1 also called CD54 is a single chain membrane glycoprotein expressed on the surface of a variety of non-haematopoietic and haematopoietic cell types and has roles in signal transduction, cell signaling and lymphocyte adhesion. ICAM1 binds to integrins such as CD11a / CD18, or CD11b / CD18. ICAM1 is also used by Rhinovirus as a receptor. ICAM-1 is an intercellular adhesion molecule constantly present in low concentrations in the membranes of leukocytes and endothelial cells. When stimulated by cytokine the concentrations significantly increase. ICAM-1 can be stimulated by interleukin-1 (IL-1) and tumor necrosis factor alpha (TNFA) and is expressed by the vascular endothelium, macrophages and lymphocytes. ICAM-1 is a ligand for LFA-1 which is a receptor found on leukocytes. Upon activation, leukocytes bind to endothelial cells via ICAM-1/LFA-1 and then transmigrate into tissues. ICAM-1 is implicated in subarachnoid hemorrhage (SAH). Levels of ICAM-1 are shown to be notably elevated in patients with SAH. Soluble ICAM-1 is detectable in the plasma and is elevated in patients with various inflammatory conditions.

    • Background

Intercellular adhesion molecule-1 (ICAM-1) is a cell surface glycoprotein that plays a pivotal role in immune responses and inflammatory processes. This research aims to investigate the function and significance of ICAM-1 protein in various physiological and pathological conditions. Understanding the molecular mechanisms and regulatory roles of ICAM-1 can provide valuable insights into its potential as a therapeutic target for immune-related disorders.

Structure and Expression of ICAM-1 Protein:

ICAM-1 belongs to the immunoglobulin superfamily and is composed of five immunoglobulin-like domains. It is primarily expressed on the surfaces of endothelial cells, leukocytes, and other immune cells. ICAM-1 expression can be induced by pro-inflammatory cytokines, such as tumor necrosis factor-alpha (TNF-α) and interleukin-1 (IL-1), during immune responses and inflammatory conditions.

      ICAM-1 and Leukocyte Adhesion:

One of the critical functions of ICAM-1 is its involvement in leukocyte adhesion and migration. ICAM-1 interacts with its primary receptor, lymphocyte function-associated antigen-1 (LFA-1), expressed on leukocytes. This interaction facilitates the firm adhesion of leukocytes to endothelial cells, leading to their transmigration into inflamed tissues. The ICAM-1/LFA-1 axis plays a crucial role in immune surveillance, inflammation, and host defense against pathogens.

Implications of ICAM-1 in Inflammatory Disorders:

ICAM-1 has been implicated in various inflammatory diseases, including rheumatoid arthritis, inflammatory bowel disease, and multiple sclerosis. Enhanced expression of ICAM-1 on endothelial cells promotes leukocyte recruitment and contributes to the perpetuation of chronic inflammation. Therefore, targeting ICAM-1-mediated leukocyte adhesion has emerged as a potential therapeutic strategy to alleviate inflammation and attenuate disease progression.

      ICAM-1 in Viral Infections:

ICAM-1 also plays a role in viral infections, as several viruses exploit ICAM-1 to facilitate their entry into host cells. For instance, rhinoviruses, which cause the common cold, utilize ICAM-1 as a receptor for attachment and entry into respiratory epithelial cells. The interaction between ICAM-1 and viral proteins promotes viral internalization and subsequent infection. Understanding the mechanisms of ICAM-1-mediated viral entry can aid in the development of antiviral strategies.

      Therapeutic Targeting of ICAM-1:

Given its crucial involvement in immune responses and disease pathogenesis, ICAM-1 has emerged as a potential therapeutic target. Strategies aimed at blocking ICAM-1/LFA-1 interactions have shown promise in preclinical and clinical studies. Monoclonal antibodies targeting ICAM-1 or LFA-1 have been developed to prevent leukocyte adhesion and reduce inflammation. Additionally, small molecule inhibitors and gene therapies targeting ICAM-1 expression are being explored as potential therapeutic interventions.

      Challenges and Future Directions:

Although therapeutic targeting of ICAM-1 shows promise, several challenges need to be addressed. Specific targeting of ICAM-1 without affecting its physiological functions and potential off-target effects are important considerations. Additionally, the complex and dynamic nature of ICAM-1 expression and regulation require further investigation to optimize therapeutic strategies.

      Conclusion:

The investigation of ICAM-1 protein provides insights into its pivotal role in immune responses, leukocyte adhesion, and inflammatory processes. Understanding the molecular mechanisms and functional implications of ICAM-1 opens avenues for the development of targeted therapies for inflammatory disorders and viral infections. Further research on ICAM-1 protein

    • Introduction

Synovial Sarcoma, X Breakpoint 1 (SSX1) is a member of a family of highly homologous synovial sarcoma X (SSX) breakpoint proteins. SSX proteins are localized to the nucleus and expressed in testis and some types of cancers and, hence, they are classified as C/T (cancer/testis) antigens. These proteins may serve as transcriptional repressors. SSX1 genes are involved in the t (X;18) translocation typically found in all synovial sarcomas. This translocation leads to the fusion of the synovial sarcoma translocation gene on chromosome 18 to one of the SSX genes on chromosome X. These hybrid proteins are most likely responsible for transforming activity.

