About NUCB / Nucleobindin:
Nucleobindin 2 (NUCB 2) is a multifunctional protein with many functional domains that are involved in many biological processes, including food intake and energy balance. NUCB 2 has been linked to worse malignant outcomes and cell migration in breast and prostate cancer.
NUCB in Cancer
Yet, although studies are still ongoing and the final outcome is yet to be determined, it would appear that it has no clinical or biological relevance in renal cell cancer. It would seem that the NUCB 2 level in patients with renal cell carcinoma was considerably higher than in normal control patients.
In the renal cancer cell line SK RC 52, NUCB 2 deletion reduced migration and invasion. Furthermore, NUCB 2 suppression and the zinc finger E box binding to homeobox 1 (ZEB1) dependent pathway influenced the expression levels of molecules involved with epithelial mesenchymal transition (EMT), such as E cadherin and catenin.
Nucleobindin Mechanism
The AMPK target of rapamycin complex (mTORC) 1 signaling pathway is involved in the control of NUCB 2 mediated metastasis and EMT. In a mouse renal cell carcinoma tumor model, NUCB 2 inhibition also reduced tumor nodule development. In conclusion, NUCB 2 enhanced migration, invasion, and EMT in renal cell carcinoma cells in vitro and in vivo via the AMPK/TORC1/ZEB1 pathway.
