    • Introduction

IL2 is a secreted cytokine that is important for the proliferation of T and B lymphocytes. The receptor of this cytokine is a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL4) and interleukin 7 (IL7). The expression of this gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a similar gene in mice leads to ulcerative colitis-like disease, which suggests an essential role of this gene in the immune response to antigenic stimuli.

    • Introduction

IL-17 family members are glycoproteins secreted as dimers which induce local cytokine production and recruit granulocytes to sites of inflammation. The IL-17 family is comprised of at least six pro-inflammatory cytokines that share a conserved cysteine-knot structure but diverge at the N-terminus. IL-17 is induced by IL-15 and IL-23, mostly in activated CD4+ T cells distinct from Th1 or Th2 cells. IL-17B binds the IL-17B receptor, but not the IL-17 receptor; it is most homologous with IL-17D, which is expressed by resting CD4+ T cells and CD19+ B cells. IL17B Diseases associated with IL17B include spondyloarthropathy, and neuronitis, and among its related super-pathways are Mucin expression in CF via IL-6, IL-17 signaling pathways and STAT3 Pathway.

    • Introduction

OTOR proteins is also known as fibrocyte-derived protein (Fdp) and Melanoma inhibitory activity-like (MIAL). Otoraplin is a member of the melanoma-inhibiting activity gene family. Otoraplin is a secreted 16 kDa globular protein that is expressed in the inner ear by periotic mesenchyme and developing and mature fibrocytes. OTOR is highly homologous to MIA/cartilage-derived retinoic acid-sensitive protein (CD-RAP), which is a cartilage-specific protein that is also expressed in malignant melanoma cells. The 111 amino acid mature human otoraplin contains 1 SH3 domain (46 – 107 amino acids) and a Tyr at position 50 that is reportedly sulfated. Otoraplin takes pasrt in the initiation of periotic mesenchyme chondrogenesis.

    • Introduction

IL4 is a pleiotropic cytokine produced by activated T cells. IL4 is a ligand for interleukin 4 receptor. The interleukin 4 receptor also binds to IL13, which contributes to various overlapping roles of this cytokine and IL13. STAT6, a signal transducer and activator of transcription plays a main role in mediating the immune regulatory signal of this cytokine. IL4, IL3, IL5, IL13, and CSF2 form a cytokine gene cluster on chromosome 5q, with this gene particularly close to IL13. IL4, IL13 and IL5 are regulated co-ordinately by several long-range regulatory elements in an over 120 kilobase range on the chromosome.

    • Introduction

Inhibitor of DNA Binding 2 (ID2) is a part of the inhibitor of DNA binding family, whose members are transcriptional regulators that contain a helix-loop-helix (HLH) domain but not a basic domain. Members of the ID family inhibit the functions of basic helix-loop-helix transcription factors in a dominant-negative way by suppressing their heterodimerization partners through the HLH domains. ID2 play a role in negatively regulating cell differentiation and may be an inhibitor of tissue-specific gene expression.

The IL-6 receptor complex is composed of two membrane glycoproteins: the low affinity receptor and the signaltransducing component. The soluble form of IL6R is found in the urine of healthy adult humans and the serum of HIV positive individuals, as well as in the cell culture supernatants of stimulated PBMC’s. This soluble form of IL6R results from either proteolytic cleavage from the membrane, or an isoform derived splice variant.

    • Introduction

Interleukin-23 receptor (IL23R) is a member of the type I cytokine receptor family, Type 2 subfamily. IL23R contains two fibronectin type-III domains and is expressed by monocytes, Th1, Th0, NK and dendritic cells. IL23R protein pairs with the receptor molecule IL12RB1/IL12Rbeta1, and both are necessary for IL23A signaling. IL23R protein associates constitutively with JAK2 (Janus kinase 2), and it also binds to transcription activator STAT3 in a ligand-dependent manner. IL23R binds IL23 and mediates T-cells, NK cells and perhaps certain macrophage/myeloid cells stimulation most likely by way of activation of the Jak-Stat signaling cascade. IL23R might be responsible for autoimmune inflammatory diseases and be vital for tumorigenesis.

HCV is a small 50nm, enveloped, single-stranded, positive sense RNA virus in the family Flaviviridae. HCV has a high rate of replication with approximately one trillion particles produced each day in an infected individual. Due to lack of proofreading by the HCV RNA polymerase, the HCV has an exceptionally high mutation rate, a factor that may help it elude the host''s immune response. Hepatitis C virus is classified into six genotypes (1-6) with several subtypes within each genotype. The preponderance and distribution of HCV genotypes varies globally. Genotype is clinically important in determining potential response to interferon-based therapy and the required duration of such therapy. Genotypes 1 and 4 are less responsive to interferon-based treatment than are the other genotypes (2, 3, 5 and 6).

