Small nuclear ribonucleoprotein complexes (abbreviated as U-snRNP) are essential for splicing of precursor mRNA molecules. U1-snRNP is the most abundant RNP particle in the nucleus and consists of one small uridylate-rich RNA (U1 RNA) complexed with several proteins: the three 68/70 kDa, A, C polypeptides are unique to the U1-snRNP particle, whereas 7 so-called Sm proteins (B/B', D1, D2, D3, E, F, G) form a core subparticle that is common to all U-snRNP complexes.
Both the U1-specific proteins and the Sm core particle are targets of autoantibodies which classically have been called the RNP and RNP-Sm specificities, respectively. A clean diagnostic distinction of these specificities has been complicated by the biochemical difficulties of producing clean subparticle fractions from native sources. The use of single recombinant proteins as antigenic targets guarantees a much higher sensitivity and specificity and is the only way to determine RNP antibodies sensu stricto without the disturbing influence of Sm antigens; also with single U1 proteins antibodies will be detected which can be missed because of steric hindrance when using the RNP-Sm complex in an assay.
Autoantibodies to U1-snRNP are present in 95% of patients with Mixed Connective Tissue Disease (MCTD) and 30% of patients with SLE. Antibodies against the 68/70 kDa protein are known to have a high clinical significance in MCTD patients. The 68/70 kDa nomenclature of this protein refers to the fact that different splice variants of the protein are found in human cells.

PET117 is nuclear gene essential for the assembly of active cytochrome c oxidase in S. cerevisiae. Nevertheless the gene products are not subunits of the final assembled cytochrome c oxidase complex. PET117 is found in chromosome V proximate the HIS1 gene.

Prefoldin subunit 2 (PFDN2) belongs to the prefoldin beta subunit family. The PFDN2 protein is one of 6 subunits of prefoldin, which is a molecular chaperone complex that binds and stabilizes newly synthesized polypeptides, thus allowing them to fold correctly. PFDN2 binds specifically to cytosolic chaperonin (c-CPN) and transfers target proteins to it. Vitronectin (VTN) which is a part of the pexin family is a cell adhesion and spreading factor found in serum and tissues. VTN interacts with glycosaminoglycans and proteoglycans. PTEN, a tumor suppressor, has been implicated in a large number of human tumors and is conserved from humans to worms. PTEN has a tensin like domain and a catalytic domain similar to that of the dual specificity protein tyrosine phosphatases. Characterization of PTEN protein showed that it is a phosphatase that acts on proteins and on 3-phosphorylated phosphoinositides, and can therefore modulate signal transduction pathways that involve lipid second messengers. In contrast to most of the protein tyrosine phosphatases, PTEN preferentially dephosphorylates phosphoinositide substrates. PTEN negatively regulates intracellular levels of phosphatidylinositol-3,4,5-trisphosphate in cells and acts as a tumor suppressor by negative regulation of AKT/PKB signaling pathway. Ribonuclease P protein subunit p30 (RPP30) is a member of the eukaryotic/archaeal RNase P protein component 3 family. RPP30 is component of ribonuclease P, which is a protein complex that generates mature tRNA molecules by cleaving their 5'-ends. Ribonuclease P (RNase P) is small nuclear ribonucleoprotein (snRNPs) which acts on RNA substrates in vitro. Streptavidin is a tetrameric protein secreted by Streptomyces avidinii which binds firmly to biotin. Streptavidin is widely used in molecular biology through its unique high affinity for the vitamin biotin. The dissociation constant (Kd) of the biotin-streptavidin complex is about ~10-15 mol/L. The strong affinity recognition of biotin and biotinylated molecules has made streptavidin one of the most important components in diagnostics and laboratory kits. S100A9 is part of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100A9 protein is localized in the cytoplasm and/or nucleus of a wide range of cells, and participates in the regulation of several cellular processes such as cell cycle progression and differentiation. S100 genes include no less than 13 proteins which are localized as a cluster on chromosome 1q21. Dickkopf-related protein 3 (DKK3) belongs to the DKK protein family including Dkk-1, 2, 3 and -4. DKK3 is a 350 amino acid secreted glycoprotein which is comprised of an N-terminal signal peptide and 2 conserved cysteine-rich domains that are separated by a 12 amino acid linker region. DKK3 is involved in embryonic development through its inhibition of the WNT signaling pathway. DKK3 gene expression is decreased in a variety of cancer cell lines and it may act as a tumor suppressor gene.

GTPase, IMAP Family Member 5 (GIMAP5) is a part of the GTP-binding superfamily and the immuno-associated nucleotide subfamily of nucleotide-binding proteins. PABPN1 is an highly expressed nuclear protein which has high affinity to nascent poly(A) tails. BTLA, also known as B & T Lymphocyte Associated, is an inhibitory molecule which is part of the Ig superfamily. BTLA is a type 1 transmembrane glycoprotein in the CD28 family of T cell costimulatory molecules. BTLA is a 3rd inhibitory receptor on T lymphocytes with resemblances to CTLA-4 & PD-1. Moreover, BTLA is a ligand for TNF (receptor) superfamily, TNFRSF14, and HVEM. BTLA-HVEM complexes negatively regulate T-cell immune responses.

The hnRNP proteins have distinctive nucleic acid binding properties. HNRNPA1 has 2 repeats of quasi-RRM domains that bind to RNAs. HNRNPA1 is one of the most copious core proteins of hnRNP complexes and it is restricted to the nucleoplasm. The HNRNPA1 protein, along with other hnRNP proteins, is exported from the nucleus, most likely bound to mRNA, and is immediately re-imported. The M9 domain of HNRNPA1 functions as both a nuclear localization and nuclear export signal.
