    • Introduction

Complement Factor B, also known as CFB, encodes complement factor B which is a component of the alternative pathway of complement activation. Factor B circulates in the blood as a single chain polypeptide. Once the alternative pathway is activated it is cleaved by complement factor D yielding the noncatalytic chain Ba and the catalytic subunit Bb. The active subunit Bb is a serine protease which connects with C3b to form the alternative pathway C3 convertase. Also, Bb is involved in the proliferation of preactivated B lymphocytes, while Ba inhibits their proliferation.

    • Introduction

Chemokine (C-X-C motif) ligand 1 (CXCL1) is a small cytokine belonging to the CXC chemokine family that was previously called GRO1 oncogene, Neutrophil-activating protein 3 (NAP-3) and melanoma growth stimulating activity, alpha (MSGA-a). It is secreted by human melanoma cells, has mitogenic properties and is implicated in melanoma pathogenesis. CXCL1 is expressed by macrophages, neutrophils and epithelial cells, and has neutrophil chemoattractant activity. CXCL1 plays a role in spinal cord development by inhibiting the migration of oligodendrocyte precursors and is involved in the processes of angiogenesis, inflammation, wound healing, and tumorigenesis. This chemokine elicits its effects by signaling through the chemokine receptor CXCR2. The gene for CXCL1 is located on human chromosome 4 amongst genes for other CXC chemokines.

    • Introduction

Cyclosporin A is a noncytotoxic, natural, 11 amino acid cyclic peptide used clinically as an immunosuppressant for the treatment of autoimmune and inflammatory disorders and to prevent organ rejection after transplantation. Cyclosporin acts chiefly by inhibiting T lymphocyte function, which is vital for the propagation of inflammation. Cyclosporin A does not suppress the activity of other hematopoietic cells, does not cause bone marrow suppression and has a rapid onset of action as opposed to other immunosuppressive agents. Nevertheless, Cyclosporin A -induced nephrotoxicity remains an important clinical problem, and oxidative stress has been implicated as a potential responsible mechanism.

    • Background

Transforming Growth Factor Beta Receptor 1 (TGFBR1), a transmembrane protein crucial in the TGF-β signaling pathway, holds a paramount position in regulating diverse cellular processes. Its intricate involvement in development, immune responses, tissue homeostasis, and disease has elevated TGFBR1 to a central role in biology and medicine. This research embarks on a comprehensive exploration of the TGFBR1 protein, unraveling its structural complexities, signaling mechanisms, and its far-reaching implications in various physiological and pathological contexts. By dissecting the intricacies of TGFBR1, scientists aim to decode the fundamental cellular processes it governs and explore potential therapeutic avenues in the domains of cancer, fibrosis, and immunology.

      Structural Complexity of TGFBR1:

TGFBR1 is a serine/threonine kinase receptor with an extracellular ligand-binding domain, a transmembrane domain, and an intracellular kinase domain. Its structure allows it to interact with TGF-β ligands and initiate downstream signaling cascades. Understanding the three-dimensional architecture of TGFBR1 is pivotal for deciphering its interactions with ligands, co-receptors, and intracellular signaling partners, shedding light on the molecular intricacies of its function.

      Signaling Pathways and Physiological Functions:

Upon ligand binding, TGFBR1 phosphorylates downstream effectors, regulating processes like cell proliferation, differentiation, apoptosis, and immune responses. TGF-β signaling mediated by TGFBR1 is vital in embryogenesis, tissue repair, and immune tolerance. Dysregulation of this pathway is implicated in numerous diseases, including cancer, fibrosis, and autoimmune disorders, underscoring the significance of TGFBR1 in maintaining cellular and tissue homeostasis.

      TGFBR1 in Cancer Biology:

TGFBR1's dual role as a tumor suppressor and a promoter of cancer progression reflects its complexity in cancer biology. In early stages, TGFBR1 signaling suppresses cell growth and promotes apoptosis, acting as a defense against tumorigenesis. However, in advanced stages, cancer cells exploit TGFBR1 signaling to facilitate invasion, metastasis, and immune evasion. Understanding the context-dependent nature of TGFBR1's functions in cancer is pivotal for developing targeted therapies.

      Targeting TGFBR1 in Therapeutics:

Given its critical roles in various diseases, TGFBR1 has emerged as an attractive target for therapeutic interventions. In cancer, efforts are underway to develop small molecule inhibitors and monoclonal antibodies that modulate TGFBR1 signaling, aiming to curb tumor progression. Additionally, in fibrotic disorders, targeting TGFBR1 offers hope for halting the pathological tissue remodeling characteristic of these diseases, providing potential treatments for conditions such as pulmonary fibrosis and liver cirrhosis.

