a-Synuclein (amino acids 1-140), an acidic neuronal protein of 140 amino acids, is extremely heat-resistant and is natively unfolded with an extended structure primarily composed of random coils. a-synuclein has been suggested to be implicated in the pathogenesis of Parkinson’s disease and related neurodegenerative disorders, and more recently, to be an important regulatory component of vesicular transport in neuronal cells. Moreover, recent studies have shown that a-synuclein has chaperone activity and that this activity is lost upon removing its C-terminal acidic tail (amino acids 96-140).

Vps20-associated 1 homolog (VTA1) is a member of the VTA1 family. The VTA1 protein is involved in the endosomal multivesicular bodies (MVB) pathway. Furthermore, VTA1 is interacts with CHMP1B, CHMP2A, CHMP5, VPS4B, KIAA0174 (IST1) and possibly CHMP3. VTA1 is believed to be a cofactor of VPS4A/B, which catalyzes membrane-associated ESCRT-III assemblies. Microfibrillar-associated protein 4 (MFAP4) is a member of Fibrinogen protein family and contains 1 fibrinogen C-terminal domain. The MFAP4 protein has similarity to a bovine microfibril-associated protein. MFAP4 has binding specificities for both collagen and carbohydrate. MFAP4 is believed to be an extracellular matrix protein that is involved in cell adhesion or intercellular interactions. MFAP4 deletion was found in 30 of 31 Smith-Magenis syndrome (SMS) patients.

VAPB Vesicle-associated membrane protein (VAMP)-associated protein B (aka VAPB) is a type IV transmembrane protein and belongs to the VAP family of proteins. VAPB may have a role in vesicle trafficking. VAPB is found in plasma and intracellular vesicle membranes as a homodimer and heterodimer with VAPA, and interacts with VAMP1 and VAMP2. VAPB defects are the basis for the amyotrophic lateral sclerosis type 8 and spinal muscular atrophy autosomal dominant Finkel type.

Alpha-synuclein or SNCA is a synuclein protein. SNCA mainly found in the brain, small concentration of the protein can also be located in other tissues such as heart and muscle. When looking in the brain tissue, SNCA is located in the end of the neuron, in an area called presynaptic terminal. In the presynaptic terminal SNCA has interaction with phospholipids & other proteins. Neurotransmitters are released from the synaptic vesicles in the Presynaptic terminals and act as messengers. Once released, the neurotransmitters send signals across the neurons that are crucial for the brain’s operation.

SNCG is also identified in the brain, ovarian tumors, and in the olfactory epithelium. SNCG expression in breast tumors is a marker for tumor progression. A modification in the expression of gamma-synuclein has been detected in the retina of Alzheimer's patients.

Syntaxin is membrane integrated Q-SNARE protein participating in exocytosis. Syntaxin is composed of an N-terminal regulatory domain (Habc), a SNARE domain (known as H3), and a single C-terminal transmembrane domain. The SNARE (H3) domain binds to both synaptobrevin and SNAP-25 forming the core SNARE complex. Synaptic vesicles store neurotransmitters that are released during calcium-regulated exocytosis.
