    • Introduction

      Keratin 8 and 18 (K8/18) are the major components of intermediate filament (IF) proteins of simple or single-layered epithelia.

    • Synonyms

      Keratin type II cytoskeletal 8, Cytokeratin-8, CK-8, Keratin-8, K8, KRT8, CYK8, KO, CK8, K2C8, CARD2.

    • Physical Appearance

      Sterile Filtered White lyophilized (freeze-dried) powder.

    • Stability

      Lyophilized KRT8 although stable at room temperature for 3 weeks, should be stored at 2-8°C. Upon reconstitution KRT8 should be stored at 4°C between 2-7 days and for future use below -18°C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.

    • Solubility

      It is recommended to reconstitute the lyophilized KRT8 in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

    • Reconstitution to filaments

      Performed by mixing equimolar amounts of cytokeratins of type I and type II at concentrations of approx. 0.5 mg/ml, both dissolved in 9.5 M urea buffer (see above). Protofilaments and filament complexes are obtained by dialyzing the resulting polypeptide solution stepwise to a concentration of 4 M urea and then to low salt condition (50 mM NaCl, 2 mM dithiothreitol, 10 mM Tris-HCl, pH 7.4). For immunization purposes, the solution can be further dialyzed against PBS (phosphate buffered saline, e.g. Dulbecco's PBS).

    • Introduction

      PA2G4 belongs to the peptidase M24C family and functions as an RNA-binding protein involved in cellular proliferation and differentiation processes. PA2G4 is a component of pre-ribosomal ribonucleoprotein complexes, participating in ribosome assembly and regulating the later steps of rRNA processing. Also, PA2G4 interacts with ErbB-3 and may function as a modulator of the ErbB-3 mediated signal transduction pathway by regulating the effects of Neuregulin-1. Furthermore, PA2G4 is a transcriptional co-repressor of androgen receptor-regulated genes and other cell cycle regulatory genes through its interactions with histone deacetylases. PA2G4 is implicated in growth inhibition and the induction of differentiation of human cancer cells. In addition, PA2G4 mediates cap-independent translation of specific viral IRESs (internal ribosomal entry site). PA2G4 associates with 28S, 18S and 5.8S mature rRNAs, several rRNA precursors and probably U3 small nucleolar RNA.

      FAM84A is a member of the FAM84 family of proteins and is primarily expressed in testis. FAM84A is restricted to a subcellular membrane region in which there is no interaction between adjoining cells and takes part in cell morphology. Additionally, FAM84A expression increases cell motility which can result in invasion and metastasis of cancer cells. FAM84A is up-regulated in pancreatic cancer, colorectal cancer lung cancer, bladder cancer tissues and cholangiocarcinoma.

      Proteasome subunit alpha type-2 (PSMA2) is a multicatalytic proteinase complex which is characterized by its ability to cleave peptides with Arg, Phe, Tyr, Leu, and Glu adjacent to the leaving group at neutral or slightly basic pH. PSMA2 is a 20S core alpha subunit of proteasome belonging to the peptidase T1A family. PSMA2 functions as a docking domain for the regulatory particles and exterior gates blocking unregulated access to the inner cavity.

    • Introduction

      The conserved 76 amino acid protein ubiquitin (Ub) regulates a host of intracellular processes through its enzymatic conjugation to other cellular proteins.
      Ubiquitination occurs through sequential steps catalyzed by activating (E1), conjugating (E2), and ligase (E3) enzymes. The final step results in the formation of an isopeptide bond between Ub’s C-terminal glycine residue (G76) and a lysine residue of the target protein, although N-terminal ubiquitination is also known.
      Outcomes of this modification include destabilization of the conjugated protein, altered protein trafficking and functional modulation.
      After targeting the protein for specific localizations, ubiquitin is released from the substrate by deubiquitinating enzymes.
      A mutant ubiquitin, having a Gly to Ala substitution at the C-terminus (G76A ubiquitin) supported several downstream reactions of the proteolytic pathway but inhibits the deubiquitination process.
      As consequence, the Ub derivative becomes irreversibly conjugated to protein, shifting the equilibrium between the bound and unbound form in the direction of conjugation, at the expense of the free form.

      DsbA appears to be necessary for correct formulation of disulfide bonds in exported proteins in vivo. DsbA is useful as a standard in immunoblotting.       Calcium-activated nucleotidase 1 (CANT1) is a member of the apyrase family. The CANT1 protein is calcium-dependent nucleotidase with a preference for UDP.       EDA2R (ectodysplasin A2 receptor) mediates the activation of the NF-kappa-B and JNK pathways. Activation seems to be mediated through binding to TRAF3 and TRAF6. In addition, Mutations in EDA give rise to a clinical syndrome characterized by loss of hair, sweat glands, and teeth. EDA2R specifically binds to EDA-A2 isoform. This protein is a type III transmembrane protein of the TNFR (tumor necrosis factor receptor) superfamily, and contains 3 cysteine-rich repeats and a single transmembrane domain however it lacks an N-terminal signal peptide. Alternatively spliced transcript variants have been found for this gene.
