    • Introduction

ANGPTL3 and ANGPTL4 are angiopoietin-like proteins secreted and expressed mainly by the liver, their role being the regulation of triglyceride metabolism by inhibiting the lipolysis of triglyceride-rich lipoproteins. During different nutritional states (feeding/fasting) the levels of the circulating triglycerides are regulated by Angptl3 and Angptl4 through differential inhibition of Lipoprotein lipase (LPL) as shown by the experimental data. The molecular structure of ANGPTL3 is similar to that of the angiopoietins (vascular endothelial growth factors). Deletion mutants of human Angiopoietin 5 were used in order to demonstrate that the N-terminal domain (fragment 17-207) and not the C-terminal fibrinogen-like domain (fragment 207-460) increased the plasma triglyceride levels in mice.

    • Synonyms

      Angiopoietin 5, ANGPT5, ANGPTL3, Angiopoietin Like Protein 3.

    • Stability

Store lyophilized ANGPTL3 Human at -20°C. Aliquot the product after reconstitution to avoid repeated freezing/thawing cycles. Reconstituted Angiopoietin 5 can be stored at 4°C for a limited period of time; it does not show any change after two weeks at 4°C.

    • Solubility

Add 0.1M Acetate buffer pH-4 and let the lyophilized pellet of ANGPTL3 Human dissolve completely. For conversion into higher pH value, we recommend intensive dilution by relevant buffer to a concentration of 10μg/ml. In higher concentrations the solubility of Angiopoietin 5 is limited.

    • Background

Angiopoietin-Like Protein 3 Human Recombinant: An Emerging Target in Metabolic and Cardiovascular Disorders

      Abstract:

Angiopoietin-like protein 3 (ANGPTL3) is a key regulator of lipid metabolism and has garnered considerable attention for its involvement in metabolic and cardiovascular disorders. ANGPTL3 plays a crucial role in lipid homeostasis, including the regulation of triglycerides, cholesterol, and lipoprotein metabolism. The availability of human recombinant ANGPTL3 protein has provided a valuable tool for investigating its biological functions and therapeutic potential. This review aims to provide a comprehensive overview of the role of ANGPTL3 in metabolic disorders, cardiovascular diseases, and lipid metabolism, highlighting the potential of ANGPTL3 human recombinant protein as a therapeutic target.

      Introduction:

Metabolic disorders, such as dyslipidemia and obesity, significantly contribute to the development of cardiovascular diseases. ANGPTL3, a member of the angiopoietin-like protein family, has emerged as a key player in lipid metabolism and cardiovascular health. ANGPTL3 regulates lipoprotein metabolism, affecting triglyceride-rich lipoproteins, low-density lipoproteins (LDL), and high-density lipoproteins (HDL).

      Molecular Mechanisms of ANGPTL3 Action:

ANGPTL3 exerts its effects through inhibition of lipoprotein lipase (LPL) and endothelial lipase (EL), key enzymes involved in lipoprotein metabolism. By inhibiting LPL and EL activities, ANGPTL3 increases plasma triglyceride and LDL cholesterol levels. ANGPTL3 also influences hepatic cholesterol metabolism and HDL metabolism through modulation of the receptor-mediated uptake of lipoproteins.

Role of ANGPTL3 in Metabolic Regulation:

ANGPTL3 plays a critical role in metabolic regulation, particularly in lipid metabolism and dyslipidemia. Loss-of-function mutations in the ANGPTL3 gene result in decreased plasma triglycerides, LDL cholesterol, and total cholesterol levels, highlighting the potential therapeutic relevance of ANGPTL3 inhibition. Conversely, elevated ANGPTL3 levels are associated with increased cardiovascular risk and atherogenic lipid profiles.

ANGPTL3 in Cardiovascular Health and Disease:

ANGPTL3 has emerged as a key modulator of cardiovascular diseases, including atherosclerosis and coronary artery disease. ANGPTL3 influences vascular endothelial function, inflammation, and plaque formation through its effects on lipoprotein metabolism and lipid accumulation. Inhibition of ANGPTL3 has shown promising results in preclinical studies, reducing atherosclerosis and improving cardiovascular outcomes.

Therapeutic Potential of ANGPTL3 Human Recombinant Protein:

The development of ANGPTL3 human recombinant protein provides a novel avenue for therapeutic interventions targeting metabolic and cardiovascular disorders. Inhibition of ANGPTL3 using monoclonal antibodies or other approaches has demonstrated efficacy in lowering plasma lipid levels, particularly triglycerides and LDL cholesterol. Clinical trials investigating the safety and efficacy of ANGPTL3 inhibition are underway.

      Conclusion:

ANGPTL3 is a key regulator of lipid metabolism and a promising therapeutic target for metabolic and cardiovascular disorders. The availability of ANGPTL3 human recombinant protein has facilitated in-depth investigations into its biological functions and therapeutic potential. Targeting ANGPTL3 holds promise for improving lipid profiles, reducing cardiovascular risk, and managing metabolic disorders.

    • Introduction

Shiga-like toxin (verotoxin) is a toxin produced by some strains of Escherichia coli. Shiga-like toxin is named for its similarity to the AB5-type Shiga toxin produced by the bacteria Shigella dysenteriae. There are two known types-SLT1 and SLT2. The Shiga-like toxin is linked with hemolytic-uremic syndrome. Shiga-like toxin requires highly specific receptors on the cells'' surface in order to attach and enter the cell. Species such as cattle, swine, and deer which do not carry these receptors may harbor toxigenic bacteria without any ill effect, dropping them in their feces, from where they may be distributed to humans. Shiga Like Toxin-1 Subunit B has nontoxic action, it is the functional region which binds to the receptor. The Shiga Like Toxin-1 Subunit B can be useful in vaccine study, antibody test and other functional research.

