      Cadherin-5, also known as CDH5 is a classical cadherin which belongs to the cadherin superfamily. The CDH5is located in a six-cadherin cluster in a region on the long arm of chromosome 16 which is involved in loss ofheterozygosity events in breast as well as prostate cancer.       Cadherin-5 (CDH5) belongs to the atypical/type 2 subgroup of Cadherin homophilic adhesion proteins. Cadherin-5 is a calcium-dependent cell-cell adhesion molecule, which is comprised of 5 extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. Cadherin-5 protein plays a role in the formation, maturation and remodeling of the vascular wall. CDH5 is generally considered to be specific for vascular endothelia in which it is either the sole or the predominant cadherin, often co-existing with N-cadherin. Moreover, the CDH5 protein regulates or is regulated by VEGF R2, type 1 and type 2 TGF-beta receptors, and other endothelial junction proteins such as JAM-C, Claudin-5, and N-Cadherin. CDH5 also connects with alpha-catenin forming a link to the cytoskeleton. CDH5 functions jointly with KRIT1 to create and maintain correct endothelial cell polarity and vascular lumen. Furthermore, Cadherin-5 is essential for activation of PRKCZ and for the localization of phosphorylated PRKCZ, PARD3, TIAM1 and RAP1B to the cell junction.     • Introduction

      E-cadherin (uvomorulin, cell-CAM120/80) is a calcium dependent cell adhesion molecule expressed predominately in epithelial tissues. It plays an important role in the growth and development of cells via the mechanisms of control of tissue architecture and the maintenance of tissue integrity. Numerous studies have demonstrated that reduction and/or loss of Ecadherin expression in carcinomas correlates positively with the potential of these tumors for invasion and metastasis.

Upon reconstitution CDH1 should be stored at 4°C between 2-7 days and for future use below -18°C.For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).Please prevent freeze-thaw cycles.

About CDH / Cadherin:

A cadherin, or CDH, is a type of cell adhesion molecule that facilitates the formation of adherens junctions. These junctions play a critical role in enabling cells to bind to one another, hence CDH is also an important element in this process. There are numerous classes of cadherin molecules. Although there are believed to be over 100 types of CDHs, they are usually split into four groups: classical, protocadherins, desmosomal and unconventional

CDH Function
CDHs act as ligands and receptors for other molecules. They assist in the development of cells, specifically the separation of tissue layers and cellular migration. Different cadherins are required for different functions, hence they are present at varying stages of cell development.
Following development, CDHs help to maintain tissue and cell structure, as well as playing a role in facilitating cellular movement.

Cadherin Mechanism
Adhesions between two cells are mediated by extracellular cadherin domains, while intracellular cytoplasmic tails are facilitated by multiple adaptors and signaling proteins. Collectively, these adaptors and signaling proteins are known as the cadherin adhesome.

CDH Interactions
Cadherins on one cell interact with cadherins on different cells to facilitate adhesions.

Cadherin Structure
Synthesized as polypeptides, cadherins are modified numerous times before they impact on cellular migration and adhesions. Typically, these peptides are around 720–750 amino acids long. Every CDH has a transmembrane component, which is comprised of a single chain glycoprotein repeats, and a small C-terminal cytoplasmic component, while the remaining protein is extra-cellular. In order to function, CDHs form two identical subunits: homodimers, which facilitate cell to cell adhesions.
