TIE-1 (tyrosine kinase with Ig and EGF homology domains 1) and TIE-2/Tek comprise a receptor tyrosine kinase (RTK) subfamily with unique structural characteristics: two immunoglobulin-like domains flanking three epidermal growth factor (EGF)-like domains and followed by three fibronectin type III-like repeats in the extracellular region and a split tyrosine kinase domain in the cytoplasmic region. These receptors are expressed primarily on endothelial and hematopoietic progenitor cells and play critical roles in angiogenesis, vasculogenesis and hematopoiesis. Human TIE-1 cDNA encodes a 1124 amino acid (aa) residue precursor protein with an 18 residue putative signal peptide, a 727 residue extracellular domain and a 354 residue cytoplasmic domain. Whereas two ligands have been described for TIE-2 [angiopoietin-1 (Ang1) and angiopoietin-2 (Ang2)], so far no ligand was found for TIE-1.

BEN domain-containing protein 6 (BEND6) is a neural BEN-solo factor which shares many functional attributes with Drosophila Insensitive, a co-repressor for the Drosophila CSL factor. The BEND6 protein binds the mammalian CSL protein CBF1 and antagonizes Notch-dependent target activation. Furthermore, its association with Notch- and CBF1-regulated enhancers is stimulated by CBF1 and antagonized by activated Notch. In utero electroporation experiments demonstrated that ectopic BEND6 impeded Notch-mediated self-renewal of neocortical neural stem cells and promoted neurogenesis.

Grancalcin is calcium-binding protein which is particularly abundant in human neutrophils. GCA is a member of the penta EF-hand (PEF) subfamily of EF-hand proteins, who also comprises calpain, sorcin, peflin, and ALG-2. GCA undergoes essential conformational changes upon binding of calcium, which subsequently exposes hydrophobic amino acid residues, that direct the protein to hydrophobic surfaces. Death effector domain-containing protein (DEDD) is a cytoplasmic protein. The cell death activity of DEDD relates to its nuclear localization. DEDD translocates to the nucleus during CD95-mediated apoptosis, there it localizes to nucleoli-like structures, activates caspase-6 and particularly inhibits RNA polymerase I-dependent transcription. DEDD is usually expressed in a variety of tissues, and found in the highest levels in the testis. Overexpression of DEDD was shown to induce weak apoptosis.
