Introduction
TNFR1 belongs to the TNF-receptor superfamily. TNFR1 is a receptor for TNFSF2/TNF-alpha and homotrimeric TNFSF1/lymphotoxin-alpha.
There are 2 types of soluble TNF receptors: sTNFR-I and sTNFR-II, which act to neutralize the biological activities of TNF alpha and TNF beta. The levels of these soluble receptors seem to increase as a result of shedding of the extracellular domains of the membrane bound receptors. TNF-a, TNFR1 and TNFR2 have roles in cellular differentiation. TNFR1 and TNFR2 function in cell type-specific renal injury.
TNFR1 is capable of signaling both cell survival and apoptosis. TNFR1-induced apoptosis requires 2 sequential signaling complexes. TNFR1 is capable of activating NF-kappaB, mediate apoptosis, and function as a regulator of inflammation. Oxidative stress promotes TNFR1 and TNFR2 self-interaction, ligand-independent and enhanced ligand-dependent TNF signaling. TNFR1 contributes to the induction of non-cytocidal TNF effects including anti-viral state and activation of the acid sphingomyelinase. Human TNFR1 has a major region which controls cell surface expression. High levels of soluble TNF receptors are found in the amniotic fluid of pregnant women.
Germline mutations of the extracellular domains of TNFR1 are linked to the autosomal dominant periodic fever syndrome. The impaired receptor clearance is believed to be a mechanism of the disease. Familial hibernian fever (FHF) is caused by defects in TNFRSF1A gene. -

Introduction
Regulated upon Activation, Normal T-cell Expressed, and Secreted or RANTES is an 8 kDa protein classified as a chemotactic cytokine or chemokine. It has recently been renamed CCL5. RANTES is chemotactic for T cells, eosinophils and basophils and plays an active role in recruiting leukocytes into inflammatory sites. With the help of particular cytokines (i.e. IL-2 and IFN-?) that are released by T cells, RANTES also induces the proliferation and activation of certain natural killer (NK) cells to form CHAK (CC-Chemokine-activated killer) cells. It is also a HIV-suppressive factor released from CD8+ T cells. This chemokine has been localized to chromosome 17 in humans.

Introduction
RARRES2 is a secreted chemotactic protein that initiates chemotaxis through the ChemR23 G protein-coupled seven-transmembrane domain ligand. RARRES2 is upregulated by the synthetic retinoid tazarotene and found in a vast variety of tissues. RARRES2 acts as an adipokine, and is truncated on both termini from the proprotein. RARRES2 is structurally related to the cathelicidin precursors, cystatin C and kininogens. RARRES2 promotes calcium mobilization and chemotaxis of immature dendritic cells and macrophages. RARRES2 is secreted as a precursor of little biological activity, which requires proteolytic cleavage of its COOH-terminal domain to be exchangeed into a potent and highly specific agonist of ChemR23. RARRES2 signals via its receptor, ChemR23 (CMKLR1), as a positive regulator of adipocyte differentiation and metabolic function. The Chemerin receptor acts as a coreceptor for SIV and some primary HIV-1 strains. The Chemerin receptor has another ligand, called tazarotene-induced gene.

The RAN complex with BIRC5 participates in mitotic spindle formation by being as a physical scaffold to assist the delivery of the RAN effector molecule TPX2 to microtubules. RUN increases AR-mediated transactivation.

Introduction
The gene for TNFRSF12A was initially recognized as a fibroblast growth factor inducible immediate early response gene Fn14 in mouse NIH 3T3 fibroblasts. Human TNFRSF12A cDNA encodes a 129 amino acid residue type I transmembrane protein with a 27 aa signal peptide, a 53 aa extracellular domain, a 21 aa transmembrane domain and a 28 aa cytoplasmic domain. Human and mouse TNFRSF12A hold 82% aa sequence identity. TNFRSF12 is the tiniest member of the TNF receptor superfamily and has only one cysteine rich region in its extracellular domain. The TNFRSF12A cytoplasmic domain holds one TRAF binding motif which binds TRAFs 1, 2, and 3. TNFRSF12A binds its ligand TWEAK/TNFSF12A with high affinity to initiate a signal transduction cascade which subject to the cell type, causes different cellular responses such as cell death, cell proliferation, and angiogenesis.

