S100A6 is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized either in the cytoplasm or the nucleus of a wide range of cells. S100 proteins are involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. There are at least 13 members in the S100 gene family, which are located as a cluster on chromosome 1q21.
S100A6 may function in stimulation prolactin secretion, and exocytosis. Chromosomal rearrangements and altered expression of the S100A6 gene are implicated in melanoma.

DFF is a heterodimeric protein of 40kDa (DFFB) and 45kDa (DFFA) subunits. DFFA (DNA fragmentation factor subunit alpha) is the substrate for caspase-3 and triggers DNA fragmentation during apoptosis. DFF is activated once DFFA is cleaved by caspase-3. The cleaved fragments of DFFA detach from DFFB (the active component of DFF), which in turn triggers DNA fragmentation as well as chromatin condensation during apoptosis. Apoptosis is accompanied by shrinkage and fragmentation of the cells and nuclei and degradation of the chromosomal DNA into nucleosomal units.
A reduced level of DFFA detected in ovarian endometriosis may be a part of an apoptosis-resistant mechanism enhancing the disease progression.
DFFA at chromosome 1 shows rare allelic variants in neuroblastoma tumors.

The 14-3-3 family of proteins plays a key regulatory role in signal transduction, checkpoint control, apoptotic and nutrient-sensing pathways. 14-3-3 proteins are highly conserved and ubiquitously expressed. There are at least seven isoforms, β, γ, ε, σ, ζ, τ and η that have been identified in mammals. The 14-3-3 epsilon, a subtype of the 14-3-3 family of proteins, was thought to be brain and neuron-specific. It has been shown to interact with CDC25 phosphatases, RAF1 and IRS1 proteins, suggesting its role in diverse biochemical activities related to signal transduction, such as cell division and regulation of insulin sensitivity. It has also been implicated in the pathogenesis of small cell lung cancer.

Secreted frizzled-related protein 4 (SFRP4) belongs to the SFRP family which contains a cysteine-rich domain homologous to the putative Wnt-binding site of Frizzled proteins. SFRPs serve as soluble modulators of Wnt signaling. SFRP4 may serve as a regulator of adult uterine morphology and function. SFRP4 increases apoptosis during ovulation possibly via modulation of FZ1/FZ4/WNT4 signaling. SFRP4 also has phosphaturic effects by specifically inhibiting sodium-dependent phosphate uptake. SFRP4 is expressed in proliferative endometrium and several types of ovarian, endometrial and Brest tumors. SFRP4 is expressed in mesenchymal cells and in cardiomyocytes. SFRP4 expression in ventricular myocardium correlates with apoptosis related gene expression. SFRP4 is up-regulated in failing myocardium.

BUD31 is a member of the BUD31 (G10) family. BUD31 is a nuclear protein which contains a putative nuclear translocation sequence, an N-terminal acidic domain and a cysteine-rich C-terminal domain containing a putative Zinc-finger structure. Based on the structure of BUD31 it is suggested that BUD31 might be a nuclear regulator of transcription.

High levels of DKK1 in bone marrow plasma and peripheral blood is associated with the presence of osteolytic bone lesions in patients with multiple myeloma. Induction of DKK1 contributes to the pathological cascade triggered by beta-amyloid and is significantly involved in the development of tau phosphorylation.
DKK1 reduces the differentiation of osteoblastic precursor cells. DKK1 is an important member in multiple myeloma bone disease. DKK1 expression is reduced in human colon tumors, implying that DKK-1 plays a role as a tumor suppressor gene in this neoplasia; the Wnt/beta-catenin pathway is downregulated by the induction of DKK1 expression, a mechanism that is missing in colon cancer.

Hemochromatosis (HFE) is a member of the MHC class I family. The Hemochromatosis protein contains 1 Ig-like C1-type (immunoglobulin-like) domain. HFE is a membrane protein, which is similar to MHC class I-type proteins and associates with beta-2 microglobulin (beta2M). It is assumed that the HFE protein acts to regulate iron absorption by regulating the interaction of the transferrin receptor (TFR) with transferrin. FABP multigene family has almost 20 known members. FABPs are divided into 3 different types: hepatic, intestinal and cardiac which form 14-15 kDa proteins that take part in the uptake, intracellular metabolism and/or transport of long-chain fatty acids. FABPs are involved in the modulation of cell growth and proliferation. Intestinal FABP (FABP2) gene has a polymorphism at codon 54 that identified an alanine-encoding allele and a threonine-encoding allele. Thr-54 protein is associated with increased fat oxidation and resistance. High serum levels of FABP2 is ulcerative colitis indicates ileitis. FABP2 is has part in triglyceride-rich lipoprotein synthesis. BEN domain-containing protein 6 (BEND6) is a neural BEN-solo factor which shares many functional attributes with Drosophila Insensitive, a co-repressor for the Drosophila CSL factor. The BEND6 protein binds the mammalian CSL protein CBF1 and antagonizes Notch-dependent target activation. Furthermore, its association with Notch- and CBF1-regulated enhancers is stimulated by CBF1 and antagonized by activated Notch. In utero electroporation experiments demonstrated that ectopic BEND6 impeded Notch-mediated self-renewal of neocortical neural stem cells and promoted neurogenesis.
