    • Introduction

Carboxypeptidase-E (CPE ) is a carboxypeptidase that cleaves C-terminal amino acid residues and is involved in the biosynthesis of peptide hormones and neurotransmitters. CPE is a peripheral membrane protein. CPE specifically connects regulated secretory pathway proteins, including prohormones, but constitutively secreted proteins. Mutations in CPE are implicated in type II diabetes.

    • Introduction

ALAD form porphobilinogen (a precursor of heme, cytochromes and other hemoproteins) by catalyzing the compression of 2 molecules of delta-aminolevulinate. ALAD catalyzes the second step in the porphyrin and heme biosynthetic pathway; zinc is vital for enzymatic activity. ALAD has 8 identical subunits and its enzymatic activity is inhibited by lead. Mutations in the ALAD structural gene are the source for high sensitivity to lead poisoning and acute hepatic porphyria.

MUTM is a base excision repair enzyme that identifies and eliminates a large variety of oxidized purines from correspondingly impaired DNA. MUTM is nondismissable and essential to remove quickly its substrate lesions on the chromosome. MUTM, additionally, mends a large number of the lesions recognized by Endo III, signifying that MUTM takes a prominent part in the overall repair of both purine damage and pyrimidine damage in vivo.

    • Introduction

Glutamate Decarboxylase 1 (GAD1) is one of several forms of glutamic acid decarboxylase. GAD1 is responsible for catalyzing the production of gamma-aminobutyric acid from L-glutamic acid. A pathogenic role for the GAD1 enzyme has been identified in the human pancreas since it has been detected as an autoantigen and an autoreactive T cell target type II diabetes. The GAD1 protein may also have a role in the stiff man syndrome. GAD1 enzyme deficiency leads to pyridoxine dependency with seizures. GAD1 also catalyzes the production of GABA.

    • Introduction

Angiotensin is an oligopeptide in the blood that causes vasoconstriction, increased blood pressure, and release of aldosterone from the adrenal cortex. It is a powerful dipsogen. It is derived from the precursor molecule angiotensinogen, a serum globulin produced in the liver. It plays an important role in the renin-angiotensin system.
The protein encoded by this gene, pre-angiotensinogen or angiotensinogen precursor, is expressed in the liver and is cleaved by the enzyme renin in response to lowered blood pressure. The resulting product, angiotensin I is then cleaved by angiotensin converting enzyme (ACE) to generate the physiologically active enzyme angiotensin II. The protein is involved in maintaining blood pressure and in the pathogenesis of essential hypertension and preeclampsia.

    Description

Hyaluronidase is an enzyme that temporarily and reversibly breaks down the polysaccharide, hyaluronic acid, which is found between the cells of connective tissue. Hyaluronic acid may be thought of as the "glue" that holds cells together. Hyaluronic acid is a mucopolysaccharide that exists in the human tissue matrix. It can constrain the diffusion of the extracellular fluid. Hyaluronidase makes the glucoseamine of the hyaluronic acid molecules hydrolyzed and depolymerized, thus decreases the viscosity of the body fluids and increases the flow and diffusion of the intercellular fluids. In this way the physic liquor, exudates or blood in local areas can be more easily diffused, and the drug can be more easily absorbed. Thus the local tissue tension and pains can be relieved. And it will also be easier for the edema and inflammatory exudates to be absorbed and dissolved. This product is a basic component of the articular cartilage. It can nourish, protect and maintain the functions of the joints.

Adenine DNA glycosylase (mutY) is an adenine DNA glycosylase active on DNA substrates including A/G, A/8-oxoG, or A/C mismatches and also has a weak guanine glycosylase activity on G/8- oxoG-containing DNA. mutY is essential for the prevention of mutations resulting from oxidative DNA impairment. Rising levels of mutY in A549 cells exposed to oxygen and infrared radiation leads to improvements in cell survival. mutY is common in neurons where mitochondrial genomes exposed to reactive oxygen species that damage DNA must uphold integrity over the whole mammalian life span.

    • Introduction

Acyl CoA dehydrogenase is the enzymeused to catalyzethe first step of ?-oxidationin Fatty acid metabolism.
Acyl-coenzyme A (CoA) dehydrogenases (ACADs) are a family of mitochondrial enzymes that catalyze the first dehydrogenation step in the bets-oxidation of fatty acyl-CoA derivatives. Several human ACADs exist and all ACADs catalyze the same initial dehydrogenation of the substrate at the beta-carbon atom and require electron transfer flavoprotein as an alectron acceptor. The predicted 415-amino acid ACAD8 protein contains many of the residues conserved in most other ACADs, including an active site glutamic acid residue and residues important for tetramer formation.

    • Introduction

Glutamate Decarboxylase 1 Isoform-1, also known as GAD1 iso1 belongs to the group II decarboxylase family. GAD1 iso1 encodes one of several forms of glutamic acid decarboxylase, and is identified as a major autoantigen in insulin-dependent diabetes. GAD1 iso1 is in charge for catalyzing the production of gamma-aminobutyric acid from L-glutamic acid. In addition, a pathogenic function for GAD1 iso1 has been shown in the human pancreas while it has been identified as an autoantigen & an autoreactive T cell target in insulin-dependent diabetes. GAD1 iso1 play a role in the stiff man syndrome. It has been shown that deficiency in this enzyme has led to pyridoxine dependency with seizures.

    • Introduction

L-threonine to glycine degradation consists of a two-step biochemical pathway which involvs the enzymes L-threonine dehydrogenase and 2-amino-3-ketobutyrate coenzyme A ligase. L-Threonine is initially converted into 2-amino-3-ketobutyrate by L-threonine dehydrogenase. Glycine C-Acetyltransferase (GCAT) is the 2nd enzyme in this pathway, which subsequently catalyzes the reaction between 2-amino-3-ketobutyrate and coenzyme A to form glycine and acetyl-CoA. The GCAT enzyme is regard as a class II pyridoxal-phosphate-dependent aminotransferase. GCAT is strongly expressed in the heart, brain, liver and pancreas. GCAT is also found in lung.
