      Leukemia Inhibitory Factor also called LIF is a lymphoid factor that promotes long-term maintenance of embryonic stem cells by suppressing spontaneous differentiation. Leukemia Inhibitory Factor has several functions such as cholinergic neuron differentiation, control of stem cell pluripotency, bone & fat metabolism, mitogenesis of factor dependent cell lines & promotion of megakaryocyte production in vivo. Human and mouse LIF exhibit a 78% identity in its amino acid sequence.

    • Introduction

      Chemokine (C-X-C motif) ligand 11 (CXCL11) is a small cytokine belonging to the CXC chemokinen family. I-TAC is highly expressed in peripheral blood leukocytes, pancreas and liver, with moderate levels in thymus, spleen and lung and low expression levels were in small intestine, placenta and prostate. Gene expression of CXCL11 is strongly induced by IFN-g and IFN-b, and weakly induced by IFN-a. The I-TACchemokine elicits its effects on its target cells by interacting with the cell surface chemokine receptor CXCR3, with a higher affinity than do the other ligands for this receptor, CXCL9 and CXCL10.       100mM KCl, 50mM Tris-HCl, pH-8.5, 10mM MgCl2, 0.1mM EDTA, 1mM DTT and 2% Glycerol.

    • Preparation protocol

      1. After first round PCR, purify PCR fragments using Qiagen QIAquick PCR purification kit with elution in dH2O.
      2. Dilute PCR product to 250 ng/µl in 10 mM Tris–HCl, pH 7.8, 50 mM NaCl and heat to 95 °C for 5 min followed by cooling at 0.1°C /s to 25 °C.
      3. Add reaction buffer (100mM KCl, 50 mM Tris-HCl, pH 8.5, 10 mM MgCl2, 0.1 mM EDTA, 1 mM DTT, 2% Glycerol) to annealed PCR product, adjust DNA concentration to 11.5 ng/µl, add 6×His-MutS dimers to 950 nM.
      4. Incubate the mixture at room temperature for 10 min, then add an equal volume of Ni-NTA beads pre-equilibrated with 1×binding buffer, and incubate for 30 min at room temperature. 5. Gently spin down beads and save supernatant for subsequent processing (second round PCR, cloning etc).

      GLUL catalyzes the synthesis of glutamine from glutamate and ammonia. Glutamine is a major source of energy and that takes part in cell proliferation, inhibition of apoptosis, and cell signaling.       SIRT5 belongs to the Sirtuin family of proteins, homologs to the yeast Sir2 protein. Sirtuin family members hold a sirtuin core domain and grouped into four classes. The protein encoded by this gene is included in class III of the sirtuin family. Human sirtuins functions were not yet determined; still, yeast sirtuin proteins are known to regulate epigenetic gene silencing and suppress recombination of rDNA.       Hypoxia-inducible factor-1 (HIF-1), identified as one of the transcription factors, has been found to play an essential role in cellular and systemic oxygen homeostasis. HIF-1 is a heterodimer composed of HIF-1b subunit and one of three subunits (Hif-1a, Hif-2 (or Hif-3)). The activation of Hif-1 (is closely associated with a variety of tumors and oncogenic pathways. Hif-1 (consists of DNA binding domain (DBD domain), Dimerization domain and C-terminal regulatory domains, including two transactivation domains (TAD), an oxygen-dependent degradation (ODD) domain, and inhibitory domains. Under hypoxic conditions HIF1A activates the transcription of more than 40 genes, including, erythropoietin, glucose transporters, glycolytic enzymes, VEGF, and other genes whose protein products increase oxygen delivery or facilitate metabolic adaptation to hypoxia.