Background
Severe acute respiratory syndrome coronavirus (SARS-CoV) and Middle East respiratory syndrome coronavirus (MERS-CoV) have caused significant outbreaks with severe respiratory illness in recent years. The spike proteins of these viruses play a crucial role in viral entry and host cell recognition. This research aims to explore the structure, function, and implications of SARS and MERS spike proteins in viral pathogenesis and the development of therapeutic interventions. Understanding the mechanisms of viral entry can provide valuable insights into the design of effective antiviral strategies.
Structure and Function of SARS and MERS Spike Proteins:
The spike proteins of SARS-CoV and MERS-CoV are trimeric glycoproteins located on the viral surface. They consist of S1 and S2 subunits, with the S1 subunit responsible for receptor binding and the S2 subunit involved in membrane fusion. The spike proteins contain key domains, including the receptor-binding domain (RBD) and the fusion peptide, which mediate the interactions with host cell receptors and membrane fusion during viral entry.
Interaction with Host Cell Receptors:
The SARS-CoV spike protein primarily interacts with the angiotensin-converting enzyme 2 (ACE2) receptor, which is expressed in various tissues, including the respiratory tract. The binding of the SARS-CoV spike protein to ACE2 is crucial for viral entry into host cells. Similarly, the MERS-CoV spike protein interacts with the dipeptidyl peptidase 4 (DPP4) receptor, which is predominantly expressed in the lungs and other tissues. These receptor interactions determine the host range and tissue tropism of the viruses.
Implications for Viral Pathogenesis:
The interaction between the spike proteins of SARS-CoV and MERS-CoV and their respective receptors initiates a cascade of events leading to viral entry and replication. The binding of the spike protein to the receptor triggers conformational changes that expose the fusion peptide, facilitating membrane fusion and viral entry into host cells. The specificity and affinity of the spike-receptor interaction play a critical role in determining viral tropism, disease severity, and transmission dynamics.
Therapeutic Strategies Targeting Spike Proteins:
The spike proteins of SARS-CoV and MERS-CoV present attractive targets for the development of antiviral interventions. Various approaches have been explored, including the development of neutralizing antibodies that specifically target the spike proteins and inhibit viral entry. Additionally, small molecule inhibitors and peptide-based therapeutics are being investigated to disrupt the spike-receptor interaction and block viral entry. Vaccine development efforts have also focused on generating immune responses against the spike proteins to prevent infection.
Challenges and Future Directions:
Although significant progress has been made in understanding the structure and function of SARS and MERS spike proteins, several challenges remain. The emergence of novel coronavirus variants necessitates continuous surveillance and adaptation of therapeutic strategies. Additionally, potential cross-reactivity and immunogenicity concerns need to be addressed to ensure the safety and efficacy of spike protein-based therapeutics.
Conclusion:
The study of SARS and MERS spike proteins provides valuable insights into the mechanisms of viral entry and pathogenesis. Understanding the interactions between spike proteins and host cell receptors is crucial for the development of effective antiviral strategies. Further research on spike protein structure, receptor binding, and fusion mechanisms is essential to combat current and future coronavirus outbreaks. By unraveling the complexities of viral entry, we can pave the way for the development of novel therapeutics to mitigate the impact of SARS-CoV, MERS-CoV, and related coronaviruses.

Introduction
Since April 2012, cases of the Middle East Respiratory Syndrome Coronavirus (MERS-CoV) have been identified in various countries. Coronaviruses are the cause of the common cold, SARS (severe acute respiratory syndrome) and other severe illnesses with high mortality rates, all are classified into coronavirus family. MERS-CoV is a new type of SARS found in the coronavirus family causing severe pneumonia with sudden and serious respiratory illness with high mortality rates as well. Since January 27th 2015, the WHO has reported 956 human cases, including 351 deaths. More cases of the new coronavirus strain are expected. Like in other coronaviruses, large surface spike glycoprotein is a central structural protein of this virus; it is located above the virion surface to bind and enter into the target cell. Spike protein has 2 domains- S1 and S2. The S1 domain is responsible for cellular tropism and interaction with target cell, while the S2 domain is responsible for membrane fusion. The C-terminal of S1 domain contains a receptor binding domain, and is also a potential target for vaccine development and an antigen for diagnosis.

Introduction
The Fas receptor (CD95) mediates apoptotic signaling by Fas-ligand expressed on the surface of other cells. The Fas-FasL interaction plays an important role in the immune system and lack of this system leads to autoimmunity, indicating that Fas-mediated apoptosis removes self-reactive lymphocytes. Fas signaling is also involved in immune surveillance to remove transformed cells and virus infected cells. Binding of FAS to oligimerized FasL on another cell activates apoptotic signaling through a cytoplasmic domain termed the death domain that interacts with signaling adaptors including FAF, FADD and DAX to activate the caspase proteolytic cascade. Caspase-8 and caspase-10 are first activated, to then cleave and activate downstream caspases, and a variety of cellular substrates that lead to cell death. Caspases cleave nuclear lamins, causing the nucleus to break down and lose its normal structure and another caspase substrate is DFF, inducing cleavage and degradation of the genome. Other caspase substrates are involved in cytoskeletal structure, cell cycle regulation and signaling pathways. Activation of JNK kinase, activation of Jun, and production of ceramide may also play roles in Fas-mediated apoptosis. Activation of fas-mediated apoptosis is opposed by I-FLICE and FAP. Viruses and tumors may escape immune surveillance in part through suppression of fas-mediated apoptosis using similar mechanisms.

Introduction
Cleavage stimulation factor subunit 1 (CSTF1) is involved in the polyadenylation and 3'end cleavage of pre-mRNAs. The CSTF1 gene encodes one of three subunits which merge to produce cleavage stimulation factor (CSTF). Like the mammalian G protein beta subunits, CSTF1 contains transducin-like repeats. CSTF1 is one of the numerous factors necessary for polyadenylation and 3'-end cleavage of mammalian pre-mRNAs. CSTF1 is responsible for the interaction of CSTF with other factors to create a stable complex on the pre-mRNA.
