    More Info

    • Introduction

Uteroglobin (SCGB1A1) which belongs to the Secretoglobin (SCGBs) superfamily, is a multifunctional protein that exerts anti-inflammatory and anti-tumorigenic effects by binding small hydrophobic molecules such as phospholipids and prostaglandins. Uteroglobin is involved in numerous functions including anti-inflammation, inhibition of phospholipase A2 and the sequestering of hydrophobic ligands. SCGB1A1 is expressed by Clara cells, the non-ciliated, non-mucous secretory cells predominant in lung bronchioles, and by other epithelia which communicate with the external environment. On top of sequestering pro-inflammatory mediators and carcinogens, Uteroglobin is implicated in the inhibition of cell migration and invasion, platelet aggregation, and T cell differentiation. SCGB1A1 gene defects are associated with a susceptibility to asthma.

    • Synonyms

Uteroglobin, Clara cell 17 kDa protein, Clara cell phospholipid-binding protein, CCPBP, Clara cells 10 kDa secretory protein, CC10, PCB-binding protein, Secretoglobin family 1A member 1, Scgb1a1, Ugb, Utg, UG, CC16, CCSP, PCB-BP.

    • Physical Appearance

Sterile Filtered White lyophilized (freeze-dried) powder.

    • Stability

Lyophilized SCGB1A1 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution SCGB1A1 should be stored at 4°C between 2-7 days and for future use below -18°C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.

    • Solubility

It is recommended to reconstitute the lyophilized SCGB1A1 in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

    • Introduction

The bone morphogenetic proteins (BMPs) are a family of secreted signaling molecules, which can induce ectopic bone growth. Various BMPs are part of the transforming growth factor-beta (TGFB) superfamily. BMPs were initially identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. Based upon its expression early in embryogenesis, BMP6 has a suggested role in early development. Moreover, the fact that the BMP6 is closely related to BMP5 and BMP7 leads to an assumption of possible bone inductive activity.

      Abstract:

Bone Morphogenetic Protein-6 (BMP-6) human recombinant is a critical member of the bone morphogenetic protein family, known for its pivotal role in tissue development, repair, and regeneration. This research paper aims to provide a comprehensive analysis of BMP-6, including its characteristics, signaling pathways, and potential therapeutic applications. Moreover, innovative methodologies for the production and optimization of BMP-6 human recombinant are proposed, shedding light on its future implications in the field of tissue engineering and regenerative medicine.

      Introduction:

Tissue engineering and regenerative medicine have emerged as promising approaches to address the challenges associated with tissue repair and regeneration. BMP-6, a prominent member of the BMP family, plays a key role in regulating cellular responses during tissue development and healing. This paper explores the distinctive features of BMP-6 and presents novel approaches for the production and optimization of BMP-6 human recombinant, aiming to unlock its therapeutic potential in diverse regenerative contexts.

      Characteristics and Signaling Pathways:

BMP-6 is a secreted growth factor belonging to the transforming growth factor-beta (TGF-β) superfamily. It exerts its biological effects by binding to specific cell surface receptors, initiating intricate intracellular signaling pathways. BMP-6 signaling cascades, including Smad-dependent and Smad-independent pathways, orchestrate critical processes such as cell differentiation, proliferation, and extracellular matrix synthesis, thereby influencing tissue development and repair.

      Production of BMP-6 Human Recombinant:

Efficient production methodologies are vital for harnessing the therapeutic potential of BMP-6 human recombinant. Recombinant protein expression systems, including mammalian cells or baculovirus-insect cell systems, have been employed for the production of functional BMP-6. Optimization strategies, such as codon optimization, signal peptide engineering, and protein folding enhancement, have been implemented to enhance the yield and bioactivity of BMP-6 recombinant protein.

      Potential Therapeutic Applications:

BMP-6 human recombinant holds immense promise in the field of tissue engineering and regenerative medicine. Its regulatory role in bone formation, cartilage regeneration, and wound healing positions it as a potential therapeutic candidate for the treatment of skeletal disorders, osteoarthritis, and tissue injuries. Furthermore, the ability of BMP-6 to modulate cell behavior and tissue remodeling indicates its wider therapeutic applications in various regenerative processes.

      Conclusion:

BMP-6 human recombinant emerges as a crucial regulator in tissue engineering and regenerative medicine, offering significant potential for tissue repair and regeneration. Optimizing production methodologies and further unraveling its signaling mechanisms will undoubtedly enhance its therapeutic applications. Given its involvement in bone and cartilage formation, as well as wound healing, BMP-6 human recombinant represents a valuable tool for promoting tissue regeneration and addressing the unmet clinical needs in regenerative medicine.

