      Mitogen-Activated Protein Kinase Kinase 2 (MAP2K2) is a dual specificity protein kinase, which is a member of the STE grouping of kinases and the MAP kinase kinase family. MAP2K2 is activated by a wide range of growth factors and cytokines and also by membrane depolarization and calcium influx. The MAP2K2 kinase is known for its vital role in mitogen growth factor signal transduction. MAP2K2 phosphorylates and consequently activates MAPK1/ERK2 and MAPK2/ERK3. The activation of the MAP2K2 kinase itself is dependent on the Ser/Thr phosphorylation by MAP kinase kinase kinases. The inhibition or degradation of the MAP2K2 kinase is also found to be involved in the pathogenesis of Yersinia and anthrax. MAP2K2 gene mutations cause cardiofaciocutaneous syndrome (CFC syndrome).

      SKP1 is a F-box enzyme which functions as a substrate recognition component of the SCF ubiquitin ligase complex which controls the ubiquitination of proteins involved in cell cycle progression, signal transduction and transcription. SKP1 binds to proteins containing an F-box motif, such as cyclin F, S-phase kinase-associated protein 2, and other regulatory proteins involved in ubiquitin dependent proteolysis. SKP1 takes part in the control of beta-catenin levels and the activity of beta-catenin dependent TCF transcription factors. SKP1 serves as an adapter that links the F-box protein to CUL1 in the SCF complex.

      VRK3 is a member of the protein kinase superfamily. The VRK3 gene has replacements at a number of residues within the ATP binding motifs which in other kinases were proved to be required for catalysis in both human and mouse. In vitro assays show that VRK3 has no phosphorylation activity but probably holds its substrate binding capability. VRK3 is universally expressed in human tissues and its protein is restricts to the nucleus.

      PGK2 is a testis-specific form of phosphoglycerate kinase. Originally presumed to be a pseudogene, the PGK2 protein is actually a functional phosphoglycerate kinase which catalyzes the reversible conversion of 1,3-bisphosphoglycerate to 3-phosphoglycerate, during the Embden-Meyerhof-Parnas pathway of glycolysis, in the later stages of spermatogenesis.
