    • Introduction

CD5 is a suitable immunohistochemical marker for T-cells, though not as sensitive as CD3. Approximately 76% of T-cell neoplasms are reported to express CD5, and it is also observed in chronic lymphocytic leukemia, hairy cell leukemia, and mantle cell lymphoma cells. CD5 is commonly lost in cutaneous T-cell lymphoma, and its deficiency can be used as an indicator of malignancy in this condition. CD5’s absence in T cell acute lymphoblastic leukemia, though relatively rare, is linked with a poor prognosis. CD5 serves as a receptor in regulating T-cell proliferation.

    • Synonyms

      CD5 Molecule, CD5, LEU1, CD5 Antigen (P56-62), Lymphocyte Antigen T1/Leu-1, T1, T-Cell Surface Glycoprotein CD5, CD5 Antigen.

    • Physical Appearance

      Sterile Filtered colorless solution.

    • Stability

Store at 4°C if entire vial will be used within 2-4 weeks. Store, frozen at -20°C for longer periods of time. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).Avoid multiple freeze-thaw cycles.

    • Introduction

The T1/Leu1/CD5 molecule, a human T-cell surface glycoprotein of relative molecular mass (Mr) 67,000, has been implicated in the proliferative response of activated T cells and in T-cell helper function. Ly1 is the murine homolog. (In the mouse, lymphocyte surface antigens that are markers for function have been identified. Allelic forms are known and chromosomal assignment of the loci has been achieved. Ly1 is on mouse chromosome 19. Ly2 and Ly3 are on mouse chromosome 6. Ly5 is on mouse chromosome 1. Ly6 is on mouse chromosome 15.

    • Synonyms

      T-cell surface glycoprotein CD5, Lymphocyte antigen T1/Leu-1, CD5, T1, LEU1.

Decay-Accelerating Factor Isoform 1 or CD55, is a protein that attached to the cell membrane through glycophosphatidylinositol (GPI) anchor. CD55 regulates the complement system in the outer membrane. The protein is a receptor to many types of enteroviruses such as coxsackieviruses. CD55 is widely scattered amidst hematopoietic & non-hematopoietic cells. CD52 is a small GPI (glycosylphosphatidylinositol) anchored glycoprotein which is tied to the cell surface with a GPI linkage. CD52 induces immune suppression by complement-meditated cell lysis. Moreover, CD52 is expressed on lymphocytes, eosinophils, monocytes-derived dendritic cells, monocytes, as well as neutrophils, however not on the stem cells from which these lymphocytes were derived. • Introduction

Cell division protein kinase 5 (CDK5) belongs to the cyclin-dependent kinase family. CDK5 is essential for appropriate development of the brain and in order to be activated CDK5 must link to CDK5R1 or CDK5R2. CDK5 doesn't need phosphorylation on the T loop so that binding with the activator is enough to activate the kinase. CDK5 is engaged in the processes of neuronal maturation and migration, phosphorylating the central intracellular adaptor of the reeling signaling chain.

    • Introduction

Cadherin-5 (CDH5) belongs to the atypical/type 2 subgroup of Cadherin homophilic adhesion proteins. Cadherin-5 is a calcium-dependent cell-cell adhesion molecule, which is comprised of 5 extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. Cadherin-5 protein plays a role in the formation, maturation and remodeling of the vascular wall. CDH5 is generally considered to be specific for vascular endothelia in which it is either the sole or the predominant cadherin, often co-existing with N-cadherin. Moreover, the CDH5 protein regulates or is regulated by VEGF R2, type 1 and type 2 TGF-beta receptors, and other endothelial junction proteins such as JAM-C, Claudin-5, and N-Cadherin. CDH5 also connects with alpha-catenin forming a link to the cytoskeleton. CDH5 functions jointly with KRIT1 to create and maintain correct endothelial cell polarity and vascular lumen. Furthermore, Cadherin-5 is essential for activation of PRKCZ and for the localization of phosphorylated PRKCZ, PARD3, TIAM1 and RAP1B to the cell junction. The CDH5 gene is located in a gene cluster in a region on the long arm of chromosome 16 which is involved in loss of heterozygosity events in breast and prostate cancer.

    • Introduction

CD59 is a cell surface glycoprotein which regulates complement-mediated cell lysis, and it is involved in lymphocyte signal transduction. CD59 is an effective inhibitor of the complement membrane attack complex, whereby it binds complement C8 and/or C9 during the compilation of this complex, thus inhibiting the incorporation of multiple copies of C9 into the complex, which is essential for osmolytic pore formation. In addition, CD59 has a role in signal transduction pathways in the activation of T cells. CD59 gene mutations cause CD59 deficiency, a disease resulting in hemolytic anemia and thrombosis, and which causes cerebral infarction.

PDCD5 is expressed in tumor cells during apoptosis independent of the apoptosis-inducing stimuli. Prior to apoptosis induction, PDCD5 is distributed in both the nucleus and cytoplasm. Once apoptosis is induced, the amount of PDCD5 increases and by relocation from the cytoplasm, it accumulates in the nucleus.PDCD5 protein has a stable helical core conformation consisting of a triple-helix bundle and two dissociated terminal regions. PDCD5 is an important novel protein that regulates both apoptotic and non-apoptotic programmed cell death. PDCD5 functions in the process of apoptosis.
PDCD5 plays an important role in regulation of apoptotic processes in gastric cancer cells and gastric tumors.
PDCD5 plays a role in the pathogenesis of rheumatoid arthritis.
-27G/-11A SNP is associated with reduced PDCD5 promoter activity and increased susceptibility to chronic myelogenous leukemia.
PDCD5 gene may be a target gene under the control of some important apoptosis-related transcriptional factors during the cell apoptosis.

    • Introduction

CD33 is putative adhesion molecule of myelomonocytic-derived cells which mediates sialic-acid dependent binding to cells. CD33 prefers to bind to alpha-2,6-linked sialic acid. The sialic acid recognition site is masked by cis interactions with sialic acids on the same cell surface. In the immune response, CD33 perform as an inhibitory receptor upon ligand induced tyrosine phosphorylation by recruiting cytoplasmic phosphatases through their SH2 domains which block signal transduction through dephosphorylation of signaling molecules. In addition, CD33 induces apoptosis in acute myeloid leukemia.
