THAP domain-containing protein 1 (THAP1) is a 213 amino acid protein which is restricted to the nucleoplasm and contains one THAPtype zinc finger, a conserved DNA-binding domain. THAP1 is a DNA-binding transcription regulator which regulates endothelial cell proliferation and G1/S cell-cycle progression. In addition, THAP1 has a pro-apoptopic activity by potentiating both serum-withdrawal and TNF-induced apoptosis. THAP1 protein colocalizes with the apoptosis response protein PAWR/PAR-4 in promyelocytic leukemia (PML) nuclear bodies, and serves as a proapoptotic factor which connects PAWR to PML nuclear bodies.

PBLD is member of the phenazine biosynthesis-like protein (PhzF) family. PBLD which is expressed in most tissues is the only representative of the PhzF family in the human genome. PBLD participates in the MAPK signaling pathway. PBLD is involved in multiple basic cellular functions, its expression is elevated in several disease processes, including folate deficiency and hypotension.

MDS1 is a transcriptional regulator and oncoprotein which takes part in hematopoiesis, development, cell differentiation, apoptosis and proliferation. MDS1 is able to interact with CTBP1, SMAD3, CREBBP, KAT2B, MAPK8, and MAPK9. Overexpression, as a result of translocation with the AML1 gene, is known to cause leukemia. Dynein light chain roadblock-type 2 (DYNLRB2) functions as one of several non-catalytic accessory components of the cytoplasmic dynein complex which are considered to be involved in linking dynein to cargos and to adapter proteins that regulate dynein function. Cytoplasmic dynein functions as a motor for the intracellular retrograde motility of vesicles and organelles along microtubules.

Myelin Protein Zero-Like 1 (MPZL1) is a cell surface receptor, which is involved in signal transduction processes. MPZL1 recruits PTPN11/SHP-2 to the cell membrane and is a putative substrate of PTPN11/SHP-2. MPZL1 is a main receptor for concanavalin-A (ConA) and it is involved in cellular signaling induced by ConA, which most likely includes Src family tyrosine-protein kinases. MPZL1 Isoform 3 appears to have a dominant negative role by blocking tyrosine phosphorylation of MPZL1 induced by ConA. Isoform 1, but not isoform 2 and isoform 3, might be involved in regulation of integrin-mediated cell motility.

LIN28 plays an important role as a 'translational enhancer', leading specific mRNAs to polysomes and therefore increasing the competence of protein synthesis. LIN28 is a marker of undifferentiated human embryonic stem cells and it enhances the efficiency of the formation of induced pluripotent stem (iPS) cells from human fibroblasts. LIN28 binds to the let-7 pre-miRNA and blocks production of the mature let-7 microRNA in mouse embryonic stem cells. Overexpression of LIN28 is associated with human germ-cell tumors.

MAX Dimerization Protein 3 (MXD3) belongs to the Myc superfamily of basic helix-loop-helix leucine zipper transcriptional regulators. MXD3 forms a heterodimer with the cofactor MAX that connects specific E-box DNA motifs in the promoters of target genes and regulates their transcription. Disturbance of the MAX-MXD3 complex is correlated with uncontrolled cell proliferation and tumorigenesis. Transcript variants of MXD3 encoding different isoforms have been described.

Human and mouse CCL28 RNA expression was found to be highest in normal and pathologic colon with the protein being expressed by epithelial cells. Human CCL28 RNA was also present in normal and asthmatic lung tissues.

Human proteins MECP2, MBD1, MBD2, MBD3, and MBD4 form a family of nuclear proteins linked by the existence in each of a methyl-CpG binding domain (MBD). Each one of these proteins, with the exception of MBD3, can bind specifically to methylated DNA. In addition, MECP2, MBD1 and MBD2 can inhibit transcription from methylated gene promoters. Unlike other MBD family members, MECP2 is X-linked and subject to X inactivation. MECP2 is expendable in stem cells, but is vital for embryonic development. Von Hippel-Lindau disease is a dominant inherited syndrome characterized by the predisposition to develop various kinds of benign and malignant tumors, including clear cell renal carcinomas, pheochromocytomas and hemangioblastomas of the central nervous system and retina. VHL syndrome is caused by germline mutation in the VHL tumor suppressor, and VHL tumors are associated with loss or mutation of the remaining wild-type allele. VHL has two domains: a roughly 100-residue NH2-terminal domain rich in b sheet (b-domain) and a smaller a-helical domain (a-domain), held together by two linkers and a polar interface. VHL protein is also involved in the degradation of hypoxia-inducible factor (HIF).

It has been associated as the major inductive signal in patterning of the ventral neural tube, the anterior-posterior limb axis, and the ventral somites. Sonic HedgeHog binds to the patched receptor, which functions in association with smoothened, to activate the transcription of target genes. In the absence of sonic HedgeHog, patched receptor represses the constitutive signaling activity of smoothened. Sonic HedgeHog also regulates another factor, the gli oncogene. Sonic HedgeHog intercellular signal is essential for a various patterning events during development: signal produced by the notochord that induces ventral cell fate in the neural tube and somites, and the polarizing signal for patterning of the anterior-posterior axis of the developing limb bud. Ataxin 3 is otherwise known as Machado-Joseph disease protein 1. Machado–Joseph disease is a hereditary autosomal dominant neurodegenerative disorder. ATXN3 contains trinucleotide CAG repeats in the coding region, and the expansion of these repeats from the normal 13-36 to 68-79 causes the Machado-Joseph disease. ATXN3 is a poly-ubiquitin-binding protein whose cellular turnover is regulated by its catalytic activity.
In addition, ATXN3 is a proteasome-associated factor which mediates the degradation of ubiquitinated proteins. ATXN3 folds reversibly using a single intermediate; partial destabilization of ATXN3 by chemical denaturation causes the formation of fibrillar aggregates by the non-pathological variant.
Ataxin-3 interacts with the major histone acetyltransferases cAMP-response-element binding protein (CREB)-binding protein, p300, and p300/CREB-binding protein-associated factor and hinders transcription by these coactivators.
