Vav 1 Guanine Nucleotide Exchange Factor (VAV1), belongs to the VAV gene family. The VAV1 proteins are guanine nucleotide exchange factors (GEFs) for Rho family GTPases which activate pathways leading to actin cytoskeletal rearrangements and transcriptional alterations. VAV1 is essential in hematopoiesis, taking part in T-cell and B-cell development and activation. VAV1 has been identified as the specific binding partner of Nef proteins from HIV-1. In addition, VAV1 contains an SH2 domain, which could show its function as a substrate for tyrosine kinases. Expression of VAV1 is limited solely to cells of hematopoietic origin, as well as those of the erythroid, lymphoid and myeloid lineages. Endophilin-B1 (SH3GLB1) is a part of the endophilin family and highly expresses in heart, skeletal muscle, kidney and placenta. Endophilin B1 belongs to the B subgroup of the endophilin family which is needed for preservation of mitochondrial morphology and for the regulation of the outer mitochondrial membrane dynamics. SH3GLB1 is required for normal outer mitochondrial membrane dynamics. Furthermore, SH3GLB1 is required for coatomer-mediated retrograde transport in certain cells. BPI fold containing family A Member 1 (BPIFA1) belongs to the short subfamily of PLUNC family proteins and have homology only to the Nterminal domains of BPI. BPIFA1 is a secreted protein which is expressed in the secretory ducts and submucosal glands of tracheobronchial tissues. BPIFA1 binds to LPS (lipopolysaccharide) in nasal lavage fluid (NLF) which alludes to its role in the inflammatory response of the upper airways after exposure to irritants. BPIFA1 may also serve as a potential molecular marker for detection of micrometastasis in non-small-cell lung cancer. Decreased levels of BPIFA1 are observed in the NLF of smokers and people who have been exposed to reactive epoxy chemicals, which indicats that long-term exposure to airway irritants damages the production of BPIFA1 in the upper respiratory tract.

COP9 signalosome complex subunit 8 isoform 1 (COPS8) is one of the 8 subunits of COP9 signalosome, which is a much conserved protein complex that functions as an imperative regulator in multiple signaling pathways. The structure and function of COP9 signalosome is analogous to that of the 19S regulatory particle of 26S proteasome. COP9 signalosome interacts with SCF-type E3 ubiquitin ligases and acts as a positive regulator of E3 ubiquitin ligases.

PRDX2 is part of the peroxiredoxin family of antioxidant enzymes, which reduce hydrogen peroxide and alkyl hydroperoxides. Methionine sulfoxide reductase B1 (SEPX1 or MSRB1), is a selenoprotein that contains a selenocysteine (Sec) residue at its active site. The selenocysteine is encoded by the UGA codon that usually signals translation termination. SEPX1 is a member of the methionine sulfoxide reductase B (MsrB) family, and is expressed in an assortment of adult and fetal tissues. MSRs (Methionine sulfoxide reductases) catalyze the reduction of free and protein-bound methionine sulfoxides to corresponding methionines. The oxidation of methionine by ROS creates a diastereomeric mixture of methionine-S-sulfoxide (Met-S-SO) and methionine-R-sulfoxide (Met-R-SO). Two separate enzyme families evolved for reduction of these sulfoxides, with methionine-S-sulfoxide reductase (MsrA) being stereospecific for Met-S-SO and methionine-R-sulfoxide reductase (MsrB) for Met-R-SO.

Sialic acid-binding Ig-like lectin 6 isoform 1 (SIGLEC6) is a member of immunoglobulin superfamily and SIGLEC (sialic acid binding Ig-like lectin) family. SIGLEC6 mediates sialic-acid dependent binding to cells and binds to alpha-2, 6-linked sialic acid. The SIGLEC6 protein localizes in numerous compartments such as membrane fraction, extracellular region and so on. The SIGLEC6 receptor binds sialyl-TN glycans and leptin. Placental expression SIGLEC6 is upregulated in preeclampsia.

AURKA (Aurora Kinase A) belongs to the mitotic serine/threonine kinases family. AURKA is a cell cycle-regulated kinase which may be involved in microtubule formation and/or stabilization at the spindle pole during chromosome segregation. AURKA is found at the centrosome in interphase cells and at the spindle poles in mitosis. Since the AURKA expression is cell-cycle regulated, it is low in G1/S, it accumulates during G2/M, and it decreases rapidly after. AURKA is involved in important processes during mitosis and meiosis whose proper function is essential for healthy cell proliferation. In addition, AURKA plays an essential role in tumourigenesis and is overexpressed in various types of cancers. AURKA is strongly expressed in the testis, colon, ovarian, prostate, neuroblastoma, breast and cervical cancer cell lines and weakly in skeletal muscle, thymus and spleen. AURKA interacts with its substrates BORA and ARHGEF2, as well as with TACC1 and CPEB1.
Defects in the AURKA gene cause numerical centrosome aberrations including aneuploidy. AURKA overexpression has been linked to chromosomal instability in colorectal cancer. AURKA expression may have a prognostic significance in ovarian carcinoma.

Betaine-Homocysteine Methyltransferase 2 (BHMT2) is involved in the regulation of homocysteine metabolism. Homocysteine is a sulfur-containing amino acid which has a key role in methylation reactions. Transfer of the methyl group from betaine to homocysteine generates methionine, which donates the methyl group to methylate DNA, proteins, lipids, and other intracellular metabolites. BHMT2 is one of two methyl transferases which can catalyze the transfer of the methyl group from betaine to homocysteine. BHMT2 converts homocysteine to methionine using S-methylmethionine (SMM) as a methyl donor. Homocysteine metabolism anomalies are implicated in disorders varying from vascular disease to neural tube birth defects such as spina bifida.

PSME3 is a member of the PA28 family. The 26S proteasome is an extremely organized multicatalytic proteinase complex composed of 2 complexes, a 20S core and a 19S regulator. Proteasomes are spread all over the eukaryotic cells in large quantities and cleave peptides in an ATP/ubiquitin-dependent process in a non-lysosomal pathway. PSME3 stimulates the trypsin-like catalytic subunit of the proteasome and inhibits the chymotrypsin-like and postglutamyl-preferring (PGPH) subunits. PSEM3 enables the MDM2-p53/TP53 collaboration that encourages ubiquitination- and MDM2-dependent proteasomal degradation of p53/TP53, restricting its growth and causing inhibited apoptosis after DNA damage.

VCAM1 is a member of the Ig superfamily. It is a cell surface sialoglycoprotein expressed by cytokine activated endothelium. VCAM-1 contains 6 or 7 immunoglobulin domains, and is expressed on both large and small vessels only after the endothelial cells are stimulated by cytokines. The protein has a number of functions including the regulation of leukocyte migration, leukocyte endothelial cell adhesion and signal transduction and may play a role in a number of inflammatory diseases (artherosclerosis and rheumatoid arthritis).