TGFBR1 Protein, with its intricate signaling mechanisms and diverse physiological roles, stands at the crossroads of fundamental cellular processes and disease pathogenesis. Its involvement in development, immune regulation, cancer, and tissue homeostasis underscores its significance in biology and medicine. As researchers delve deeper into the complexities of TGFBR1, they pave the way for innovative therapies and a deeper understanding of diseases, ultimately shaping the future of healthcare and scientific exploration. This research not only illuminates the pivotal role of TGFBR1 but also holds the promise of transformative advancements in medicine and our understanding of cellular signaling pathways.

    • Introduction

Alpha crystallins are composed of two gene products; alpha-A and alpha-B, for acidic and basic, respectively. Alpha crystallins can be induced by heat shock and are members of the small heat shock protein (sHSP also known as the HSP20). They act as molecular chaperones and hold them in in large soluble aggregates. These heterogeneous aggregates consist of 30-40 subunits; the alpha-A and alpha-B subunits have a 3:1 ratio, respectively. Two additional function of -crystallins are an autokinase activity and participation in the intracellular architecture. Alpha-B is expressed widely in many tissues and organs and occurs in many neurological diseases.

Streptavidin is a tetrameric protein secreted by Streptomyces avidinii which binds firmly to biotin. Streptavidin is wilyde used in molecular biology through its unique high affinity for the vitamin biotin. The dissociation constant (Kd) of the biotin-streptavidin complex is about ~10-15 mol/L. The strong affinity recognition of biotin and biotinylated molecules has made streptavidin one of the most important components in diagnostics and laboratory kits. The streptavidin/biotin system has one of the biggest free energies of association of yet observed for noncovalent binding of a protein and small ligand in aqueous solution (K_assoc = 10**14). The complexes are also extremely stable over a wide range of temperature and pH.

    • Introduction

Lipopolysaccharides (LPS) are a type of glycolipids on the outer cell wall of Gram-negative bacteria. Lipopolysaccharide binding protein (aka LBP) is a plasma protein which facilitates the diffusion of bacterial LPS (endotoxin). LBP is involved in the acute-phase immunologic response to gram-negative bacterial infections. In cooperation with bactericidal permeability-increasing protein (BPI), LBP binds LPS and interacts with the CD14 receptor, most likely playing a role in regulating LPS-dependent monocyte responses. LBP belongs to a family of structurally and functionally related proteins, including BPI, plasma cholesteryl ester transfer protein (CETP), and phospholipid transfer protein (PLTP). The LBP gene is found on chromosome 20, directly downstream of the BPI gene. LBP catalyzes the transfer of LPS monomers from LPS aggregates to HDL particles, to phospholipid bilayers, and to a binding site on soluble CD14 (sCD14). sCD14 is capable of speeding up the transfer by receiving an LPS monomer from an LPS aggregate, and then yielding it to an HDL particle, therefore acting as a soluble "shuttle" for an insoluble lipid.

    • Introduction

Avidin is a tetrameric protein of 4 identical subunits (homotetramer) each of which can bind to biotin with a high degree of affinity and specificity. Avidin molecular weight in its tetrameric form is estimated to be between 66-69 kDa. Avidin is produced in the oviducts of birds, reptiles and amphibians and is subsequently deposited in the whites of their eggs. In the chicken egg white, avidin makes up roughly 0.05% of total protein (approximately 1.8 mg per egg). 10% of Avidin’s molecular weight is ascribed to carbohydrate content which is composed of four to five mannose and three N-acetylglucosamine residues. Avidin has at least three distinctive oligosaccharide structural type which are similar in structure and composition. The dissociation constant (KD) of avidin is approximately 10-15M, making it one of the strongest known non-covalent bonds.

    • Introduction

Babesiosis is a disease caused by apicomplexan parasites of the Babesia genus. The Babesia microti life cycle involves 2 hosts, which include a rodent, mainly the white-footed mouse (Peromyscus leucopus) and a tick in the Ixodes genus. During a blood meal, a Babesia-infected tick introduces sporozoites into the mouse host. Sporozoites pass into the erythrocytes and undergo asexual reproduction (budding). In the blood, some parasites differentiate into male and female gametes, though these cannot be distinguished by light microscopy. The definitive host is the tick. Once ingested by an proper tick, gametes unite and undergo a sporogonic cycle resulting in sporozoites. Transovarial transmission (aka vertical or hereditary transmission) has been detected for "large" Babesia species but not for the "small" Babesia, such as B. microti. Humans enter the cycle when bitten by the infected ticks. Thus during a blood meal, a Babesia-infected tick introduces sporozoites into the human host. Sporozoites then enter erythrocytes and undergo asexual replication (budding). Multiplication of the blood-stage parasites is responsible for the clinical manifestations of the disease. Humans typically are dead-end hosts. However, human-to-human transmission is well acknowledged to occur via contaminated blood transfusions.

