The protein is fused to a 6xHis tag at N-terminal and purified by chromatography techniques. P-domain (225-520 a.a.) forms P1- P2-P1 structure.     • Introduction

      Human norovirus is classified into two groups, group 1& group 2. Norwalk virus is the species which belongs to group 1 and was discovered in 1968 at Ohio. Norovirus is a familiar virus which causes human gastroenteritis with the following symptoms vomiting, diarrhea and sickness. CDC report revealed that there are 19-21 million Americans infected by Nororvirus annually with 800 deaths, 1 in 15 people with infection. Around the world, this virus affects about 267 million people and causes over 200,000 deaths each year; these losses are mostly in less developed countries and in the very young, elderly and immuno-suppressed population, though, most cases are self-limited with a full recovery within just a few days. Norovirus is extremely contagious and can spread from human to human through infected food, water or contaminated surfaces. The outbreaks usually occur from November-April, while the peak is in January. Norovirus is a positive sense RNA virus with 7.5 kb nucleotides, encoding a major structural protein VP1 with 50-55kDa. The full length of VP1 capsid comprises the internal N-terminal, Hinge, shell (S) and protruding (P) domains. P domain from 225 to 520 forms P1- P2-P1 structure. Moreover, P domain has a receptor binding region which recognizes human histo-blood group antigens (HBGAs). P domain expressed in bacteria can spontaneously form a P dime as well as a P particle aggregated by 12 P dimmers. P particle displays an increased binding activity to HBGAs higher than virus-like particle (VLP) formed by the full-length capsid. For norovirus vaccine development, we consider P domain as a good candidate.

    • Introduction

      CD4 is a cell-surface glycoprotein found on the mature helper T cells and immature thymocytes, as well as on monocytes and macrophages. (Some cytotoxic T cells have CD4 protein as well.) Normally, about 65% of T cells in the blood are CD4+ (have CD4 protein protruding from their membrane). A mature T cell with either have CD4 or CD8, but not both. During one stage of development T cells develop CD4 and CD8 receptors, but they eventually are differentiated in the thymus and become more specialized.

      Dengue fever is a mosquito-borne viral disease caused by the four serotypes of Dengue virus (DENV-1 to DENV-4). With an estimated 390 million infections occurring annually, dengue fever poses a significant global health burden, particularly in tropical and subtropical regions. The DENV core protein is a vital component of the viral particle, playing a crucial role in viral assembly and genome packaging. Understanding the structural and functional characteristics of the DENV-2 core protein is essential for unraveling the viral assembly process and identifying potential targets for antiviral intervention.
      This research aims to characterize the DENV-2 core protein in its recombinant form to gain insights into its role in viral replication and explore potential strategies for combating dengue fever.
      The primary objective of this study is to express and purify recombinant DENV-2 core protein using various expression systems. Recombinant DNA techniques will be employed to construct expression vectors containing the DENV-2 core gene, followed by expression in bacterial, yeast, or mammalian cell-based systems. The recombinant core protein will be purified using affinity chromatography or other suitable methods, enabling subsequent biochemical and biophysical characterization.
      The second objective is to investigate the assembly and structural properties of the DENV-2 core protein. In vitro assays will be conducted to analyze the ability of the purified core protein to self-assemble into oligomeric structures resembling the viral nucleocapsid. Techniques such as electron microscopy and atomic force microscopy will be utilized to determine the morphology and organization of the assembled structures. Furthermore, the interactions between the core protein and other viral and host factors will be explored using protein-protein interaction assays.
      The third objective is to identify potential antiviral targets within the DENV-2 core protein. Structural studies, including X-ray crystallography or cryo-electron microscopy, will be employed to elucidate the three-dimensional structure of the recombinant core protein. This structural information will provide valuable insights into the protein's functional domains and potential druggable sites, guiding the development of novel antiviral compounds targeting the DENV-2 core protein.
      By characterizing the DENV-2 core protein in its recombinant form, this research aims to deepen our understanding of the assembly process and structural properties of the virus, facilitating the identification of novel antiviral targets. The findings from this study may contribute to the development of effective therapeutic strategies against dengue fever, ultimately reducing the global impact of this mosquito-borne viral disease.

      HCV is a small 50nm, enveloped, single-stranded, positive sense RNAvirus in the family Flaviviridae.
      HCV has a high rate of replication with approximately one trillion particles produced each day in an infected individual. Due to lack of proofreading by the HCV RNA polymerase, the HCV has an exceptionally high mutation rate, a factor that may help it elude the host's immune response. Hepatitis C virus is classified into six genotypes(1-6) with several subtypes within each genotype. The preponderance and distribution of HCV genotypes varies globally. Genotype is clinically important in determining potential response to interferon-based therapy and the required duration of such therapy. Genotypes 1 and 4 are less responsive to interferon-based treatment than are the other genotypes (2, 3, 5 and 6).

      Galectin-3 mediates with the alpha-3, beta-1 integrin the stimulation by cspg4 of endothelial cells migration. Galectin-3 plays an necessary part during the acquisition of vasculogenic mimicry and angiogenic properties associated with melanoma progression. LGALS3 overexpression is highly expressed in early stages of papillary carcinoma, and its expression intensity declines during tumor progression. Serum levels of LGALS3 are high in patients with thyroid malignancy but there is considerable overlap in serum LGALS3 concentrations between those with benign and malignant nodular thyroid disease. LGLAS3 takes part as an immune regulator to inhibit T-cell immune responses and promote tumor growth, as a result providing a new mechanism for tumor immune tolerance.

      Trypanosoma cruzi, better known as T. cruzi, is a deadly parasite that causes Chagas’ disease. This disease is a chronic infection, which primarily affects the heart and nervous system, causing severe neurological disorders, as well as swelling or denervation of nervous tissue in the heart, colon and esophagus. Chagas’ disease often goes undiagnosed due to close association of symptoms to heart disease and a variety of other disorders. The organism can circulate in the blood of infected patients for many years after infection, and can lead to transfusion-acquired infections in blood recipients from these infected donors. Contaminated blood transfusions are suspected to be the primary way in which the parasite has been transmitted to industrialized countries.

      VZV is closely related to the herpes simplex viruses(HSV), sharing much genomehomology. The known envelope glycoproteins (gB, gC, gE, gH, gI, gK, gL) correspond with those in HSV, however there is no equivalent of HSV gD. VZV virons are spherical and 150-200 nm in diameter.
