The HD domain consists of highly conserved residues, in particular histidines or aspartates. Porphyria cutanea tarda, Parkinson’s disease and Stickler syndrome have all been related with HDDC2.

Guanosine-3',5'-bis(diphosphate)-pyrophosphohydrolase MESH1 (HDDC3) contains an active site for ppGpp hydrolysis and a conserved His-Asp-box motif for Mn(2+) binding. HDGF is a self-regulating factor connected with the prognosis of liver cancer, non-small cell lung cancer and pancreatic cancer. HDGF has proliferative, angiogenic, and neurotrophic activity. HDGF is a distinctive nuclear targeting growth factor that is vastly expressed in HCC cells and is a prognostic factor for HCC.

Hepatoma-derived growth factor, related protein 3 (HDGFRP3/HDGF2) is the original member of a family of polypeptides labeled HDGF related proteins (HRPs). The HDGF2 protein augments DNA synthesis and may play a part in cell proliferation. HDGF2 is expressed primarily in the testis and the brain, to an intermediate extent in the heart, and to a slight extent in the ovaries, kidneys, spleen, and liver in humans.

Hepatoma Derived Growth Factor-Like 1 (HDGFL1) is a member of the HDGF family and contains 1 PWWP domain.

Haloacid dehalogenase-like hydrolase domain-containing protein 1A (HDHD1A) is a member of the adiponutrin family. HDHD1A is interesting because it is an X-linked gene that escapes X-inactivation. HDHD1A is particularly important in the perception of human X chromosome structural organization as well as the mechanism of X-inactivation.

HDHD2 is a member of the HAD-like hydrolase superfamily which includes L-2-haloacid dehalogenase, epoxide hydrolases and phosphatases. There are two active sites in HDHD2 - an L-2-haloacid dehalogenase and a carboxylate group. The L-2-haloacid dehalogenase active site catalyzes the hydrolytic dehalogenation of D- and L-2-haloalkanoic acids, producing L- and D-2-hydroxyalkanoic acids.

HDL transports their cholesterol generally to the liver or steroidogenic organs such as adrenals, ovary and testes by direct and indirect pathways. The release of HDL cholesterol to adrenals, ovaries and testes is important for the synthesis of steroid hormones. Since the triglycerides are not stable in HDL, they’re degraded by hepatic lipase in order that finally small HDL particles are left which resume the uptake of cholesterol from cells. The cholesterol supplied to the liver is excreted into the bile and consequently intestine either directly or indirectly after conversion into bile acids. High levels of HDL are associated with a decreased risk of atherosclerosis and coronary heart disease.

The HDV genome exists as a negative sense, single-stranded, closed circular RNA. Because of a nucleotide sequence that is 70% self-complementary, the HDV genome forms a partially double stranded RNA structure that is described as rod-like. With a genome of approximately 1700 nucleotides, It has been proposed that HDV may have originated from a class of plant viruses called viroids. Evidence in support of this hypothesis stems from the fact that both HDV and viroids exist as single-stranded, closed circular RNAs that have rod-like structures. Likewise, both HDV and viroids contain RNA sequences that can assume catalytically active structures called ribozymes.

Heme Binding Protein 1 (HEBP1) is an intracellular tetrapyrrole-binding protein. HEBP1 is comprised of a natural chemoattractant peptide of 21 amino acids at the N-terminus, which is a natural ligand for formyl peptide receptor-like receptor 2 (FPRL2) and stimulates calcium mobilization and chemotaxis in monocytes and dendritic cells.
