    • Introduction

Cytochrome c oxidase subunit VB (COX5B) is the terminal enzyme of the mitochondrial respiratory chain. COX5B is a multi-subunit enzyme complex which couples the transfer of electrons from cytochrome c to molecular oxygen and contributes to a proton electrochemical gradient across the inner mitochondrial membrane. There are two isoforms of COX5:COX5a and COX5b. Transcription of COX5A (the aerobic isoform) is up-regulated as the rate of cellular respiration increases, when oxygen levels within the cell are high. However, when oxygen levels are low, COX5B (the hypoxic isoform) transcription increases and functions to maximize the turnover rate of the COX apoenzyme.

    • Introduction

NAGAT (ABO) is a member of the glycosyltransferase 6 family. The ABO protein is the basis of the ABO blood group system and related to the first discovered blood group system, ABO. The allele that is present in an individual determines the blood group. The histo-blood group ABO is comprised of 3 carbohydrate antigens: A, B, and H. A, B, and AB individuals express a glycosyltransferase activity which converts the H antigen to the A antigen (by addition of UDP-GalNAc) or to the B antigen (by addition of UDP-Gal), whereas O individuals are deficient of such activity.

    • Introduction

Platelet-derived growth factor D (PDGFD) belongs to the platelet-derived growth factor family. PDGFD gene product only forms homodimers and, thus, does not dimerize with the other 3 family members. PDGFD has an imperative role in wound healing. PDGFD induces macrophage recruitment, increased interstitial pressure, and blood vessel maturation during angiogenesis. PDGFD initiates events which lead to a mesangial proliferative glomerulonephritis, including influx of monocytes and macrophages and production of extracellular matrix. The 4 members of the PDGF family are mitogenic factors for cells of mesenchymal origin and are distinguished by a core motif of eight cysteines, 7 of which are found in this factor. PDGFD differs from alpha and beta members of this family by having an odd N-terminal domain, the CUB domain.

    • Introduction

NEDD8 is part of the ubiquitin family. Human NEDD8 shares 60% amino acid sequence homology to ubiquitin. The NEDD8 system is essential for the regulation of protein degradation pathways involved in cell cycle progression, morphogenesis and tumorigenesis. NEDD8 is involved in cell cycle control and embryogenesis. Covalent attachment to its substrates requires prior activation by the E-1 complex UbE1c- appbp1 and linkage to the E-2 enzyme UbE2m. Attachment of NEDD8 to cullins activates their associated E-3 ubiquitin ligase activity, and thus promotes polyubiquitination and proteasomal degradation of cyclins and other regulatory proteins.

    • Background

N-terminal pro-brain natriuretic peptide, or NT-proBNP, has emerged as a significant biomarker in cardiovascular medicine, providing crucial insights into heart function and aiding in the diagnosis and management of heart-related conditions. This peptide, released in response to cardiac stress, reflects the heart's intricate signaling mechanisms and physiological responses. The study of NT-proBNP, especially in its recombinant form, has not only deepened our understanding of heart diseases but has also led to the development of advanced diagnostic tools and potential therapeutic interventions. This research delves into the world of NT-proBNP Recombinant Protein, exploring its biochemical intricacies, physiological roles, and its profound impact on cardiovascular healthcare.

Structural and Molecular Complexity of NT-proBNP:

NT-proBNP, derived from proBNP through enzymatic cleavage, is a stable peptide fragment. It acts as a precursor to the biologically active B-type natriuretic peptide (BNP) hormone. Understanding the molecular structure and processing of NT-proBNP is essential for unraveling the regulatory mechanisms governing its release and its significance in cardiovascular physiology.

      Physiological Significance in Heart Function:

BNP and its precursor, NT-proBNP, play pivotal roles in cardiac homeostasis. They are secreted by ventricular myocardial cells in response to increased wall stress, primarily due to volume and pressure overload. Elevated levels of NT-proBNP serve as a sensitive indicator of heart failure, reflecting the heart's struggle to maintain effective circulation. Monitoring NT-proBNP levels aids clinicians in diagnosing heart failure, assessing its severity, and guiding therapeutic strategies, ultimately improving patient outcomes.

      Diagnostic Applications and Clinical Relevance:

NT-proBNP assays, often utilizing recombinant NT-proBNP proteins, have become cornerstones in cardiovascular diagnostics. Elevated levels of NT-proBNP are indicative of heart failure, providing vital information for early intervention and personalized treatment plans. Moreover, the peptide's utility extends to predicting cardiovascular events, aiding in risk stratification among individuals with or at risk of heart diseases. The incorporation of NT-proBNP testing into clinical practice has significantly enhanced the accuracy and timeliness of cardiovascular disease diagnosis.

      Potential Therapeutic Implications:

Research into NT-proBNP’s regulatory pathways has opened avenues for therapeutic interventions. Targeted therapies aimed at modulating the BNP system are under investigation, offering potential strategies for managing heart failure and related conditions. By understanding the complex interplay between NT-proBNP, its receptor systems, and cardiac function, researchers are exploring novel therapies to optimize cardiovascular health.

    • Introduction

The Melanoma Inhibitory protein (MIA) was identified as an inhibitor of in vitro growth of malignant melanoma cells. The protein contains a SH3 domain.
MIA acts as a potent tumor cell growth inhibitor for malignant melanoma cells and some other neuroectodermal tumors, including gliomas, in an autocrine fashion. In a study of human melanoma cell lines with different metastatic capacity MIA mRNA expression appeared to be inversely correlated with pigmentation. MIA has been shown to represent a very sensitive and specific serum marker for systemic malignant melanoma that might be useful for staging of primary melanomas, detection of progression from localized to metastatic disease during follow-up, and monitoring therapy of advanced melanomas.