    • Introduction

Yersinia enterocolitica is a Gram-negative bacillus-shaped bacterium, which is a member of the Enterobacteriaceae family. Y.Enterocolitica is motile at temperatures between 22-29°C, however becomes non-motile at normal human body temperature. Y. Enterocolitica infection causes the yersiniosis disease, which is an animal-borne disease occurring in humans, as well as in a various groups of animals such as cattle, deer, pigs, and birds. Yersinia enterocolitica is a heterogeneous group of strains, which are conventionally classified by bio-typing into six bio-groups on the basis of phenotypic characteristics, and by serotyping into more than 57 “O” serogroups, on the basis of their O (lipopolysaccharide or LPS) surface antigen. Five of the six biogroups (1B and 2–5) are considered as pathogens. Nevertheless, only a few of these serogroups have been linked with disease in either humans or animals. Strains which belong to serogroups O:3 (biogroup 4), O:5,27 (biogroups 2 and 3), O:8 (biogroup 1B), and O:9 (biogroup 2) are most frequently isolated worldwide from human samples. Still, the main Y. enterocolitica serogroup in many European countries is serogroup O:3 followed by O:9, whereas the serogroup O:8 is mostly detected in the United States.

    Description

Somatotropin Chicken Antagonist Recombinant mutein G119R produced in E.Coli is a single, non-glycosylated polypeptide chain containing 191 amino acids with an additional Ala at the N-terminus and having a molecular mass of 22.3 kDa. The Chicken Growth-Hormone Antagonist Recombinant is purified by proprietary chromatographic techniques.

    Source

    Escherichia Coli.

    Formulation

The protein was lyophilized from a concentrated (1mg/ml) solution with 0.03% NaHCO3 pH-8.

    Purity

Greater than 98.0% as determined by:
    (a) Analysis by SEC-HPLC.
    (b) Analysis by SDS-PAGE.

    Biological Activity

Recombinant Chicken Growth Hormone G119R mutant did not bind to ovine GHR-ECD and was devoid of any biological activity in FDC-P1 3B9 cells.
However, in binding experiments that were carried out using chicken liver membranes, both ovine GH and chicken GH showed similar IC50 values in competition with 125I-ovine GH, while the IC50 of G119R mutein was 10-fold higher.
These results emphasize the importance of species specificity and indicate the possibility of antagonistic activity of chGH G119R in homologous system.

    More Info

    • Introduction

GH is a member of the somatotropin/prolactin family of hormones which play an important role in growth control. The gene, along with four other related genes, is located at the growth hormone locus on chromosome 17 where they are interspersed in the same transcriptional orientation; an arrangement which is thought to have evolved by a series of gene duplications. The five genes share a remarkably high degree of sequence identity. Alternative splicing generates additional isoforms of each of the five growth hormones, leading to further diversity and potential for specialization. This particular family member is expressed in the pituitary but not in placental tissue as is the case for the other four genes in the growth hormone locus. Mutations in or deletions of the gene lead to growth hormone deficiency and short stature.

    • Synonyms

GH1, GH, GHN, GH-N, hGH-N, Pituitary growth hormone, Growth hormone 1, Somatotropin.

    • Physical Appearance

Sterile Filtered White lyophilized (freeze-dried) powder.

    • Stability

Lyophilized Growth-Hormone Chicken antagonist although stable at room temperature for at least two weeks, should be stored desiccated below -18°C. Upon reconstitution and filter sterilization GH can be stored at 4°C, pH 9 for up to 4 weeks. For long term storage and more diluted solutions it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.

    • Solubility

It is recommended to reconstitute the lyophilized Growth-Hormone Chicken antagonist in 0.4% NaHCO3 or water adjusted to pH 8-9, not less than 100µg/ml, which can then be further diluted to other aqueous solutions, preferably in a presence of a carrier protein such as BSA or similar.

    • Amino Acid Sequence

The sequence of the first five N-terminal amino acids was determined and was found to be Ala-Thr-Phe-Pro-Ala.

    • Protein content

Protein quantitation was carried out by: UV spectroscopy at 280 nm using the absorbency value of 0.74 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