    • Introduction

Simian immunodeficiency virus, SIV, is a retrovirusthat is found in strains, having the HIV-1and HIV-2specific strains that infect humans. SIV strains may cause an AIDS-like immune deficiency known as SAIDS (simian acquired immunodeficiency syndrome) if they cross species boundaries. HIV-2 is more similar to SIV strains than to HIV-1, suggesting for the first time the simian origin of HIV. HIV-2 is derived from the SIV strain found in sooty mangabeys whereas HIV-1, the predominant virus found in humans, is derived from SIV strains infecting chimpanzees.

    • Introduction

RARRES2 is a secreted chemotactic protein that initiates chemotaxis through the ChemR23 G protein-coupled seven-transmembrane domain ligand. RARRES2 is upregulated by the synthetic retinoid tazarotene and found in a vast variety of tissues. RARRES2 acts as an adipokine, and is truncated on both termini from the proprotein. RARRES2 is structurally related to the cathelicidin precursors, cystatin C and kininogens. RARRES2 promotes calcium mobilization and chemotaxis of immature dendritic cells and macrophages. RARRES2 is secreted as a precursor of little biological activity, which requires proteolytic cleavage of its COOH-terminal domain to be exchangeed into a potent and highly specific agonist of ChemR23. RARRES2 signals via its receptor, ChemR23 (CMKLR1), as a positive regulator of adipocyte differentiation and metabolic function. The Chemerin receptor acts as a coreceptor for SIV and some primary HIV-1 strains. The Chemerin receptor has another ligand, called tazarotene-induced gene.

MGAT2 is an enzyme which takes part in the catalyzation of a crucial step in the reaction of oligomannose which converts to complex N-glycans. MGAT2 has three domains, classic to glycosyltransferase: short N-terminal cytoplasmic domain, a C-terminal catalytic domain and hydrophobic non-cleavable signal-anchor domain. The enzyme MGAT2 is encoded by the MGAT2 gene in humans. There are no introns in the DNA coding the gene, therefore mutations in the MGAT2 will result in carbohydrate-deficient glycoprotein syndrome, type II.

    • Introduction

The cytokine IL-1 alpha or hematopoietin 1 is a member to the interleukin 1 family, encoded by the IL1A gene in humans. Essentially Il-1 is in charge of inflammation and the rising of fever and sepsis. In order to disturb the mentioned processes and treatment for diseases, inhibitors for Interleukin 1 alpha are being developed. Neutrophils and macrophages are the main activators of IL-1 alpha, as well as endothelial and epithelial cells. By binding to the il1 receptor it is a major part in the immune response regulation. In the activation process of tumor necrosis factor-alpha the IL1A has a part as well, and has physiological, metabolic and hematopoietic activities.

    • Introduction

CHMP4A is a member of the SNF7 family and operates as chromatin-modifying protein. CHMP4A is a key component of the endosomal sorting vital for transport complex III (ESCRT-III) which takes part in multivesicular bodies (MVBs) formation and sorting of endosomal cargo proteins into MVBs. Additionally, during HIV-1 infection, the virus utilizes the ESCRT-III complex to facilitate budding and exocytosis of viral proteins through the connection of CHMP4 and a protein engaged by HIV-1 p6, which exists in viral Gag assembly and budding. CHMP4A is expressed in higher quantities in skeletal muscle, kidney, liver and heart.

    • Introduction

The heat-shock response is elicited by exposure of cells to thermal and chemical stress and through the activation of HSFs (heat shock factors) results in the elevated expression of heat-shock induced genes. Heat shock factor binding protein-1 (HSBP1), is a 76-amino-acid protein that binds to heat shock factor 1(HSF1), which is a transcription factor involved in the HS response. During HS response, HSF1 undergoes conformational transition from an inert non-DNA-binding monomer to active functional trimers. HSBP1 is nuclear-localized and interacts with the active trimeric state of HSF1 to negatively regulate HSF1 DNA-binding activity. Overexpression of HSBP1 in mammalian cells represses the transactivation activity of HSF1. When overexpressed in C.elegans HSBP1 has severe effects on survival of the animals after thermal and chemical stress consistent with a role of HSBP1 as a negative regulator of heat shock response.

    • Introduction

Uteroglobin (SCGB1A1) which belongs to the Secretoglobin (SCGBs) superfamily, is a multifunctional protein that exerts anti-inflammatory and anti-tumorigenic effects by binding small hydrophobic molecules such as phospholipids and prostaglandins. Uteroglobin is involved in numerous functions including anti-inflammation, inhibition of phospholipase A2 and the sequestering of hydrophobic ligands. SCGB1A1 is expressed by Clara cells, the non-ciliated, non-mucous secretory cells predominant in lung bronchioles, and by other epithelia which communicate with the external environment. On top of sequestering pro-inflammatory mediators and carcinogens, Uteroglobin is implicated in the inhibition of cell migration and invasion, platelet aggregation, and T cell differentiation. SCGB1A1 gene defects are associated with a susceptibility to asthma.