    • Introduction

The enzyme Carbonic anhydrase II having an accession number of NP_414668 is also called carbonate dehydratase which is part of the enzyme family that catalyses rapid inter-conversion of carbon dioxide & water to bicarbonate, carbonic acid and protons (CO2 + H2O ? HCO3? + H+), a reaction that occurs rather slowly in the absence of a catalyst. The majority of carbonic anhydrases enclose a zinc ion in their active site and therefore is classified as metalloenzymes.
The most important function of Carbonic anhydrase is known to preserve acid-base balance in blood and other tissues, and to help transport carbon dioxide of tissues. Carbonic anhydrases have been found in all kingdoms of life. Carbonic anhydrase has 3 different classes: alpha, beta and gamma which share very little sequence or structural similarity, thus far they all perform the same function and require a zinc ion at the active site. Mammalian carbonic anhydrase is monomeric and belongs to the alpha class. Plant carbonic anhydrase is dimeric and belongs to the beta class.
Methane-producing bacteria carbonic anhydrase is trimeric and grows in hot springs which forms the gamma class.

There are several proteolytic fragments or specific domains of proteins that act as inhibitors of angiogenesis. These include fragments of plasminogen such as Angiostatin protein kringles 1-4 and kringles 1-5, Endostatin, Restin, PEX, the N-terminal fragment of prolactin, and the Nterminally truncated platelet factor. Angiostatin is a proteolytic protein fragment of plasminogen that is comprised of the first 4 kringle regions. Angiostatin k1-4 prevents the growth of endothelial cells, and its systemic administration inhibits the growth of primary carcinomas in mice. Angiostatin Kringles 1-3 segment has a larger inhibitory activity than the Angiostatin kringles 1-4 fragment. The protease-activated angiostatin kringles 1-5 is the most potent plasminogen fragment with over 50 times larger endothelial cell specific inhibitory activity. Angiostatin kringles 1-5 systemic administration inhibits growth of fibrosarcoma and significantly reduces neovascularization.
Angiostatin is an angiogenesis inhibitor in mouse serum and urine. Angiostatin is a 38 kDa protein fragment of the plasminogen composed of the 1st 4 kringle domains of plasminogen. Angiostatin K1-4 is also named plasminogen kringles 1-4 and PK1-4.
Angiostatin protein is manufactured by the protelytic cleavage of plasminogen by a serine protease from several prostate carcinoma cell lines. The manufacturing of angiostatin by pancreatic cancer cells can be inhibited by TGF-beta 1 along with plasminogen activator inhibitor type-1 (PAI1).

    • Introduction

HP-NAP protects DNA from oxidative damage by sequestering intracellular Fe2+ ion and storing it in the form of Fe3+ oxyhydroxide mineral. One hydrogen peroxide oxidizes two Fe2+ ions, thus preventing hydroxyl radical production by the Fenton reaction. HP-NAP is necessary for the survival in the presence of oxidative stress. Dps is also a virulence factor which activates neutrophils, mast cells and monocytes. HP-NAP binds to neutrophil-glycosphingolipids and to sulfated carbohydrates on mucin. HP-NAP might play a part in the accumulation of neutrophils and monocytes at the site of infection. HP-NAP induces superoxide anion generation, adhesion and chemotaxis of neutrophils, via a pertussis toxin-sensitive pathway involving MAP kinases.

    • Introduction

Carbonic Anhydrase VIII or CA8 was previously called CA-related protein due to its sequence resemblance to additional recognized carbonic anhydrase genes. Nonetheless CA8 doesn’t have carbonic anhydrase function. This protein keeps bearing a carbonic anhydrase classification because of coherent sequence similarity to additional proteins in carbonic anhydrase family. Mutations in this protein may lead to cerebellar dysequilibrium syndrome type 3 or ataxia mental retardation.

    • Introduction

HAAO is a monomeric cytosolic protein which is a member of intramolecular dioxygenases family containing nonheme ferrous iron. The HAAO protein catalyzes the synthesis of quinolinic acid (QUIN) from 3-hydroxyanthranilic acid. QUIN is an excitotoxin whose toxicity is facilitated by its ability to activate glutamate N-methyl-D-aspartate receptors. Amplified cerebral levels of QUIN may contribute to the pathogenesis of neurologic and inflammatory disorders. HAAO is widely distributed in peripheral organs, such as liver and kidney, and is also present in low amounts in the central nervous system.

    • Introduction

Resistin, a product of the RSTN gene, is a peptide hormone belonging to the class of cysteine-rich secreted proteins which is termed the RELM family, and is also described as ADSF (Adipose Tissue- Specific Secretory Factor) and FIZZ3 (Found in Inflammatory Zone). Human resistin contains 108 amino acids as a prepeptide, and its hydrophobic signal peptide is cleaved before its secretion. Resistin circulates in human blood as a dimeric protein consisting of two 92 amino acid polypeptides, which are disulfide-linked via Cys26.
Resistin may be an important link between obesity and insulin resistance. Mouse resistin, specifically produced and secreted by adipocyte, acts on skeletal muscle myocytes, hepatocytes and adipocytes themselves so that it reduces their sensitivity to insulin. Steppan et al. have suggested that resistin suppresses the ability of insulin to stimulate glucose uptake. They have also suggested that resistin is present at elevated levels in blood of obese mice, and is down regulated by fasting and antidiabetic drugs. Way et al., on the other hand, have found that resistin expression is severely suppressed in obesity and is stimulated by several antidiabetic drugs.
Other studies have shown that mouse resistin increases during the differentiation of adipocytes, but it also seems to inhibit adipogenesis. In contrast, the human adipogenic differentiation is likely to be associated with a down regulation of resistin gene expression.
