Ku antigen consists of two polypeptides (70 kDa and 80 kDa; the p80 subunit is occasionally called p86). The Ku heterodimer provides DNA-binding and regulatory functions to DNA-dependent protein kinase which is a central enzyme for DNA recombination processes in eucaryotic cells. The Ku protein plays important roles in multiple nuclear processes including DNA repair, V(D)J recombination, telomere maintenance, and the regulation of specific gene transcription.

TMEFF1 is a type I transmembrane glycoprotein which includes 2 follistatin modules and an EGF domain in its extracellular domain, a transmembrane domain and a short cytoplasmic tail. The extracellular domain of TMEFF1 can be released as a soluble protein. TMEFF1 is primarily expressed in brain, but is downregulated in brain neoplasms. TMEFF1 selectively regulates nodal but not activin signaling through direct binding to the nodal co-receptor, Cripto. TMEFF inhibits NODAL and BMP signaling through neural patterning and also a possible tumor suppressor in brain cancers.

ETHE1 is a mitochondrial sulfur dioxygenase involved in catabolism of sulfide that accumulates to toxic levels in ethylmalonic encephalopathy. Mutations of ETHE1 were detected in all the typical ethylmalonic encephalopathy patients analysed, but no ETHE1 mutations were identified in patients presenting with early onset progressive encephalopathy with ethylmalonic aciduria.

PTTG1 (Securin) is mainly involved in the regulation of sister chromatid separation during cell division. Securin is a regulatory protein, which has a central role in chromosome stability, in the p53/TP53 pathway, and DNA repair. Additional two roles have been identified for PTTG1; the first one is to facilitate the transport of separase (cysteine protease) to the nucleus and the second role is to hinder the catalytic activity of separase. PTTG1 is ubiquitinated by the APC (Anaphase Promoting Complex), and subsequently degraded by the Proteasome, releasing separase. During the mitosis, PTTG1 blocks Separase/ESPL1 function, thus preventing the proteolysis of the cohesin complex and the ensuing segregation of the chromosomes. However, PTTG1 function is not restricted to a blocking activity only, since it is required to activate ESPL1. PTTG1 is ubiquitinated at the beginning of anaphase, leading to its destruction and to the liberation of ESPL1.

Glycoprotein Nmb (GPNMB) is a member of the PMEL/NMB family. GPNMB is a type I transmembrane glycoprotein which exhibits homology to the pMEL17 precursor, a melanocyte-specific protein. GPNMB is expressed in the lowly metastatic human melanoma cell lines and xenografts but has no expression in the highly metastatic cell lines. GPNMB might be involved in growth delay and reduction of metastatic potential. GPNMB is up-regulated in a number of cancer cells, including in glioblastoma multiforme. GPNMB is expressed in many melanoma cells, as well as in tissue macrophages, including liver Kuppfer cells and lung alveolar macrophages, in podocytes and in some cells of the ciliary body of the eye (at protein level). Dynein light chain Tctex-type 1 (DYNLT1) is a member of the dynein light chain Tctex-type family. DYNLT1 is a dynein light chain involved in cargo binding. DYNLT1 acts as one of several non-catalytic accessory components of the cytoplasmic dynein 1 complex which are believed to be involved in linking dynein to cargos and to adapter proteins that regulate dynein function. Cytoplasmic dynein is a key motor protein complex responsible for minus-end, microtubule-based motile processes. STIP1 is an adaptor protein that mediates the functions of HSP70 & HSP90 in protein folding. The HDGF2 protein augments DNA synthesis and may play a part in cell proliferation. HDGF2 is expressed primarily in the testis and the brain, to an intermediate extent in the heart, and to a slight extent in the ovaries, kidneys, spleen, and liver in humans.