    • Introduction

Alpha-synuclein or SNCA is a synuclein protein. SNCA mainly found in the brain, small concentration of the protein can also be located in other tissues such as heart and muscle. When looking in the brain tissue, SNCA is located in the end of the neuron, in an area called presynaptic terminal. In the presynaptic terminal SNCA has interaction with phospholipids & other proteins. Neurotransmitters are released from the synaptic vesicles in the Presynaptic terminals and act as messengers. Once released, the neurotransmitters send signals across the neurons that are crucial for the brain’s operation.

    • Introduction

TIE-1 (tyrosine kinase with Ig and EGF homology domains 1) and TIE-2/Tek comprise a receptor tyrosine kinase (RTK) subfamily with unique structural characteristics: two immunoglobulin-like domains flanking three epidermal growth factor (EGF)-like domains and followed by three fibronectin type III-like repeats in the extracellular region and a split tyrosine kinase domain in the cytoplasmic region. These receptors are expressed primarily on endothelial and hematopoietic progenitor cells and play critical roles in angiogenesis, vasculogenesis and hematopoiesis. Human TIE-1 cDNA encodes a 1122 amino acid (aa) residue precursor protein with an 18 residue putative signal peptide, a 726 residue extracellular domain and a 353 residue cytoplasmic domain. Two ligands, angiopoietin-1 (Ang1) and angiopoietin-2 (Ang2), which bind TIE-2 with high-affinity have been identified. Ang2 has been reported to act as an antagonist for Ang1. Mice engineered to overexpress Ang2 or to lack Ang1 or Tie-1 display similar angiogenic defects.

    • Background

BACE2 protein, a member of the beta-secretase family, has gained attention as a key player in the pathogenesis of neurological disorders, particularly Alzheimer's disease. This research aims to explore the function and potential therapeutic implications of BACE2 protein in neurodegenerative conditions. Understanding the role of BACE2 protein can provide valuable insights into its significance as a therapeutic target for the development of novel treatment strategies.

      Function of BACE2 Protein:

BACE2 is a transmembrane aspartic protease predominantly expressed in the central nervous system. It exhibits distinct cleavage activity on various protein substrates, including neuregulins, APP-like proteins, and TGF-β. Unlike its close homolog BACE1, BACE2 has been proposed to have non-amyloidogenic processing capabilities and has shown potential neuroprotective effects.

Implications of BACE2 Protein in Alzheimer's Disease:

Alzheimer's disease is characterized by the accumulation of amyloid-beta (Aβ) peptides in the brain, which are generated through the sequential cleavage of amyloid precursor protein (APP). BACE1 is primarily responsible for the cleavage of APP, leading to the production of toxic Aβ peptides. In contrast, BACE2 has been suggested to compete with BACE1, thereby reducing the levels of Aβ generation. This has led to speculation about the neuroprotective role of BACE2 and its potential as a therapeutic target for Alzheimer's disease.

      BACE2 Protein and Neuronal Survival:

Emerging evidence suggests that BACE2 may play a role in promoting neuronal survival and function. Studies have shown that BACE2 deficiency leads to impaired synaptic plasticity, reduced dendritic branching, and altered neurotransmitter release. BACE2 has also been implicated in the regulation of axonal growth and guidance during development. These findings highlight the potential importance of BACE2 in maintaining neuronal integrity.

Association of BACE2 Protein with Other Neurological Disorders:

Apart from Alzheimer's disease, BACE2 has been implicated in other neurological conditions as well. Genetic studies have identified BACE2 gene variants associated with an increased risk of Parkinson's disease, suggesting its involvement in the pathogenesis of this disorder. Furthermore, BACE2 has been linked to the regulation of insulin signaling and glucose homeostasis, making it a potential target for diabetes-associated cognitive decline.

      Therapeutic Implications of BACE2 Protein:

Given its potential neuroprotective effects and modulatory role in amyloid processing, BACE2 protein has emerged as a promising therapeutic target for neurodegenerative disorders. Strategies aimed at enhancing BACE2 activity or selectively activating BACE2-mediated non-amyloidogenic processing pathways hold promise for reducing amyloid pathology and preserving neuronal function. However, further research is needed to better understand the complex mechanisms underlying BACE2 function and to develop safe and effective therapeutic interventions.

      Conclusion:

The investigation of BACE2 protein has provided valuable insights into its role in neurodegenerative diseases, particularly Alzheimer's disease. The potential neuroprotective effects and modulation of amyloid processing pathways by BACE2 make it an intriguing therapeutic target. Future studies should focus on unraveling the precise mechanisms by which BACE2 influences disease pathogenesis and developing strategies to harness its therapeutic potential. The exploration of BACE2 protein opens new avenues for the development of innovative treatment approaches for neurodegenerative disorders.

Greater than 90.0% as determined by SDS-PAGE.

    More Info

    • Synonyms

Angiopoietin-1 receptor precursor, Tyrosine-protein kinase receptor TIE-2, hTIE2, Tyrosine-protein kinase receptor TEK, p140 TEK, Tunica interna endothelial cell kinase, CD202b, VMCM, VMCM1, TIE2.

    • Physical Appearance

Sterile Filtered White lyophilized (freeze-dried) powder.

    • Stability

Lyophilized sTIE-2 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution TEK should be stored at 4°C between 2-7 days and for future use below -18°C.For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).Please prevent freeze-thaw cycles.

    • Solubility

It is recommended to reconstitute the lyophilized TIE-2 Fc Chimera in sterile water not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
