HLA

HLA-G (Major Histocompatibility Complex Class I G) is a member of the HLA class I heavy chain paralogues. This class I molecule is a heterodimer which comprises a heavy chain as well as a light chain, beta-2 microglobulin, while the heavy chain is fixed in the membrane. HLA-G is expressed on fetal derived placental cells. HLA-G is a non-classical class-I HLA molecule linked with immuno-modulatory & anti-inflammatory properties which interacts with inhibitory receptors such as, ILT2/ILT4/KIR2DL4, that are present on different immune cells. HLA-G inhibits the proliferation of T cells, B cells & natural killer cells, moreover it also induces regulatory T cells.

Major Histocompatibility Complex Class I F (HLA-F) belongs to the MHC family which takes part in the presentation of antigens to the T cell receptor. HLA-F is a member of the class I molecules which are expressed in virtually all cells. There are 2 classes of HLA antigens. Class I molecules takes an important part in the immune system by presenting peptides derived from the endoplasmic reticulum.

Major Histocompatibility Complex Class II DR Beta 1 also known as HLA-DRB1 ia a member of the HLA class II beta chain paralogs. Molecule class II is a heterodimer consisting of an alpha (DRA) and a beta chain (DRB), both anchored in the membrane. HLA-DRB1 takes an essential part in the immune system by presenting peptides derived from extracellular proteins. Class II molecules are expressed in antigen presenting cells (APC: B lymphocytes, dendritic cells, macrophages). Furthermore, the beta chain is approximately 26- 28 kDa. It is encoded by 6 exons. While exon one encodes the leader peptide; exons 2 and 3 encode the two extracellular domains; exon 4 encodes the transmembrane domain; and exon 5 encodes the cytoplasmic tail.

Major histocompatibility complex, class II, DR alpha (HLA-DRA) belongs to the MHC class II family. HLA-DRA binds peptides derived from antigens which access the endocytic route of antigen presenting cells (APC) and presents them on the cell surface for identification by the CD4 T-cells. The peptide binding cleft accommodates peptides of 10-30 residues. The peptides presented by MHC class II molecules are generated mainly by degradation of proteins which access the endocytic route, where they are processed by lysosomal proteases and other hydrolases.

Major Histocompatibility Complex Class II DO Beta (HLA-DOB) is a part of the HLA class II beta chain paralogues. This class II beta chain is a heterodimer which is located in intracellular vesicles and consists of an alpha (DOA) and a beta chain (DOB), both anchored in the membrane. HLA-DOB which interacts with the HLA-DM molecule in B-cells is a significant modulator in the HLA class II restricted antigen presentation pathway. Class II molecules are expressed in antigen presenting cells such as B lymphocytes, dendritic cells and macrophages.

Major Histocompatibility Complex Class II DO Alpha (HLA-DOA) is a significant modulator in the HLA class II restricted antigen presentation pathway by interaction with the HLA-DM molecule in B-cells. HLA-DOA forms a heterodimer along with HLA-DOB. The heterodimer is located in lysosomes in B cells and regulates HLA-DM-mediated peptide loading on MHC class II molecules. HLA-DOA exhibits very little sequence variation, particularly at the protein level.

The HLA-C (class I molecule) is a heterodimer comprised of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is anchored in the membrane. Class I molecules have a key role in the immune system by presenting peptides derived from endoplasmic reticulum lumen. Class I molecules are expressed in virtually all cells.