Human Immunodeficiency Virus type 1 (HIV1) is the causative agent of Acquired Immunodeficiency Syndrome (AIDS), a devastating disease that affects millions of people worldwide. The HIV1 life cycle is a complex process involving several key viral enzymes, one of which is the integrase (IN). HIV1 integrase plays a crucial role in viral replication by catalyzing the integration of viral DNA into the host cell genome, an essential step for the establishment of a persistent infection.
The study of HIV1 integrase has been of great interest to researchers due to its significance as a therapeutic target. In recent years, advances in recombinant DNA technology have allowed for the production and purification of HIV1 integrase in recombinant form, enabling detailed structural and functional studies. This research aims to characterize the HIV1 integrase recombinant and gain insights into its mechanisms of action during viral integration.
The first objective of this study is to express and purify recombinant HIV1 integrase using various expression systems such as bacterial, yeast, or mammalian cell-based systems. Recombinant DNA techniques, including cloning and expression vector design, will be employed to generate the desired constructs for protein production. The recombinant integrase will be purified using affinity chromatography, followed by characterization using biochemical and biophysical techniques.
The second objective is to investigate the enzymatic activity of the purified HIV1 integrase recombinant. In vitro assays will be performed to determine its ability to catalyze the integration of viral DNA into target DNA sequences. Various substrates, including oligonucleotides and plasmids, will be used to assess the substrate specificity and kinetics of the integrase enzyme. Furthermore, the effects of potential inhibitors or small molecules on the enzymatic activity will be evaluated.
The third objective is to elucidate the three-dimensional structure of the HIV1 integrase recombinant using techniques such as X-ray crystallography or cryo-electron microscopy. This structural information will provide valuable insights into the mechanism of integrase function and aid in the rational design of novel inhibitors targeting integrase.
By characterizing the HIV1 integrase recombinant, this research aims to contribute to our understanding of the molecular mechanisms underlying viral integration. The findings from this study may provide crucial information for the development of novel therapeutic strategies targeting HIV1 integrase, potentially leading to the discovery of more effective antiretroviral drugs.

    • Introduction

IL12B is a subunit of interleukin 12 which is a cytokine that acts on T and natural killer cells, and has a broad array of biological activities. Interleukin 12 is a disulfide-linked heterodimer composed of the 40kDa cytokine receptor like subunit encoded by the IL12B gene, and a 35kDa subunit encoded by IL12A. The IL12B cytokine is expressed by activated macrophages which serve as an important inducer of Th1 cells development. The IL12B cytokine has been found to be important for sustaining a sufficient number of memory/effector Th1 cells to mediate long-term protection to an intracellular pathogen. Overexpression of the IL12B gene was observed in the central nervous system of patients with multiple sclerosis (MS), suggesting a role of the IL12B cytokine in the pathogenesis of the disease. The promoter polymorphism of the IL12B gene has been reported to be linked to the severity of atopic and non-atopic asthma in children.

    Description

IL22RA (Y51A) Human Recombinant produced in E.Coli is a single, non-glycosylated polypeptide chain containing 147 amino acids and having a molecular mass of 16.7 kDa. IL22RA (Y51A) is purified by proprietary chromatographic techniques.

    Source

    Escherichia coli.

    Formulation

The protein was lyophilized from a concentrated (1mg/ml) solution with NaHCO3.

    Purity

Greater than 98.0% as determined by:

    (a) Gel filtration chromatography.

    (b) Analysis by SDS-PAGE.

    Biological Activity

IL22RA (Y51A) Is capable of full inhibition of STAT3 phosphorylation induced by mouse interleukin 22 in HepG cells. Its affinity toward immobilized mIL-22 receptor α1 extracellular domain (mIL-22 Rα1-ECD) or IL-22 binding protein is similar to the non mutated mouse interleukin 22. Human IL-22 antagonist (Y51A) has no agonistic activity in this bioassay.

    • Physical Appearance

Sterile Filtered White lyophilized (freeze-dried) powder.

    • Stability

Lyophilized IL22RA (Y51A) although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution IL22RA (Y51A) should be stored at 4°C between 2-7 days and for future use below -18°C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.

    • Solubility

It is recommended to reconstitute the lyophilized IL22RA (Y51A) in sterile 18MΩ-cm not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

    • Amino Acid Sequence

The sequence of the first five N-terminal amino acids was determined and was found to be Ala-Pro-Ile-Ser-Ser.

    • Background

IL-22 is a part of the IL-10 family of regulatory cytokines and produced by a few populations of immune cells at a site of inflammation. Members of this family share partial homology in their amino acid sequences, but varies in their biological functions. IL-22 effects on non-hematopoietic cells. IL-22 responsible for wound healing and takes part in protection against microbs. Produced by T lymphocytes, IL-22 inhibits IL-4 production by Th2 cells, and induces acute phase reactants in the pancreas and liver.

    • Protein content

Protein quantitation was carried out by UV spectroscopy at 280 nm using the absorbency value of 0.18 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the DNAman computer analysis program of protein sequences.
