    • Introduction

      HIV-1 and HIV-2 appear to package their RNA differently. HIV-1 binds to any appropriate RNA whereas HIV-2 preferentially binds to mRNA which creates the Gag protein itself. This means that HIV-1 is better able to mutate. HIV-2 is transmitted in the same ways as HIV-1: Through exposure to bodily fluids such as blood, semen, tears and vaginal fluids.
      Immunodeficiency develops more slowly with HIV-2.
      HIV-2 is less infectious in the early stages of the virus than with HIV-1.
      The infectiousness of HIV-2 increases as the virus progresses.
      Major differences include reduced pathogenicity of HIV-2 relative to HIV-1, enhanced immune control of HIV-2 infection and often some degree of CD4-independence. Despite considerable sequence and phenotypic differences between HIV-1 and 2 envelopes, structurally they are quite similar. Both membrane-anchored proteins eventually form the 6-helix bundles from the N-terminal and C-terminal regions of the ectodomain, which is common to many viral and cellular fusion proteins and which seems to drive fusion.
      HIV-1 gp41 helical regions can form more stable 6-helix bundles than
      HIV-2 gp41 helical regions however HIV-2 fusion occurs at a lower threshold temperature (25°C), does not require Ca2+ in the medium, is insensitive to treatment of target cells with cytochalasin B, and is not affected by target membrane glycosphingolipid composition.

      PSG5 belongs to the PSG family, which are a highly connected group of secreted glycoproteins, vastly expressed in fetal placental syncytiotrophoblast cells. PSGs are can be found in serum from the first 2-3 weeks of pregnancy and at higher levels as the pregnancy progresses, up to e point where they are the highest fetal protein found in maternal blood at term. PSG5’s role is inducing secretion of TH2-type cytokines from monocytes and modulating the maternal immune system throughout the pregnancy, thus defending the semi-allotypic fetus from rejection.

      Chromgranin-A is part of the neuroendocrine secretory protein family. CHGA is located in secretory vesicles of neurons and endocrine cells. Chromgranin-A is a precursor to three biologically active peptides; vasostatin, pancreastatin, and parastatin. These peptides act as autocrine or paracrine negative modulators of the neuroendocrine system. Other peptides, including chromostatin, beta-granin, WE-14 and GE-25, are also derived from the full-length protein. Chromgranin-A has numerous biological activities on some tissues and organs and exerts a large spectrum of homeostatic actions, including antifungal and antimicrobial effect, modulation of cell adhesion, and inhibition of parathyroid hormone secretion.

The DENV core protein is a vital component of the viral particle, playing a crucial role in viral assembly and genome packaging. Understanding the structural and functional characteristics of the DENV-2 core protein is essential for unraveling the viral assembly process and identifying potential targets for antiviral intervention.
      This research aims to characterize the DENV-2 core protein in its recombinant form to gain insights into its role in viral replication and explore potential strategies for combating dengue fever.
      The primary objective of this study is to express and purify recombinant DENV-2 core protein using various expression systems. Recombinant DNA techniques will be employed to construct expression vectors containing the DENV-2 core gene, followed by expression in bacterial, yeast, or mammalian cell-based systems. The recombinant core protein will be purified using affinity chromatography or other suitable methods, enabling subsequent biochemical and biophysical characterization.
      The second objective is to investigate the assembly and structural properties of the DENV-2 core protein. The FCGR3A receptor is expressed on natural killer (NK) cells as an integral membrane glycoprotein anchored through a transmembrane peptide, while FCGR3B is expressed on polymorphonuclear neutrophils (PMN) where the receptor is anchored through a phosphatidylinositol (PI) linkage. In addition, FCGR3A is expressed on macrophages, subpopulation of T-cells, immature thymocytes and placental trophoblasts. FCGR3A mediates antibody-dependent cellular cytotoxicity (ADCC) and other antibody-dependent responses, such as phagocytosis. FCGR3A gene mutations are linked with susceptibility to recurrent viral infections, susceptibility to systemic lupus erythematosus, and alloimmune neonatal neutropenia.

    • Introduction

      MAGEA6 belongs to the MAGEA family, which directs the expression of tumor antigens which are seen on a human melanoma by autologous cytolytic T lymphocytes (CTL). Members of the MAGEA family encode proteins with 50 to 80% sequence identity to each other. The MAGE genes, which are clustered at chromosomal location Xq28, were originally isolated from separate kinds of tumors, and based on their practically exclusive tumor-specific expression in adult tissues; they are objects for cancer immunotherapy. MAGEA genes’ promoters and first exons exhibit substantial variability, implying that the existence of the MAGEA gene family enables the same function to be expressed under different transcriptional controls. MAGEA genes are implicated in several hereditary disorders, such as dyskeratosis congenita.

    • Introduction

      Forty-two antigenic domains were identified across the hepatitis A virus (HAV) polyprotein by using a set of 237 overlapping 20-mer synthetic peptides spanning the entire HAV polyprotein. Nineteen antigenic domains were found within the structural proteins, and 22 were found within the nonstructural proteins, with 1 domain spanning the junction of VP1 and P2A proteins. Five of these domains were considered immunodominant, as judged by both the breadth and the strength of their immunoreactivity. One domain is located within the VP2 protein at position 57-90 aa. A second domain, located at position 767-842 aa, contains the C-terminal part of the VP1 protein and the entire P2A protein. A third domain, located at position 1403-1456 a.a, comprises the C-terminal part of the P2C protein and the N-terminal half of the P3A protein. The fourth domain, located at position 1500-1519 a.a, includes almost the entire P3B, and the last domain, located at position 1719-1764 aa, contains the C-terminal region of the P3C protein and the N-terminal region of the P3D protein. Four of the five most immunoreactive domains are derived from small HAV proteins and/or encompass protein cleavage sites separating different HAV proteins.

      HAV VP1-P2A protein although stable at 4°C for 1 week, should be stored below -18°C. Please prevent freeze thaw cycles.

    • Applications

      HAV VP1-P2A antigen is suitable for ELISA and Western blots, excellent antigen for detection of HAV with minimal specificity problems.
