    • Introduction

      Chemokine (C-X-C motif) ligand 2 (CXCL2) is a small cytokine belonging to the CXC chemokine family that is also called macrophage inflammatory protein 2-alpha (MIP2-alpha), Growth-regulated protein beta (Gro-beta) and Gro oncogene-2 (Gro-2). CXCL2 is 90% identical in amino acid sequence as a related chemokine, CXCL1. This chemokine is secreted by monocytes and macrophages and is chemotactic for polymorphonuclear leukocytes and hematopoietic stem cells. The gene for CXCL2 is located on human chromosome 4 in a cluster of other CXC chemokines. CXCL2 mobilizes cells by interacting with a cell surface chemokine receptor called CXCR2.

      Anthrax PA (Protective Antigen) and Anthrax LF (Lethal Factor), along with Anthrax EF (Edema Factor), are toxins produced by Bacillus anthracis. All three of the proteins must be present for toxicity to occur. It begins with the binding of PA to its cell-surface receptor, ATR (Anthrax Toxin Receptor), at which point PA is cleaved and the resulting 63 kD carboxyl-terminal fragment binds to LF and EF. LF is a protease that is known to cleave members of the mitogen-activated protein kinase kinase (MAPKK) family(2).

      Anthrax PA Antibody was developed by immunizing rabbits with synthetic peptides corresponding to amino acids 240-254 of the Anthrax PA protein (~83 kD). Thus the Anthrax PA antibody (made against amino acids 250-250) should recognize both the 83 kDa and 63 kDa PA forms, but not the 20 kDa cleavage fragment.

    • Introduction

      PTX3 is part of the pentraxin family sharing the C-terminal domain with short pentraxins and containing a unique N-terminal domain. PTX3 is produced and released at inflammatory sites by various cell types including monocytes/macrophages, endothelial cells, vascular smooth muscle cells, fibroblasts, and adipocytes. PTX3 is involved in the regulation of innate resistance to pathogens, inflammatory reactions, possibly clearance of self-components and female fertility. PTX3 is used as a marker for disease activity of psoriasis. High serum PTX3 levels are associated with the disease severity of systemic sclerosis. Elevated serum PTX3 is associated with pulmonary fungal infections.

      CD21 is expressed strongly on mature B cells, follicular dentritic cells and weakly on immature thymocytes and T lymphocytes. In B-cell ontogeny, CD21 appears after the pre-B-stage, is maintained during peripheral B-cell development and is lost upon terminal differentiation into plasma cells. CD21 expression is also gradually lost after stimulation of B cells in vitro. CD21 functions as receptor for C3d, C3dg and iC3b Complement components, for EBV and for IFNalpha. CD21 binds to CD23 and associates with CD19, CD81 and Leu13 to form a large signal-transduction complex involved in B cell activation.

      Native human C4 is aglycosylated polypeptide containing 3 disulfide-linked chains. C4 is essential to the activation of both thelectin and the classical pathways of complement activation.Initiation of each pathway generates proteolytic enzyme complexes which are bound to the target surface. These enzymes cleave a peptide bond in C4 releasing the anaphylatoxin C4a and activating C4b. Like C3, the thioester of metastable C4b is highly reactive and is capable of reacting and covalently coupling C4b to amino or hydroxyl groups on the target surface. C4A and C4B are 2 variants of C4 that are common in manwhereas, in animals who have only 1 type usually have C4B.
