    • Introduction

The Defensin family are highly similar in their protein sequence and are microbicidal & cytotoxic peptides made by neutrophils. Beta Defensin-1 is an antimicrobial peptide having the resistance of epithelial surfaces to microbial colonization. Beta Defensin-1 has close proximity to Defensin Alpha-1 and has been implicated in the pathogenesis of cystic fibrosis.
Skin of patients having atopic dermatitis patients and mycosis fungoides (non-lesional and lesional) show lower human Beta Defensin-1 mRNA expression and higher human Beta Defensin-2 and human Beta Defensin-3 mRNA expression.
Beta Defensin is highly expressed by epithelial cells.
Beta-defensin 1 may play a role in the pathogenesis of severe sepsis.

    • Synonyms

      Beta-defensin 1, BD-1, Defensin beta 1, hBD-1, HBD1, HBP1, DEFB1, HBD-1, HBP-1, DEFB101, DEFB-1, MGC51822.

    • Introduction

SARS Coronavirus is an enveloped virus containing three outer structural proteins, namely the membrane (M), envelope (E), and spike (S) proteins. Spike (S)-glycoprotein of the virus interacts with a cellular receptor and mediates membrane fusion to allow viral entry into susceptible target cells. Accordingly, S-protein plays an important role in virus infection cycle and is the primary target of neutralizing antibodies. It has recently been shown that SARS is caused by a human coronavirus. Human coronaviruses are the major cause of upper respiratory tract illness in humans, such as the common cold. Coronaviruses are positive-stranded RNA viruses, featuring the largest viral RNA genomes known to date (27-31 kb). The first step in coronavirus infection is binding of the viral spike protein, a 139-kDa protein, to certain receptors on host cells. The spike protein is the main surface antigen of the coronavirus. The most prominent protein in the culture supernatants infected with SARS virus is a 46 kDa nucleocapsid protein. This suggests that the nucleocapsid protein is a major immunogen that may be useful for early diagnostics.

    • Introduction

CD5 is a suitable immunohistochemical marker for T-cells, though not as sensitive as CD3. Approximately 76% of T-cell neoplasms are reported to express CD5, and it is also observed in chronic lymphocytic leukemia, hairy cell leukemia, and mantle cell lymphoma cells. CD5 is commonly lost in cutaneous T-cell lymphoma, and its deficiency can be used as an indicator of malignancy in this condition. CD5’s absence in T cell acute lymphoblastic leukemia, though relatively rare, is linked with a poor prognosis. CD5 serves as a receptor in regulating T-cell proliferation.

    • Introduction

CD19, also known as B-Lymphocyte Surface Antigen B4, belongs to the Ig superfamily which is expressed on the surface of all B-lymphoid cells with the exception of terminally differentiated plasma cells. CD19 is involved as a signal-transducing receptor in the control of differentiation as well as proliferation. CD19 is as an adaptor protein which drafts cytoplasmic signaling proteins to the membrane and operates within the CD19/CD21 complex to decrease the threshold for B cell receptor signaling pathways. CD19 is present on all B cells, and therefore is a biomarker for lymphoma diagnosis, B lymphocyte development and can be used as a target for leukemia immunotherapies.

    • Introduction

CD81 antigen isoform 1, also known as CD81, is part of the tetraspanin family. CD81 interacts directly with immunoglobulin superfamily member 8 (IGSF8, CD316) and CD36. In B cells, CD81 is part of a complex with CD21, CD19 and Leu13. Furthermore, this complex decreases the threshold for B cell activation through B cell receptors by linking Ag specific recognition and CD21 mediator recognition. While on T cells CD81 connects with CD4 and CD8 and provides a costimulatory signal with CD3.

Clusterin also named Apolipoprotein J (APO-J) is a 75-80 kD disulfide-linked heterodimeric protein containing about 30% of N-linked carbohydrate rich in sialic acid but truncated forms targeted to the nucleus have also been identified. The precursor polypeptide chain is cleaved proteolytically to remove the 22-mer secretory signal peptide and subsequently between residues 227/228 to generate the ? and ? chains. These are assembled in anti-parallel to give a heterodimeric molecule in which the cysteine-rich centers are linked by five disulfide bridges and are flanked by two predicted coiled-coil ? -helices and three predicted amphipathic a-helices. Across a broad range of species clusterin shows a high degree of sequence homology ranging from 70% to 80%. It is nearly ubiquitously expressed in most mammalian tissues and can be found in plasma, milk, urine, cerebrospinal fluid and semen. It is able to bind and form complexes with numerous partners such as immunoglobulins, lipids, heparin, b

CD27 belongs to the TNF-receptor superfamily. CD27 is necessary for initiation and long-term maintenance of T cell immunity. CD27 binds to ligand CD70, and has a crucial role in regulating B-cell activation and immunoglobulin synthesis. The CD27 receptor transduces signals which result in the activation of NF-kappaB and MAPK8/JNK. Adaptor proteins TRAF2 and TRAF5 mediate the signaling process of CD27. CD27-binding protein (SIVA), which is a proapoptotic protein, can bind to the CD27 receptor and is believed to have a significant role in the apoptosis induced by CD27.

    • Introduction

STAM Binding Protein (STAMBP) is a zinc metalloprotease which specifically cleaves Lys-63-linked polyubiquitin chains, however it doesn’t cleave Lys-48-linked polyubiquitin chains. STAMBP has a role in signal transduction for cell growth and MYC induction mediated by IL-2 and GM-CSF. In addition, STAMBP potentiates BMP signaling by antagonizing the inhibitory action of SMAD6 and SMAD7. STAMBP also has a major role in regulation of cell surface receptor-mediated endocytosis and ubiquitin-dependent sorting of receptors to lysosomes. Endosomal localization of STAMBP is essential for efficient EGFR degradation but not for its internalization. STAMBP is also involved in the negative regulation of PI3K-AKT-mTOR and RAS-MAP signaling pathways.