    • Introduction

Vascular endothelial growth factor is an important signaling protein involved in both angiogenesis and vasculogenesis.VEGF activity has been mostly studied on cells of the vascular endothelium, although it does have effects on a number of other cell types (e.g. stimulation monocyte/macrophage migration, neurons, cancer cells, kidney epithelial cells ).VEGF mediates increased vascular permeability, induces angiogenesis, vasculogenesis and endothelial cell growth, promotes cell migration, and inhibits apoptosis. In vitro, VEGF has been shown to stimulate endothelial cell mitogenesis and cell migration. VEGF is also a vasodilator and increases microvascular permeability and was originally referred to as vascular permeability factor.

    • Introduction

FKBP1A is a 12kDa protein initially discovered on an immune cells on the basis of its capability to bind as well as mediate the intracellular effect of the immunosuppressant FK506. FKBP1A is also known to mediate the action of Rapamycin-immunosuppressive agent. FKBP1A is part of the family of immunophilins, which have in common high affinity for immunosuppressant drugs and a peptidyl-prolyl cis-trans isomerase (PPIase). Activity which participates in folding of proline-containing protein. In the absence of immunosuppressive ligands, FKBP1A is involved in intracellular calcium regulation by associating with 3 types of Ca2+ release channel complexes: skeletal ryanodine receptors, cardiac ryanodine receptors and the inositol 1,4,5-triphosphate receptor. FKBP1A also interact with TGF-beta type I receptor exerting an inhibitory effect on the TGF-beta signaling pathway. FKBP12 plays a role in modulation of ryanodine receptor isoform-1 (ryr-1), a component of the calcium release channel of skeletal muscle sarcoplasmic reticulum. FKBP1A increase the folding of proteins and catalyzes the cis-trans isomerization of proline imidic peptide bonds in oligopeptides.

    • Introduction

FKBP4 is part of the immunophilin protein family, which takes part in immunoregulation and necessary cellular processes concerning protein folding and trafficking. FKBP4 is a cis-trans prolyl isomerase that connects to the immunosuppressants FK506 and rapamycin. FKBP4 has high structural and functional similarity to FKBP1A, though, FKBP4 does not have immunosuppressant activity when complexed with FK506. FKBP4 interacts with interferon regulatory factor-4 and is involved in immunoregulatory gene expression in B and T lymphocytes. FKBP4 is known to connect with phytanoyl-CoA alpha-hydroxylase. FKBP4 associates with HSP90 & HSP70 thus takes part in the intracellular trafficking of hetero-oligomeric forms of the steroid hormone receptors. FKBP4 highly associates with adeno-associated virus type 2 vectors (AAV) resulting in a considerable increase in AAV-mediated transgene expression in human cell lines. FKBP4 is involved in the optimal use of AAV vectors in human gene therapy.

    • Introduction

Acidic fibroblast growth factor is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes, including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth and invasion. FGF1 functions as a modifier of endothelial cell migration and proliferation, as well as an angiogenic factor.FGF1 acts as a mitogen for a variety of mesoderm- and neuroectoderm-derived cells in vitro, thus is thought to be involved in organogenesis. Three alternatively spliced variants encoding different isoforms have been described. The binding growth factors are angiogenic agents in vivo and are potent mitogens for a variety of cell types in vitro. There are differences in the tissue distribution and concentration of these 2 growth factors.

    • Introduction

Protein Interacting with PRKCA 1, also known as PICK1 includes a PDZ domain, through which it interacts with protein kinase C, alpha (PRKCA). PICK1 performs as an adaptor which binds to and arranges the subcellular localization of various membrane proteins. PICK1 interacts with multiple glutamate receptor subtypes, monoamine plasma membrane transporters, in addition to non-voltage gated sodium channels, and might target PRKCA to these membrane proteins and therefore regulates their distribution as well as function. PICK1 has also been found to perform as an anchoring protein which particularly targets PRKCA to mitochondria in a ligand-specific manner.

    • Introduction

Pleiotrophin (Osteoblast-Specific Factor-1, OSF-1) contains 136 amino acid residues. The sequence is very rich in cationic amino acids (24% of the residues); lysine cluster sequences are found in the N-terminal and C-terminal ends of the structure.
The OSF-1 gene was shown by Northern blotting analysis to be expressed in mouse calvarial osteoblast-enriched cells and in mouse brain tissues, but not in thymus, spleen, kidney, liver, lung, testis or heart. Pleiotrophin has the ability to promote adhesion, migration, expansion, and differentiation of human osteoprogenitor cells. In addition to certain types of cancer, the embryonic growth and differentiation factor pleiotrophin is found also in adults in inflammatory diseases. In osteoarthritis, pleiotrophin is especially expressed in early stages, and its concentrations in the synovial fluid could serve as a marker for the progress of the disease. Pleitrophin might be involved in cartilage repair in osteoarthritis, in particular, in earlier stages.
