    • Introduction

UBE2E1 belongs to the E2 ubiquitin-conjugating enzyme family. UBE2E1 accepts ubiquitin from the E1 complex and catalyzes its covalent attachment to other proteins. UBE2E1 catalyzes the covalent attachment of ISG15 to other proteins. UBE2E1 mediates the selective degradation of short-lived and abnormal proteins. In vitro, UBE2E1 also catalyzes 'Lys-48'-linked polyubiquitination.

      Ubiquitin carrier protein E1, Ubiquitin-conjugating enzyme E2E 1 (homologous to yeast UBC4/5), Ubiquitin-protein ligase E1, UBCH6, EC 6.3.2.19.

    • Physical Appearance

      Sterile Filtered colorless solution.

    • Stability

Store at 4°C if entire vial will be used within 2-4 weeks. Store, frozen at -20°C for longer periods of time. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).Avoid multiple freeze-thaw cycles.

    • Introduction

UFC1 is a member of the ubiquitin-conjugating enzyme family. UFC1 is an E2-like conjugating enzyme for ubiquitin-fold modifier-1. UFM1 is activated by UBA5 (a novel E1-like enzyme) by forming a high-energy thioester bond. Activated UFM1 is subsequently transferred to its cognate E2-like enzyme, UFC1, in a similar thioester linkage.

    • Introduction

Ubiquitin-like modifier activating enzyme 5 (UBA5) is a member of the ubiquitin-activating E1 family and UBA5 subfamily. Ubiquitin and ubiquitin-like proteins are recognized as covalently conjugated to various cellular substrates by a three-step enzymatic pathway. The ubiquitin-activating enzyme (E1) has a vital role in the first step of ubiquitination pathway to activate ubiquitin or ubiquitin-like proteins. UBA5 activates an ubiquitin-like protein, ubiquitin-fold modifier 1 (Ufm1), by forming a high-energy thioester bond. UBA5 is located primarily in cytoplasm, while it generally localizes to the nucleus in presence of SUMO2.

    • Introduction

Human Ubc9 is homologous to ubiquitin-conjugating enzymes (E2s). However, instead of conjugating ubiquitin, it conjugates a ubiquitin homologue, small ubiquitin-like modifier 1(SUMO-1). And hUbc9 retains striking structural and functional conservation with yeast Ubc9. The ubiquitin-dependent protein degradation system has been recognized as a complete enzymatic pathway that is responsible for the selective degradation of abnormal and short-lived proteins. The conjugation of ubiquitin requires the activities of ubiquitin-activating (E1) and –conjugating (E2) enzymes.

    • Introduction

The alteration of proteins with ubiquitin is a vital cellular mechanism for targeting atypical or short-lived proteins for degradation. Ubiquitination involves noT less than three classes of enzymes: ubiquitin-activating enzymes, or E1s, ubiquitin-conjugating enzymes, or E2s, and ubiquitin-protein ligases, or E3s. UBE2A is part of the E2 ubiquitin-conjugating enzyme family. UBE2A is necessary for post-replicative DNA damage repair. UBE2A catalyzes the covalent attachment of ubiquitin to other proteins. UBE2A is necessary for postreplication repair of UV-damaged DNA.

    • Introduction

Ubiquitin-conjugating enzyme E2 F (UBE2F) is a member of the ubiquitin-conjugating enzyme family and UBE2F subfamily. Ubiquitin-conjugating enzymes or sometimes as ubiquitin-carrier enzymes, run the second step in the ubiquitination reaction which targets a protein for degradation using the proteasome. The UBE2F receives the ubiquitin-like protein NEDD8 from the UBA3-NAE1 E1 complex and catalyzes its covalent attachment to other proteins.
RBX2-UBE2F complex neddylates specific target proteins, for example CUL5 as can be seen from the specific interaction with the E3 ubiquitin ligase RBX2, but not RBX1.

    • Introduction

UBE2D1 is a member of the ubiquitin-conjugating enzyme family. Ubiquitination involves at least 3 classes of enzymes: ubiquitin-activating enzymes (E1s), ubiquitin-conjugating enzymes (E2s) and ubiquitin-protein ligases (E3s). UBE2D1 is strongly related to a stimulator of iron transport (SFT), and is up-regulated in hereditary hemochromatosis. In addition, UBE2D1 functions in the ubiquitination of the tumor-suppressor protein p53 and the hypoxia-inducible transcription factor HIF1alpha by interacting with the E1 ubiquitin-activating enzyme and the E3 ubiquitin-protein ligases.

    • Introduction

Cytochrome P450 2D6 is a member of a complex family of microsomal enzymes (mono-oxygenases) present in the endoplasmic reticulum membrane, which perform detoxification reaction on xenobiotic compounds. Cytochrome P450 2D6 is the molecular target of autoantibodies against the "liver kidney microsomal antigen 1" (LKM 1) which has been classically defined by immunofluorescence microscopy. The presence of these autoantibodies is considered indicative of Autoimmune Hepatitis Type 2; LKM 1 antibodies have also been detected in patients with hepatitis C viral infection. The International Autoimmune Hepatitis Group therefore has subdivided the AIH type 2 into two subgroups: 2a with HCV and 2b without HCV. AIH 2a patients are often over 40 and predominantly male.
The use of a purified recombinant Cytochrome P450 2D6 antigen allows the differentiation of autoimmune hepatitis from drug-induced hepatitis where transient autoantibodies to other P450 family members occur which cannot be differentiated by immunofluorescence techniques.

    • Introduction

Protein modification with ubiquitin is an essential cellular mechanism for targeting abnormal or short-lived proteins for degradation. Ubiquitination involves at least 3 classes of enzymes: ubiquitin-activating enzymes (E1s) ubiquitin-conjugating enzymes (E2s) and ubiquitin-protein ligases (E3s). Ubiquitin-Conjugating Enzyme E2G (UBE2G1) belongs to the E2 ubiquitin-conjugating enzyme family and catalyzes the covalent attachment of ubiquitin to other proteins. UE2G1 protein is involved in degradation of muscle-specific proteins.
