      SAM Domain SH3 Domain and Nuclear Localization Signal 1 (SAMSN1) is a member of a known adaptors and scaffold proteins containing SH3 and SAM (sterile alpha motif) domains. SAMSN1 is a negative regulator of B-cell activation and is down-regulates cell proliferation. SAMSN1 is up-regulated by IL-4 in activated B cells and expressed mainly in dendritic cells. SAMSN1 owns a function which is similar to other adaptor proteins that link signaling molecules in signal transduction cascades.

    • Introduction

      Troponin I (TnI), troponin T (TnT) and troponin C (TnC) form the troponin complex of the thin filaments of striated muscle. TnI is acts as the inhibitory subunit by blocking actin-myosin interactions and thereby mediating striated muscle relaxation. The TnI subfamily contains 3 genes: TnI-skeletal-fast-twitch, TnI-skeletal-slow-twitch, and TnI-cardiac. The TNNI3 gene encodes the TnI-cardiac protein and is exclusively expressed in cardiac muscle tissues. Mutations in the TNNI3 gene cause familial hypertrophic cardiomyopathy type 7 (CMH7) and familial restrictive cardiomyopathy (RCM).

      Hsp70-binding protein 1 (HSPBP1) is a member of a family of eukaryotic proteins identified as nucleotide exchange factors for HSP 70 that exhibit varying degrees of compartment and species specificity. HSPBP1 is localized mainly in cytoplasm and nucleus but is also found extracellularly. HSPBP1 is mostly expressed in heart and skeletal muscle .HSPBP1 binds to HSP 70, inhibits its activity and promotes dissociation of nucleotides from the HSP 70 ATPase domain. In addition, HSPBP1 inhibits HSPA1A chaperone activity by changing the conformation of the ATP-binding domain of HSPA1A and obstructing the ATP binding. HSPBP1 may also have a role in tumor (dys)regulation of chaperone proteins. Furthermore, HSPBP1 hinders ubiquitination mediated by STUB1 and inhibits chaperone-assisted degradation of immature CFTR.

      Macrophage Inflammatory Proteins (MIP) belong to the family of chemotactic cytokines known as chemokines. In humans, there are two major forms, MIP-1a and MIP-1b that are now officially named CCL3 and CCL4 respectively. They activate human granulocytes (neutrophils, eosinophils and basophils) which can lead to acute neutrophilic inflammation.       FK506 Binding Protein 5 (FKBP5) belongs to the immunophilin protein family, which has a role in immunoregulation and basic cellular processes involving protein folding and trafficking. FKBP5 is a cis-trans prolyl isomerase, which binds to the immunosuppressants FK506 and rapamycin. FKBP5 is an immunophilin protein with PPIase and co-chaperone activities. FKBP5 is assumed to mediate calcineurin inhibition. FKBP5 is a component of unligated steroid receptors heterocomplexes through interaction with heat-shock protein 90 (HSP90). In addition, FKBP5 interacts functionally with mature hetero-oligomeric progesterone receptor complexes along with the 90 kDa heat shock protein and P23 protein.

      BCL2-associated athanogene 2 (BAG2) belongs to the Bag family of proteins. BAG2 is a chief component of the HSC 70/CHIP chaperone-dependent ubiquitin ligase complex and acts to upset CHIP-mediated ubiquitylation. BAG proteins compete with Hip for attaching to the Hsc70/Hsp70 ATPase domain and promote substrate discharge. All the BAG proteins have a roughly 45 amino acid BAG domain by the C-terminus but vary clearly in their N-terminal regions. The BAG domains of BAG1, BAG2, and BAG3 act together expressly with the Hsc70 ATPase domain in vitro and in mammalian cells. All three proteins attach with high affinity to the ATPase domain of Hsc70 and obstruct its chaperone activity in a Hip-repressible manner.
