    More Info

    • Introduction

RAD51 family members are evolutionarily conserved proteins vital for DNA repair by homologous recombination. RAD51B is a part of the RAD51 protein family. RAD51B is involved in the homologous recombination repair (HRR) pathway of double-stranded DNA breaks occurring during DNA replication or induced by DNA-damaging agents. RAD51B forms a stable heterodimer with the family member RAD51C, which interacts with the other family members, such as RAD51, XRCC2, and XRCC3. Overexpression of RAD51B leads to cell cycle G1 delay and cell apoptosis.

    • Synonyms

      RAD51 homolog B (S. cerevisiae), R51H2, RAD51L1, REC2, DNA repair protein RAD51 homolog 2, Rad51B, RAD51-like protein 1, RAD51B.

    Description

GADD45G Human Recombinant produced in E.Coli is a single, non-glycosylated polypeptide chain containing 159 amino acids and having a molecular mass of 17.1 kDa.

    Source

    Escherichia Coli.

    Formulation

    The GADD45G protein solution (1mg/ml) contains 20mM Tris-HCl pH-7.

    Purity

Greater than 95.0% as determined by SDS-PAGE.

    More Info

    • Introduction

GADD45G is part of the nuclear proteins to interact with various proteins whose transcript levels are raised after stressful growth arrest conditions and treatment with DNA-damaging agents. GADD45G reacts to environmental stresses by mediating activation of the p38/JNK pathway which is mediated through their protein binding and activating MTK1/MEKK4 kinase, which is an upstream activator of both p38 and JNK MAPKs. GADD45G acts as a new-age tumor suppressor however is being frequently inactivated epigenetically in multiple tumors. GADD45G mRNA expression is down-regulated in hepatocellular carcinoma. GADD45G causes cell cycle arrest at G2/M transition when transfected into Hep-G2 cells. GADD45 Gamma induction by androgens involves new protein synthesis. Overexpression of GADD45 Gamma inhibits cell growth and causes morphological modifications in prostate cell lines thus GADD45 gamma takes part in differentiation induction by androgens.

    • Synonyms

      DDIT2, GADD45gamma, GADD45G, Growth arrest and DNA-damage-inducible protein GADD45 gamma, Cytokine-responsive protein CR6, DNA-damage-inducible transcript 2, DDIT-2, CR6, GRP17.

    • Introduction

Seryl-tRNA synthetase, cytoplasmic (SARS) is a member of the class-II aminoacyl-tRNA synthetase family. Aminoacyl-tRNA synthetases’ role is to catalyze the aminoacylation of tRNAs by their corresponding amino acids, as a result linking amino acids with tRNA-contained nucleotide triplets. The SARS enzyme catalyzes the attachment of serine to tRNA (Ser). SARS enzyme is probably able to aminoacylate tRNA (Sec) with serine, to form the misacylated tRNA L-seryl-tRNA (Sec), which will be then converted into selenocysteinyl-tRNA (Sec).

    • Synonyms

Serine--tRNA ligase cytoplasmic, Seryl-tRNA synthetase, SerRS, Seryl-tRNA(Ser/Sec) synthetase, SARS, SERS.

    • Physical Appearance

      Sterile Filtered colorless solution.

    • Introduction

TIGAR is a p53-inducible enzyme which catalyzes the hydrolysis of fructose-2-6 bisphosphate (F-2-6-BP) to fructose-6-phosphate and inorganic phosphate. F-2-6-BP is an influential activator of 6-phosphofructose-1 kinase (the rate limiting enzyme of glycolysis). By lowering the intracellular level of F-2-6-BP, TIGAR expression leads to increased glucose processing through the pentose phosphate pathway, the main cellular source for NADPH. Protein transduction using TAT fusion proteins represents an alternative methodology for introducing transcription factors and other intracellular proteins into primary as well as transformed cells.

    • Introduction

Human immunodeficiency virus type-1 (HIV-1) regulatory Tat protein plays an essential role in viral replication (Jones KA, 1994) and infectivity (Arya SK, 1985; Fisher AG, 1986). In addition, during acute infection, Tat is released extracellularly by infected cells (Chang HC, 1997; Ensoli B, 1990) and is taken up by neighboring cells where it transactivates viral replication (Ensoli B, 1993) and increases virus infectivity.
HIV-1 Tat activates transcription of HIV-1 viral genes by inducing phosphorylation of the C-terminal domain (CTD) of RNA polymerase II (RNAPII). Tat can also disturb cellular metabolism by inhibiting proliferation of antigen-specific T lymphocytes and by inducing cellular apoptosis. Tat-induced apoptosis of T-cells is attributed, in part, to the distortion of microtubules polymerization. LIS1 is a microtubule-associated protein that facilitates microtubule polymerization.

    • Introduction

RAD1 is part of a heterotrimeric cell cycle checkpoint complex, recognized as the 9-1-1 complex, that is activated to stop cell cycle progression in reaction to DNA damage or incomplete DNA replication. RAD1 complex withholds the Rad9 and Hus1 proteins and takes part in cellular responses to DNA damage, by associating with Rad17 and several components of the PCNA-loading heteropentamer, replication factor C. RAD1 takes part in DNA repair and recruited to DNA lesion upon damage by the RAD17-replication factor C (RFC) clamp loader complex. RAD1 takes part as a sliding clamp platform on DNA for several proteins that play a role in long-patch base excision repair (LP-BER). RAD1 complex stimulates DNA polymerase beta (POLB) activity by raising its affinity for the 3''-OH end of the primer-template and stabilizes POLB to those sites where LP-BER proceeds.

Furthermore, POLL takes part in the mutagenic bypass of T-T dimers. Pol lambda possesses terminal transferase and 5’-deoxyribose-5-phosphate lyase activity which is synchronized by PCNA and replication protein A.

    • Synonyms

Polymerase (DNA directed) lambda, Pol beta2, DNA polymerase beta-2, DNA polymerase kappa, BETAN, POLKAPPA, DNA polymerase beta-N, EC 2.7.7.7, EC 4.2.99.

    • Physical Appearance

Sterile Filtered and lyophilized, though might appear as a solution as a result of the glycerol content.

    • Stability

Store at 4°C if entire vial will be used within 2-4 weeks. Store, frozen at -20°C for longer periods of time. Avoid multiple freeze-thaw cycles.

    • Solubility

It is recommended to reconstitute the lyophilized HIV-1 TAT in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