    • Background

CD4 protein, also known as Cluster of Differentiation 4, is a cell surface glycoprotein primarily expressed on helper T cells. It plays a crucial role in immune system regulation by mediating interactions between T cells and antigen-presenting cells (APCs). This research aims to explore the function, signaling pathways, and significance of CD4 protein in T cell biology and disease pathogenesis.

      Function of CD4 Protein:

CD4 protein serves as a co-receptor for the T cell receptor (TCR) and interacts with major histocompatibility complex class II (MHC-II) molecules on APCs. This interaction enables CD4+ T cells to recognize and respond to antigenic peptides presented by MHC-II. CD4 protein enhances TCR signaling and facilitates the formation of immunological synapses between T cells and APCs, thereby promoting efficient T cell activation, proliferation, and differentiation.

Role of CD4 Protein in T Cell Differentiation:

CD4 protein is involved in determining the differentiation fate of CD4+ T cells. By interacting with specific cytokines secreted by APCs, CD4 protein directs the differentiation of naive T cells into distinct effector T cell subsets, including Th1, Th2, Th17, and regulatory T cells (Tregs). Each subset exhibits unique functions and cytokine profiles, contributing to immune responses, inflammation, and immune tolerance.

Signaling Pathways Activated by CD4 Protein:

Upon engagement with MHC-II, CD4 protein initiates signaling cascades that regulate T cell activation and differentiation. The cytoplasmic tail of CD4 contains conserved motifs, such as immunoreceptor tyrosine-based activation motifs (ITAMs) and proline-rich regions, which recruit and activate various signaling molecules, including protein kinases and adaptor proteins. These signaling events culminate in the activation of transcription factors, such as NF-κB and AP-1, which control gene expression required for T cell function.

Implications of CD4 Protein in Disease Pathogenesis:

Dysregulation of CD4 protein expression or function can lead to immune dysfunctions and contribute to the development of various diseases. CD4 protein is a primary receptor for human immunodeficiency virus (HIV) entry into CD4+ T cells, resulting in the destruction of these crucial immune cells and the progression of acquired immunodeficiency syndrome (AIDS). Moreover, altered CD4+ T cell responses and imbalances in T cell subsets have been associated with autoimmune diseases, allergic reactions, and chronic inflammatory conditions.

      Conclusion:

The investigation of CD4 protein in T cell biology provides valuable insights into its role in immune system regulation and disease pathogenesis. Understanding the function and signaling pathways of CD4 protein enhances our knowledge of T cell activation, differentiation, and immune responses. Further research on CD4 protein may lead to the development of targeted therapies for immune-related disorders and provide novel strategies for immune modulation.

    • Introduction

CD8B encodes the CD8 beta chain isoforms of the CD8 antigen. CD8 is a cell surface glycoprotein which facilitates efficient cell-cell interactions within the immune system and is located on most cytotoxic T lymphocytes. The T-cell receptor on the T lymphocyte and the co-receptor CD8 antigen identify antigens exposed by an antigen presenting cell (APC) in the context of class I MHC molecules. The functional co-receptor is either a heterodimer composed of one alpha and one beta chain or a homodimer composed of two alpha chains. Both alpha and beta chains share substantial homology to immunoglobulin variable light chains.

    • Introduction

Treponema pallidum is a gram-negative spirochaete bacterium and is considered to be metabolically crippled. There are at least four known subspecies: T. pallidum pallidum, T. pallidum pertenue, T. pallidum carateum and T. pallidum endemicum. The helical structure of T. pallidum pallidum allows it to move in a corkscrew motion through viscous mediums such as mucus. Treponema pallidum sub sp. pallidum has one of the smallest bacterial genomes at 1.14 million base pairs (Mb) and has limited metabolic capabilities, reflecting its adaptation through genome reduction to the rich environment of mammalian tissue.

    • Stability

Protein should be stored at 4°C.Refrigerate Upon arrival. DO NOT FREEZE.

    • Applications

Treponema Pallidum protein is suitable for ELISA and Western blots, excellent antigen for detection of Trp. Pallidum with minimal specificity problems.

    • Introduction

CD4 is a cell-surface glycoprotein found on the mature helper T cells and immature thymocytes, as well as on monocytes and macrophages. (Some cytotoxic T cells have CD4 protein as well.) Normally, about 65% of T cells in the blood are CD4+ (have CD4 protein protruding from their membrane). A mature T cell with either have CD4 or CD8, but not both. During one stage of development T cells develop CD4 and CD8 receptors, but they eventually are differentiated in the thymus and become more specialized.

    • Introduction

Borrelia belongs to a genus of bacteria of the spirochete phylum. Borrelia causes borreliosis, which is a zoonotic, vector-borne disease transmitted mainly by ticks and some by lice, depending on the species. Of the 36 known species of Borrelia, 12 are distinguished to cause Lyme disease or borreliosis and are transmitted by ticks. The main Borrelia species causing Lyme disease are Borrelia burgdorferi, Borrelia afzelii, and Borrelia garinii. The Borrelia genus members have a linear chromosome which is about 900 kbp in length as well as an excess of both linear and circular plasmids in the 5-220 kbp size range. The plasmids are atypical, as compared to most bacterial plasmids, since they contain many paralogous sequences, a large number of pseudogenes and, in some cases, essential genes. Moreover, a number of the plasmids have features suggesting that they are prophages.
