    • Introduction

      ErbB3, also called Her3 (human epidermal growth factor receptor 3), is a type I membrane glycoprotein that is a member of the ErbB family of tyrosine kinase receptors. ErbB family members serve as receptors for the epidermal growth factor (EGF) family of growth factors. Among ErbB family members, ErbB3 is unique in that it contains a defective kinase domain. ErbB3 is expressed in keratinocytes, melanocytes, skeletal muscle cells, embryonic myoblasts and Schwann cells. Monomeric ErbB3 serves as a low affinity receptor for the heregulins (HRG). ErbB3 can induce specific antibody production in vivo, hence to inhibit tumor cell growth. ErbB-3 can be used to treat early, medium and advanced or post-operative breast cancer with over-expression of ErbB2. According to its mechanism of action, ErbB3 is classified as a therapeutic for cancer.

TNFRSF8 has a role in the regulation of cellular growth and transformation of activated lymphoblasts. In addition, the TNFRSF8 protein regulates gene expression via activation of NF-kappa-B.       CDC34 takes part in the control of cell cycle and DNA replication. Cdc34 in association with different E3 complexes, including SCF, targets many different substrates for ubiquitination and degradation during cell division, signal transduction, and development. Cdc34 substrates that have been characterized include B-Myb, Wee1, MyoD, ATF5, p27Xic1, and p27Kip1. Additionally, substrates such as p21Cip1, E2F, cyclin E, and cyclin D are putative substrates of Cdc34 by virtue of their SCF requirement for proteolysis. Cdc34 is self-associate through a domain in the C-terminus, and is phosphorylated and ubiquitinylated in vivo. CDC34is useful for in vitro ubiquitinylation reactions.

      Listeriolysin O (aka LLO) is a hemolysin produced by Listeria monocytogenes bacteria, the pathogen responsible for causing listeriosis. The toxin may be regarded as a virulence factor, since it is crucial for the virulence of L. monocytogenes. LLO is a single polypeptide protein encoded by the hlyA gene and composed of 529 residues. LLO is a thiol-activated cholesterol-dependent pore forming toxin protein; therefore, it is activated by reducing agents and inhibited by oxidizing agents. Still, LLO differs from other thiol-activated toxins, as its cytolytic activity is maximized at a pH of 5.5. Inside the acidic phagosomes (average pH ~ 5.9) of cells that have phagocytosed L. monocytogenes, LLO is selectively activated by maximizing activity at a pH of 5.5.       Hematological and neurological expressed 1 protein (HN1) is a small protein which is extremely preserved among species. HN1 expression is upregulated in regenerating neural tissues, including the axotomized adult rodent facial motor nerve and dedifferentiating retinal pigment epithelial cells of the Japanese newt. HN1 is also expressed in several tissues during embryonic development as well as in regions of the adult brain which display high plasticity.

    • Introduction

      HP-NAP protects DNA from oxidative damage by sequestering intracellular Fe2+ ion and storing it in the form of Fe3+ oxyhydroxide mineral. One hydrogen peroxide oxidizes two Fe2+ ions, thus preventing hydroxyl radical production by the Fenton reaction. HP-NAP is necessary for the survival in the presence of oxidative stress. Dps is also a virulence factor which activates neutrophils, mast cells and monocytes. HP-NAP binds to neutrophil-glycosphingolipids and to sulfated carbohydrates on mucin. HP-NAP might play a part in the accumulation of neutrophils and monocytes at the site of infection. HP-NAP induces superoxide anion generation, adhesion and chemotaxis of neutrophils, via a pertussis toxin-sensitive pathway involving MAP kinases.

      SF3B14 is a 125 amino acid nuclear protein which is a component of the splicing factor 3b complex. Splicing factor 3b is involved with s with both the U2 and U11/U12 small nuclear ribonucleoprotein complexes (U2 snRNP) of spliceosomes. SF3B14 which is required for the splicing of pre-mRNA enters the spliceosome and connect with the pre-mRNA branch site facilitating the interaction of snRNP with the branch sites of U2 and U12 of the 17S U2 and the 18S U11/U12 snRNP complex.

      Small nuclear ribonucleoprotein complexes (abbreviated as U-snRNP) are essential for splicing of precursor mRNA molecules. U1-snRNP is the most abundant RNP particle in the nucleus and consists of one small uridylate-rich RNA (U1 RNA) complexed with several proteins: the three 68/70 kDa, A, C polypeptides are unique to the U1-snRNP particle, whereas 7 so-called Sm proteins (B/B', D1, D2, D3, E, F, G) form a core subparticle that is common to all U-snRNP complexes.
      Both the U1-specific proteins and the Sm core particle are targets of autoantibodies which classically have been called the RNP and RNP-Sm specificities, respectively. A clean diagnostic distinction of these specificities has been complicated by the biochemical difficulties of producing clean subparticle fractions from native sources. The use of single recombinant proteins as antigenic targets guarantees a much higher sensitivity and specificity and is the only way to determine RNP antibodies sensu stricto without the disturbing influence of Sm antigens; also with single U1 proteins antibodies will be detected which can be missed because of steric hindrance when using the RNP-Sm complex in an assay.
      Autoantibodies to U1-snRNP are present in 95% of patients with Mixed Connective Tissue Disease (MCTD) and 30% of patients with SLE. Antibodies against the 68/70 kDa protein are known to have a high clinical significance in MCTD patients. The 68/70 kDa nomenclature of this protein refers to the fact that different splice variants of the protein are found in human cells.
